[Computer tomographic differentiation of intrathoracic neoplasms and inflammation in the dog].

The purpose of this study was to elaborate on the advantages and limits of computed tomography (CT) in the differentiation of thoracic lesions, in particular neoplasias. In the course of the investigation CT-scans of the thorax of 31 dogs with lesions in the area of the lungs or mediastinum were evaluated. The lesions were rated by morphology, distribution pattern, attenuation values and contrast-enhancement. Biopsies or the whole body underwent a pathohistological examination. Of the 31 dogs 17 had neoplastic and 14 had inflammatory lesions in the thoracic region. With help of the CT, the exact localisations of the different lesions was possible in most cases. Due to their characteristic morphologies, distribution patterns and attenuation values the differentiation between inflammatory and neoplastic lesions was possible in most cases (n=25/31) on the basis of the CT-scans. Mean non-enhanced CT attenuation values of the neoplastic lesions ranged between 31 and 50 HU, of the inflammatory lesions between -251 and 9 HU. Both neoplastic and inflammatory lesions showed contrast enhancement (between 14 and 38 HU and between 2 and 95 HU respectively). The mediastinal abcesses enhanced mainly on the periphery of the lesion. A differentiation of the various types of neoplastic lesions based on the non-enhanced attenuation values was not successful. Only metastasis could be differentiated because of their distribution pattern.

[Placebo-controlled blinded study of postoperative pain therapy with carprofen and levomethadone in dogs with fractures].

It was the aim of this placebo-controlled study to evaluate the analgesic efficacy of the NASAID carprofen and the pure m-agonist levomthadone over a five-day postoperative evaluation period in dogs with fractures of the humerus or the femur (n = 30). Pain and sedation evaluation was carried out with a visual analogues system (VAS) and with the aid of a numerical estimation scale(NRS). The degree of lameness, the pain treshhold, the glucose and cortisol concentration curves as well as the respiration and heart rate and the systolic blood pressure were used as further pain indicators and to identify drug side effects. The levomethadon group displayed the lowest degree of pain on postoperative examination on the first day. On days 2 to 5, the carprofen group showed the lowest degree of pain in comparison to the placebo group. The levomethadon- and the carprofen group showed no statistically proven differences from day 2 on. Due to great variations in the pain scores and comparatively high median pain score especially on the first day of this study, the efficacy of all analgesics evaluated here must be regarded as insufficient in many cases. Only the parameter nociceptive pain treshhold showed a little, the degree of lameness, the glucose and cortisol levels showed no close correlation to the VAS and NRS pain scores and were therefore of little usefulness as postoperative pain indicators. No relevant clinical side effects caused by the used analgesics were detected in the kidney, the liver, the gastrointestinal tract and the circulatory system in this study. Rather, traumatically induced elevation of enzyme levels improves or normalised until the 5th day of the study. In addition, no negative effect on wound healing was noted, especially for carprofen. Therefore, the evaluated analgesics seems to be adequate for postoperative pain therapy also in fracture patients (trauma patients). However, the efficacy of all analgesics evaluated here must be regarded as insufficient in many cases.

[Measurement of the elastic properties of the cancellous bone in the femoral head of the dog].

The degenerative wear and pathologic damage of the joints are reasons for total endoprotheses in man as well as in dogs. The main problem is the aseptic loosening of the protheses. By usig the finite-element-method, the total endoprothesis is designed with new features, with the purpose of preventing loosening and being better adapted to load transmission. In order to simulate the femur of the dog for the numerical analysis, a material law is developed. By taking into account the anisotropy and the local density of the cancellous bone in the femoral head, the young's modules are experimentally determined. The measurements are performed by ultrasonic methods on femoral heads of euthanised dogs. The results show planar isotropic cancellous bone.

[Low flow anaesthesia with isoflurane in the dog].

The aim of the present study was to compare the safety of two low flow (LF) regimes [fresh gas flow (FGF) 20 ml/kg/min (group 2) and 14 ml/kg/min (group 3)] with the high flow (HF) technique (FGF 50 ml/kg/min; group 1) of isoflurane anaesthesia. Data were gathered from ninety dogs assigned for surgery under general anaesthesia with an expected duration of 75 minutes or longer. All dogs had an anaesthetic induction with 0,6 mg/kg I-methadone (maximum 25 mg) and 1 mg/kg diazepam (maximum 25 mg) i.v. Anaesthesia was maintained with isoflurane in a mixture of 50% O2 and 50% N2O as carrier gases, with controlled ventilation. The Monitoring included electrocardiogramm, body temperature, the temperature of in- and exspired gases, arterial oxygen saturation, arterial blood pressure as well as a continuous monitoring of inhaled and exhaled gas concentrations (O2, N2O, CO2, isoflurane). The consumption of isoflurane and carrier gases as well as the recovery times were evaluated for the three groups. The inspired oxygen concentrations always ranged above the minimum value of 30 Vol.-% during low flow anaesthesia. The arterial oxygen saturation ranged between 92-98%, the end tidal concentration of CO2 between 35 and 45 mmHg. Heart rate and arterial blood pressure were within normal limits. Recovery time was significantly shorter after LF than after HF anaesthesia. The highest decrease in body temperature occurred in the HF group 1 because of a significantly lower anaesthetic gas temperature. Despite this, LF anaesthesia resulted in a reduced consumption of carrier gases and volatiles. In conclusion, low flow anaesthesia with isoflurane is a safe technique and offers substantial economic advantages over high flow techniques and is moreover better tolerated by the patients.

