[Effect of beta-sitosterol incorporated into liposomes on several indicators of lipid metabolism in experimental hypercholesteremia in rats].

Alterations in lipid content of microvilli membranes, affecting their physical structure, as well as in lipoproteins of rat small intestinal mucosa and of blood plasma were studied in experimental hypercholesterolemia after treatment with crystalline and liposomal preparations of beta-sitosterol. The liposomal preparation of beta-sitosterol exhibited distinctly higher hypocholesterolemic activity as compared with the crystalline sterol. Possible role of structural and functional alterations in enterocyte membranes and in intestinal lipoproteins during absorption of cholesterol as well as inhibition of the processes by means of beta-sitosterol are discussed.

[Potentiometric titration in biological studies].

The employment of the potentiometric titration method for determination of the nature and concentration of ionized groups in synthetic polyelectrolytes, biopolymers and in membrane fragments of the cell has been considered. The interpretation of the results of potentiometric investigations in the biological system is complicated because of a neglection of the electrostatic factor and differentiation of molecular and supramolecular, structural changes affecting the availability of ionogenic groups. The method, however, provides an important information concerning the character and, particularly, the mechanisms of structural changes in biosystems and in their components under the influence of different factors.

[Effect of ultraviolet light on sterol-containing membranes in nystatin solutions].

The nystatin induced conductivity of artificial phospholipid membranes containing ergosterol or 7-dehydrocholesterol drops sharply under the action of ultra-violet light as a result of the transformation of these provitamins into adequate D2 and D3 vitamins. The UV-light does not change the conductivity of the membranes containing cholestrol in nystatin solution. Nystatin slightly effects the membrane resistance in the presence of D2 and D3 vitamins.

[Effect of ecdysterone on the metabolism of purine and pyrimidine nucleotides in chicken tissues].

Hormonal amounts (10(-7) M) of ecdysterone phytoecdysteroid have been studied for their effect on the metabolic processes in the chicken tissues. Changes in the blood serum parameters which characterize the state of the integral systems of mineral, lipid, protein and nucleotide metabolism as well as the dynamics of metabolism of nucleic acids and their precursors--purine and pyrimidine nucleotides in the tissues with actively functioning protein-synthesizing system prove that the biological effect of ecdysterone after its introduction to the bird organism manifests both in the non-genome early phase of the action (0-1 h) and in the late genome phase (24-72 h), i.e. in analogy to the time of response to the introduction of classical steroid hormones. It is supposed that polyfunctional early effect of ecdysterone is mediated by its action on one of universal transmembrane signal systems of the cells--adenylate cyclase or polyphosphoinosite one, while the presence of the expressed changes in metabolism of nucleic acids in the late phase unambiguously indicate to its action in the bird organism as that of a typical steroid hormone.

[Immunomodulating effect of ecdysterones].

Ecdysterone, its 20-desoxy-derivative alpha-ecdysone, their 2-desoxy-derivatives ecdysterone 2, 3, 22-triacetate and preparation BTI-4 have been studied for their effect on [3H]-thymidine incorporation in different populations of animal and human lymphocytes. It is shown the ecdysteron and its analogs in concentrations of 10(-12)-10(-5) M take considerable stimulating effect on DNA biosynthesis in animal lymphocytes activated by polyclonal mitogens. The concentration of ecdysterone being increased to 10(-4) m one can observe complete inhibition of activating effect of polyclonal mitogens. Effect of the studied ecdysteroids did not considerably depend on their structure. In case of splenocytes the stimulating effect of ecdysterone on DNA biosynthesis is less expressed than in the case of activated thymocytes. Ecdysterone was established to have considerable inhibiting effect on DNA biosynthesis in the culture of activated Con A cells of lymphocytes in the peripheral blood of healthy donors.

[The composition and physicochemical properties of the blood lipoproteins in rats exposed to external gamma irradiation].

