Remarkable efficiency of the aryne chemistry of (dehydro)octafluoro[2.2]paracyclophane when using the Cadogan method.

Generation of the aryne, (dehydro)octafluoro[2.2]paracyclophane, from its acetamide derivative utilizing the Cadogan method led to remarkable results with respect to Diels-Alder and ene reactivity. A comparison was made between this new and virtually unused method and our earlier reported results using Cram methodology (reaction of aryl iodide with potassium tert-butoxide). Surprisingly, no ene reactivity was observed with the Cram methodology. This mechanistic conundrum was examined extensively with no unambiguous explanation for the difference in reactivity being found.

Synthesis of novel pentafluorosulfanylfurans. Two retro-Diels-Alder approaches.

The first examples of furan substituted with an SF5 group are reported. 3-pentafluorosulfanylfurans were prepared from their respective 2-pentafluorosulfanyl-5-cyano-7-oxabicyclo[2.2.1]hept-2-ene precursors via retro-Diels-Alder reactions. Also, a tandem cycloaddition/retrocycloaddition reaction between 4-phenyloxazole and 1-pentafluorosulfanylhex-1-yne was used to prepare 3-pentafluorosulfanyl-4-butylfuran. [reaction: see text].

Homogeneous decomposition of aryl- and alkylimido precursors for the chemical vapor deposition of tungsten nitride: a combined density functional theory and experimental study.

MOCVD growth of tungsten nitride (WN(x)()) and tungsten carbonitride (WN(x)()C(y)()) thin films has been reported from the complexes Cl(4)(CH(3)CN)W(NR) (1: R = Ph; 2: R = (i)Pr; 3: R = C(3)H(5)). To evaluate the role of the imido substituent in film growth, gas-phase homogeneous decomposition of precursor molecules was investigated using density functional theory (DFT) calculations. Computational results and NMR kinetics of acetonitrile exchange by 2 in solution verified that dissociation of the acetonitrile ligand should be facile for 1-3 in the temperature range used for film growth (>450 degrees C). A computational search for transition states for cleavage of W-Cl bonds in the presence of H(2) carrier gas was consistent with a sigma-bond metathesis pathway. Natural bonding orbital (NBO) analysis and bond energy calculations indicated that 1 has a stronger N-C(imido) bond and a slightly weaker W-N bond than 2 and 3, suggesting a greater role for W-N bond cleavage in depositions from 1. These results are consistent with mass spectrometric fragmentation patterns from 1-3 and low nitrogen content in films deposited from 1 as compared to those from 2 and 3.

Self-complementary purines by quadruple hydrogen bonding.

The first discrete, self-complementary, quadruply hydrogen-bonded complexes based on the 2,6-diaminopurine (DAP) scaffold have been prepared; regioselective urea formation at the C2 amino group of the heterocycle allows intermolecular dimerization (K(dim) approximately 1-1.6 x 10(3) M(-1) in CDCl3) through a DADA hydrogen bonding motif.

A highly pyramidalized cage alkene formed via the double Diels-Alder cycloaddition of syn-4,5,13,14-bis(dehydro)octafluoroparacyclophane to anthracene.

[reaction: see text] A double Diels-Alder reaction of the formal syn-bis(dehydro)octafluoroparacyclophane with anthracene leads to formation of a novel cage compound that contains a highly pyramidal double bond. The measures of pyramidality of this double bond constitute what appear to be the highest combined values of psi and phi (34.3 degrees and 33.5 degrees, respectively) yet reported to have been determined by X-ray crystallography. This cage compound, although stable indefinitely as a crystalline compound in air, exhibits the high reactivity with both triplet and singlet O2 in solution that is characteristic of such highly pyramidal pi systems.

Triplet acetylenes as synthetic equivalents of 1,2-bicarbenes, part II: new supramolecular scaffolds from photochemical cycloaddition of diarylacetylenes to 1,4-cyclohexadienes.

