[The inhibition of complement-dependent hemolysis of liposomes containing cerebroside sulfate].

Cerebroside sulfate (CGS) was found to be capable of inhibiting complement-dependent hemolysis. The activity dependence of CGS-containing liposomes on their composition was studied. Mixtures of CGS with phosphatidylethanolamine, phosphatidylserine, sphingomyelin from cattle brain, cerebroside from cattle spinal cord (CG), and egg yolk phosphatidylcholine (ePC) were investigated. In the case of binary CGS/ePC mixtures, the antihemolytic activity varied nonlinearly with an increase in the mass part of CGS: it sharply increased with an increase in the CGS part from 0.3 to 0.5 and decreased by 20-30% of the maximum value with an increase in the CGS part from 0.9 to 1. On the basis of these experiments, the optimum distance between the charged groups of CGS was estimated to be 0.92-1.6 nm. In the ternary compositions of 4:3:3 CGS/ePC/polar lipid, only CG increased the activity of liposomes as compared to that of liposomes from the 4:6 CGS/ePC. The preliminary incubation of CGS-containing liposomes with complement decreased hemolysis more effectively than incubation with other components of the hemolytic system. This suggests that the interaction of CGS-containing liposomes with the complement proteins is responsible for their antihemolytic activity.

[Liposomal forms of cytostatics in oncology].

The current state and prospects of using the liposomal forms of anthracyclinic antibiotics, fluoroucil agents, and platinum compounds are discussed as potential drugs for chemotherapy of tumors. Comparative studies of standard and liposomal forms of the above groups of compounds were made to determine their therapeutical effects.

[Ganglioside GM3 from horse erythrocytes: structure and effect on cell proliferation].

An increase of the mouse fibroblast proliferation by ganglioside GM3 from equine erythrocytes is described. The structure of GM3 has been established on the basis of chemical methods, enzymatic degradation, GC-MS, as well as plasma desorption mass spectrometry and HPLC of 9-anthrylmethyl esters of gangliosides to characterize the long-chain base composition. The oligosaccharide moiety includes an N-glycolylneuraminic acid residue, whereas the main components of the lipid moiety are 20:1 sphingosine and 24:0 fatty acids.

[Lipids in drugs].

Data on the use of phospho- and glycolipids in drugs with various action targets are analysed. The issues include the use of lipids in parenteral emulsions. Essentiale-type++ preparations, and lipid adjuvants. Special emphasis is given to the transport and reconstructive functions of lipids. The drug-contained lipids were found to have a high biological activity, no allergy-inducing reactions, and a high biological degradability. The analysed evidence confirms the expediency of using drugs containing lipids of different chemical structures for medicinal purposes.

[Isolation and purification of biopolymers using an affinity chromatography method. IX. Preparation, properties and use of affinity adsorbents with immobilized polypeptide fragments of collagen].

Synthesis and properties of new affinity adsorbents with immobilized polypeptide fragments of collagen molecule (alpha-chains, beta-components, cyanogen bromide peptides) were described. Adsorbents with alpha-chains and alpha 1CB7-peptide had fibronectin binding capacity 1.5-2.0 times higher than commercial gelatin-Sepharose. Commercial production of highly purified fibronectin from human plasma using affinity chromatography on immobilized individual alpha-chains of collagen was developed.

[A new chromatographic method for the preparative isolation of phosphoinositides and other anionic phospholipids].

A new chromatographic procedure for preparative isolation of mono-, di- and triphosphoinositides and other anionic phospholipids with the use of adsorbents containing primary amino groups is described. Sorbents with immobilized neomycin, L-lysine and aminoalkyl groups were tested. Conditions for isolation of chromatographically pure phospholipids of separate classes on the above sorbents were developed. Isolation of polyphosphoinositides on the amino sorbents represents a new type of chromatography involving bioaffinity and ion-exchange interaction.

[Effect of gangliosides on the resistance of mice to the rabies virus].

The survival of animals has been studied after their infection with a mixture of rabies virus and gangliosides isolated from various organs. It has been found that virus incubation with a summarized fraction of cerebral gangliosides increased considerably the survival of animals. At the same time it was shown that gangliosides from cattle spleen and red blood cells taken in the same proportions as cerebral gangliosides, do not have any protective effect. The in vivo study of the antiviral ganglioside activity has been performed.

[Action of phospholipids and gangliosides on tumor cell sensitivity to the cytostatic and membrane-toxic action of splenic effectors].

The authors attempted to study which parts of the lipid molecules are most responsible for an increase in the sensitivity of the tumor cell to the cytostatic and membrano-toxic action of natural spleen effectors. Use was made of different combinations of egg phosphatidylcholine, phosphatidylethanolamine, cardiolipin, dipalmitoyl phosphatidylcholine and a mixture of bovine brain gangliosides. Introduction of phosphatidylethanolamine into the membrane of tumor cells increased, that of phosphatidylcholine and cardiolipin did not change whereas introduction of a mixture of brain gangliosides reduced their sensitivity to spleen effectors. Introduction of brain gangliosides together with egg phosphatidylcholine into the membrane of the target cell increased whereas that together with dipalmitoyl phosphatidylcholine reduced its sensitivity to spleen effectors. It is concluded that the increase in the sensitivity of the tumor cell primarily depends on changes in the lipid template of its membrane, induced by unsaturated fatty acids of egg phosphatidylcholine, and to a less degree on the properties of carbohydrate heads of gangliosides.

[Liposomes and other nanoparticles as drug delivery systems].

The basic principles of designing of nanoparticle drug delivery systems are considered. The special attention is paid to liposomes, because the greatest success has been achieved in this area. The modern condition in such intensively developing areas as antitumor and, antibacterials drugs, gene therapy, vaccines is reviewed.

[Major trends in lipid immunochemistry].

Data are presented on immunochemical properties of lipids, the most important group of biologically active substances. Problems on antigenic, immunogenic and adjuvant activities of lipids are considered. A possible use of lipid antigens for diagnosis of different infectious diseases is demonstrated and main principles of their construction are suggested. Data are available on immunogenicity of phospho- and glycolipid mixtures as well as on practical application of the obtained antibodies. Guidelines for the use of immunochemical properties of lipids are outlined.