Content of lipids, character of chemiluminescence of blood plasma and certain classes of lipoproteins have been studied. Geometrical parameters, nature and quantity of charged groups of lipoprotein particles accessible for titration have been determined 1 and 30 days after a single external gamma irradiation of rats in a dose of 3 Gy. The used irradiation dose exerts an expressed hyperlipidemic effect retained for one month after irradiation. The disturbances in the spectrum of blood lipids and lipoproteins are of hyper-beta and hyper-prebeta lipoproteinemia character. Considerable disturbances of physicochemical properties of different classes of lipoproteins have been detected. They are exhibited in changes of the pattern of free-radical processes, state of the charge of surface ionogenic groups and geometrical parameters of lipoprotein particles. Changes registered by the methods of potentiometric titration and correlation spectroscopy are most expressed in lipoproteins of very low density and those of low density.

[Antiradical properties and antioxidant activity of ecdysterone].

Antiradical properties and total antioxidative activity of ecdysterone (10(-6)-10(-3)M were found on the model systems--photochemical one and that containing phosphatidylcholine liposomes oxidated with Fe2+ and Fe3+. The antioxidative activity of ecdysterone was shown to be comparable with that of the known inhibitors of peroxide oxidation of lipids--diethylparaphenylendiamine and ethylendiamine tetraacetate.

[Ecdysterone induces the activity of multiple forms of acid phosphatase and malate dehydrogenase in fat bodies of Anterahea pernyi pupae].

The method of electrophoresis in PAAG has been used to study activity of multiple forms of acid phosphatase and malate dehydrogenase of the fat body of Anterahea pernyi pupae in norm and affected by ecdysterone. It is shown that an increase of total activity of acid phosphatase and malate dehydrogenase with administration of a steroid hormone is promoted by growth of activity of certain inducible multiple forms of the studied enzymes. Ecdysterone injection considerably elevates activity of the most electrophoretically mobile form of acid phosphatase and provokes new formation of malate dehydrogenase form. Ecdysterone-induced increase of activity of the studied enzyme forms in the fat body of Anterahea pernyi pupae is blocked by actinomycin D, an inhibitor of transcriptional processes, and by puromycin, a translation inhibitor. These data permit concluding that an increase in activity of the mentioned forms of enzymes observed with hormonal induction is a result of their synthesis de novo and is mediated by the DNA-dependent RNA-synthesis.

[Lipid and fatty acid composition of Eisenia foetida].

It is found out that the content of lipids in the biomass of the studied populations of Eisenia foetida is rather high: 2.5-5.2% of the wet mass. The content of phospholipids is 40-55%, C27-sterols--1.5-3.4% of the mass of coarse extracts of lipids. Lipids of tissues contain also 47-54% of saturated (C10-C24) fatty acids as well as to 23% of monoene and to 13% of polyene unsaturated (C14-C22) fatty acids. The acids with the odd number of carbon atoms compose about 25% and acids with a branched carbon chain about 23% of the above percentage. Considerable content of lipids and biologically active fatty acids in tissues of the studied object permits considering it as a promising source of raw materials for production of valuable pharmacological preparations.

[Interaction of very low density lipoproteins (VLDL) with macrophages and their triboluminescence in hypercholesterolemia].

Accumulation of cholesterol esters and triglycerides in peritoneal mice macrophages in the course of their interaction with lipoproteins of very low density (VLDL) is shown to grow considerably under conditions of hypercholesterolemia. A decrease of triboluminescence intensity characterizing the surface charge has been revealed at hypercholesterolemia both in VLDL and in the blood plasma. It is supposed that the triboluminescence method may be used for testing of the atherosclerotic process development.

[Composition and physical structure of atherogenic lipoproteins in ischemic heart disease].

The results of comparative studies in atherogenic lipoproteins of blood plasma under heart ischemic disease (HID) which is accompanied by hypercholesterinemia or proceeds without disturbances in the lipid metabolism, evidence for considerable differences in the composition and physical structure of very low-density lipoproteins (VIDL) and low-density ones (LDP) under the investigated states. The decrease in the surface charge density and in sizes of VLDL, as compared to normal, which are least expressed at HID and an increase in the surface charge density with certain increase of LDL radii under this pathologic state are revealed.