1,5-Diaryl substituted homoquadricyclanes which are readily available through cascade photocycloaddition of diarylacetylenes to 1,4-cyclohexadienes are useful supramolecular scaffolds with an angle of about 60 degrees formed by the two aromatic rings defining a hydrophobic cavity. These structural features of pyridinyl homoquadricyclanes were applied to the design of composite organic/inorganic materials with topologies depending on the ratio of ligand to metal. The crystal structure of complex 1 (L1/AgNO(3) in a 1:1 ratio) shows an alternating ligand-metal polymer in which each of the silver ions in its linear coordination geometry is shared between two L1 molecules. A small change in the crystallization method yields a supramolecular rhomboid (complex 2, L1/AgNO(3) 3:2 ratio) which has two ligands that occupy opposite corners of the rhomboid and two silver atoms occupy the other two corners. Connection of the rhomboids units through a third molecule forms unique "beads on a string" polymeric chains. In complex 2, the silver ions adopt a distorted tetrahedral geometry with the nitrate anion occupying one of the vertices of the tetrahedron. The crystal packing of the chain of rhomboids generates cavities which are filled with disordered solvent molecules. Non-symmetrical homoquadricyclane L3 coordinates with silver only through the nitrogen of the pyridine ring but not through the nitrogen of the tetrafluoropyridine ring in which the electron density of the nitrogen lone-pair is very low. The substituents on the polycyclic moiety of the homoquadricyclane cause restricted rotation of the pyridine rings which suggests that the flexibility of such systems can be fine-tuned to create a family of supramolecular scaffolds of controlled rigidity.

Reactions of indole derivatives with oxiranes and aziridines on silica. Synthesis of beta-carbolin-1-one mimic of pancratistatin.

[reaction: see text] Indole and several indoles functionalized at C-2 were condensed with oxiranes, vinyloxiranes, aziridines, and vinylaziridines in the solid state on the surface of silica. The yields of these reactions were compared to those obtained from Lewis acid-catalyzed ring-opening reactions performed in solution and found to be superior in each case. The solid-phase aziridine opening constituted a key step in the synthesis of the beta-carbolin-1-one mimic of pancratistatin. Methyl 2-indolecarboxylate was found to react on the silica gel surface with N-tosylvinylaziridine in 68% yield. A nine-step synthesis of the pancratistatin mimic has been attained. The additional key transformation in this synthesis involved silica gel-catalyzed opening of an epoxide and hydrolysis of an acetonide. Detailed experimental procedures and full characterization are reported for all new compounds.

Triplet acetylenes as synthetic equivalents of 1,2-bicarbenes: phantom n,pi state controls reactivity in triplet photocycloaddition.

Diaryl acetylenes, in which one of the aryl groups is either a pyridine or a pyrazine, undergo efficient triplet state photocycloaddition to 1,4-cyclohexadiene with formation of 1,5-diaryl substituted tetracyclo[3.3.0.0(2,8).0(4,6)]octanes (homoquadricyclanes). In the case of pyrazinyl acetylenes, the primary homoquadricyclane products undergo a secondary photochemical rearangement leading to diaryl substituted tricyclo[3.2.1.0(4,6)]oct-2-enes. Mechanistic and photophysical studies suggest that photocycloaddition proceeds through an electrophilic triplet excited state whereas the subsequent rearrangement to the tricyclooctenes proceeds through a singlet excited state. Chemical and quantum yields for the cycloaddition, in general, correlate with the electron acceptor character of aryl substituents but are attenuated by photophysical factors, such as the competition between the conversion of acetylene singlet excited state into the reactive triplet excited states (intersystem crossing: ISC) and/or to the radical-anion (photoelectron transfer from the diene to the excited acetylene: PET). Dramatically enhanced ISC between pi-pi S(1) state and "phantom" n,pi triplet excited state is likely to be important in directing reactivity to the triplet pathway. The role of PET can be minimized by the judicious choice of reaction conditions (solvent, concentration, etc.). From a practical perspective, such reactions are interesting because "capping" of the triple bond with the polycyclic framework orients the terminal aryl (4-pyridyl, 4-tetrafluoropyridyl, phenyl, etc.) groups in an almost perfect 60 degrees angle and renders such molecules promising supramolecular building blocks, especially in the design of metal coordination polymers.

Synthesis and self-assembly of functionalized donor-sigma-acceptor molecules.

[structure: see text] 3,5,7-Tris(arylmethyl)-1-aza-adamantanetrione donor-sigma-acceptor compounds have been synthesized in four steps. Computational and (1)H NMR analyses rationalize the solubility, gelation, and conformational properties of the C3-symmetric molecules toward employing sigma-coupled donor-acceptor interactions in molecular self-assembly.

Stereoselective, biocatalytic reductions of alpha-chloro-beta-keto esters.

Eighteen known and putative reductases from baker's yeast (Saccharomyces cerevisiae) were tested for the ability to reduce a series of alpha-chloro-beta-keto esters. In nearly all cases, it was possible to produce at least two of the four possible alpha-chloro-beta-hydroxy ester diastereomers with high optical purities. The utility of this approach was demonstrated by reducing ethyl 2-chloroacetoacetate to the corresponding syn-(2R,3S)-alcohol on a multigram scale using whole cells of an Escherichia coli strain overexpressing a single yeast reductase identified from the screening studies.