Iron particles enhance visualization of experimental gliomas with high-resolution sonography.

BACKGROUND AND PURPOSE: Intraoperative MR imaging and sonography are used for navigation during neurosurgical procedures. The purpose of this experimental study was to evaluate the potential of high-resolution sonography using superparamagnetic iron oxide (SPIO) particles as a contrast medium to delineate brain tumors and to relate these findings with those of MR imaging. METHODS: C6 gliomas were implanted in 36 rats. Eleven days after tumor implantation, the animals underwent MR imaging with a 1.5-T MR imaging unit. Twelve animals received gadopentetate dimeglumine immediately before the MR examination, 12 animals were injected with SPIO particles 24 hours before MR imaging, and 12 animals received no contrast agent. Immediately after MR imaging, the animals were sacrificed and their brains were removed and placed in saline. Sonography was performed instantly after brain removal. Brains were embedded in paraffin, and sections were stained for iron with Perl's stain and for macrophages with ED-1 immunohistochemistry. RESULTS: At MR imaging, the tumors appeared hyperintense on T2-weighted and gadolinium-enhanced T1-weighted images. After application of SPIO particles, they became markedly hypointense on T2-weighted images and hypo- to hyperintense on T1-weighted images. On sonograms, gliomas were iso- to slightly hyperechoic to normal brain parenchyma on nonenhanced and on gadolinium-enhanced images. After application of SPIO particles, tumors became markedly hyperechoic and were distinctly demarcated from the surrounding brain tissue. CONCLUSION: SPIO particles improved the detection and demarcation of the experimental gliomas on sonograms, which may improve intraoperative neuronavigation with sonography.

Cytogenetic investigations in four canine lymphomas.

Four cases of canine lymphoma are presented, including histological examination and cytogenetic investigation. The first case showed a derivative chromosome 13, the second case showed a clonal trisomy 8 and the third case showed a complex karyotype with a clonal trisomy 13 and additional clonal trisomies of the chromosomes 20, 30 and 37, as well as a non-clonal tetrasomy 9. Case four showed a single trisomy 2. Comparing these results with human hematopoietic malignancies, there are notable similarities between both species.

Cytokine genes single nucleotide polymorphism (SNP) screening analyses in canine malignant histiocytosis.

In humans, malignant histiocytosis is a tumour-like disease characterised by increasing proliferation of macrophages and reinforced degradation of erythrocytes. High progression of this disease leads to an unfavourable prognosis for the patients, most of them children up to the age of three years. Histological and cytological findings have proposed an important role of aberrant expression of cytokines in histiocytosis. Due to the fact that Bernese Mountain Dogs (BMD) show a predisposition for spontaneously developing malignant histiocytosis, these dogs could possibly be used as a genetic model organism to elucidate the mechanisms of human malignant histiocytosis. Canine cytokine cDNA transcripts of TNFalpha, Interleukin-1-alpha (IL-1alpha) and Interleukin-1-beta (IL-1beta) were screened for single nucleotide polymorphisms (SNPs). SNP screening in canine cytokine transcripts for malignant histiocytosis has not been carried out before. Total RNA was isolated from tissue samples from lung, spleen, testis and skin of 17 different dogs (fifteen BMDs, one Collie and one West Highland Terrier). The corresponding cytokine cDNAs were amplified, sequenced and then screened for SNPs. The resulting effects on the protein sequence were analysed. Several BMDs and the West Highland Terrier showed SNPs in the coding sequences which led to missense mutations within the protein sequences of TNFalpha, IL1alpha and IL1beta.

[Comparative examination of the lateral cerebral ventricles of different dog breeds using quantitative computed tomography].

Due to the great variations among the shape of the skull in different breeds, the routine use of computed tomography in the diagnosis of central nervous diseases in the dog requires basic knowledge of the structure of the inner subarachnoidal spaces. In neoplastic as well as in inflammatory brain disease, changes of the size and shape of liquor filled spaces can be found. In the present study, the easily demonstrable lateral ventricles of the brain of ten dogs of each breed, respectively, without cerebral disease belonging to the breeds German Shepherd (DSH), Dachshund, Boxer, Labrador Retriever and Toy poodle were characterized with regard to their two-dimensional expansion. The average width and height of the lateral ventricles were ascertained for each breed. The Boxer had the relatively largest lateral ventricles, whereas in the other breeds, their sizes were very similar. 32 of the dogs investigated (64%) had symmetric lateral ventricles, and in 18 dogs (36%) they were asymmetric. Of these 18 cases, the left lateral ventricle was larger in 12 dogs (67%).