Pyridinium cationic lipids in gene delivery: a structure-activity correlation study.

Three series of pyridinium cationic lipids useful as nonviral gene delivery agents were prepared by reaction of pyrylium salts with aminodiols, followed by acylation with fatty acyl chlorides. On the basis of this set of compounds, we undertook a comprehensive structure-activity relationship study at the level of the linker, hydrophobic anchor, and counterion in order to identify the structural elements that generate the highest transfection efficiency for this new type of cationic lipid. The results revealed that when formulated with cholesterol at a 1:1 molar ratio, the 1-(1,3-dimyristoyloxyprop-2-yl)-2,4,6-trimethylpyridinium, under the form of hexafluorophosphate (5AMyr) or chloride (5DMyr), was able to transfect NCI-H23 lung carcinoma with efficiencies surpassing classic DOTAP-based formulations and with lower cytotoxicity. Subsequent tests on other malignancies yielded similarly promising results.

Electrooxidative coupling of furans and silyl enol ethers: application to the synthesis of annulated furans.

The preparation of annulated furan systems as key synthetic intermediates through the application of a two-step annulation involving an electrochemical ring closure between a furan and a silyl enol ether has been studied. The reaction was shown to be quite general for the formation of six-membered rings in good yields and was tolerant of a variety of different functional groups. The ring closure was highly stereoselective, leading to the formation of cis-fused systems. Cyclic voltammetry and probe molecules were used to gain mechanistic insight into the reaction. These studies suggested that the key ring closure involved an initial oxidation of the silyl enol ether to a radical cation followed by a furan-terminated cyclization.

Bridged synthons from tetrabromocyclopropene: studies on the rearrangement of the primary Diels-Alder adduct with 2,5-dimethylfuran.

The reaction of tetrabromocyclopropene and furan leads directly to 8-oxabicyclo[3.2.1]octadiene derivatives. It has been proposed that this involves an initial Diels-Alder reaction followed by rearrangement of the primary adduct. We have, for the first time, isolated a primary adduct and established through X-ray crystallographic analysis that the adduct is the product of an exo-selective addition. Kinetic studies suggest the intermediacy of charged intermediates during the rearrangement.

Simple synthesis of a weak nucleophilic base (4-ethyl-2,6-diisopropyl-3,5-dimethylpyridine) evidencing a double Janus group effect.

By analogy with 2,6-di-tert-butylpyridine and its 4-methyl-substituted derivatives, which are nonnucleophilic bases, 4-ethyl-2,6-diisopropyl-3,5-dimethylpyridine (4) is also such a base. The isopropyl groups (Janus-like groups) are forced by the neighboring methyl groups to turn their "tert-butyl-analogue face" toward the heteroatom, thereby protecting it sterically against electrophilic attack. The synthesis proceeds in two stages via the corresponding pyrylium salt 3 that is obtained by alkene diacylation. X-ray data for 4, its picrate, and the hexafluorophosphate of 3 confirm that the ground-state conformation agrees with the Janus effect prediction. The chemical behavior of 4 indicates that it is indeed a weak nucleophilic base, which is able to substitute the nonnucleophilic bases in organic syntheses. The compound 3 reacts at normal pressure with methylamine or ethylamine, forming N-alkylpyridinium salts. The cationic polymerization of isobutene in the presence of 4 was also investigated.

meso-Tetrahydropyranylperoxides: molecular structures in solution, in the crystal, and by DFT calculations and their isomerization to the racemate.

The crystalline peroxide 3a is the main product (out of 10 theoretically possible) from the aerial peroxidation of all-cis-2,4,6-trimethyltetrahydropyran (2a). It has a similar structure both in solution and in the crystal as shown by nuclear Overhauser effects and X-ray analysis, respectively. Theoretical calculations at a density functional theory level (B3LYP/6-31G) provide insight into the stabilities of the different stereoisomers of this peroxide, accounting for the facile, acid-catalyzed isomerization from the meso form to the racemate. Peroxide 3b, which is the 2-tert-butyl analogue of 3a, out of 22 theoretically possible isomers, crystallizes in a similar meso form. As a result of crystal packing effects and the intrinsically (axial) chiral peroxy "chromophore" that deviates slightly from the antiperiplanar conformation, both enantiomorphic forms of 3b are encountered in the lattice.
