[Changes in pain sensitivity under increased atmospheric pressure].

It has been established that augmentation of air pressure from 0.1 to 1.1 MPa (with 0.1 MPa intervals) was accompanied in rats with the development of progressive analgesia which was measured according to the threshold of vocalization in the test of electrical stimulation of the tail. The highest analgesic response arose at 0.7-1.1 MPa. All the animals might be divided into two groups: group 1-72% of the animals with a 200% increase of the threshold, group 2--animals with such an increase by 15%. The augmentation of the pressure of heliox (79.1% of helium, 20.9% of oxygen) also caused analgesia, but not so strong. In patients pain thresholds to the mechanical nociceptive stimulation also increased by about 43-67% and 95-100% under the influence of increased air pressure of 0.4 and 0.7 MPa, respectively. In group 1 patients (67%) pain threshold increased by 50-100%, in group 2 by 15-25%. Pretreatment with naloxone (1 mg/kg), atropine (1 mg/kg), yohimbine (1 mg/kg), parachloramphetamine (5 mg/kg) and prasosin (1 mg/kg) decreased hyperbaric analgesia in rats by 41-56, 41-56, 17-19, 17-19%, respectively. The role of increased partial pressure of nitrogen in hyperbaric analgesia and possible neurochemical mechanisms of its realization are discussed.

[Effect of electroacupuncture on the activity of neurons in the central gray matter of the midbrain].

The effect of electroacupuncture in locally-segment and general analgetic points on background impulse activity of central gray substance neurons and their activity caused by nociceptive stimulation of the dental pulp, infraorbital nerve and forearm skin surface was studied in acute experiments on cats. It has been established that general analgetic points are better represented in the central gray substance, as compared to locally-segment points. Different degree involvement of central gray substance in the realization of acupuncture analgetic effect in different points is postulated. The role of dorsal and ventral compartments of the central gray substance in acupunctural analgesia is discussed.

[Local anesthetic properties of a polymeric compound of trimecaine].

Original techniques were used in experiments on rabbits and rats to explore the activity and duration of a local anesthetic action of a new trimecaine polymer as compared with trimecaine hydrochloride. The polymer effect was found to be 1.5-2 times longer as compared to that of its low-molecular analog, which was discovered in experimental terminal, conduction and infiltration anesthesia in particular. According to the data obtained, the prolongation of the effect of the new drug is determined by slow separation of trimecain from the polymer.

[Effect of electroacupuncture on signs of emotional stress caused by pain].

The experiments on male white rats have shown that electroacupuncture of the point analog Da-Djuy abolished the consequences of pain-induced acute emotional stress. The stress-protective action of electroacupuncture was comparable with the effect of diazepam in a dose of 2.5 mg/kg and resulted in a significant reduction of the number of gastric erosions and in emotional reactivity, as well as in the increased aggressiveness as compared with the control group exposed to stress. The data obtained are discussed from the standpoint of electroacupuncture and diazepam action on the antinociceptive mechanisms of the brain.

[Effect of met- and leu-enkephalins and their synthetic analog on stimulation and acupunture analgesia].

Experiments on unrestrained rats were carried out to study the effect of intraventricularly injected met- and leu-enkephalins and their synthetic analog Tyr-dAla-Cly-Phe-NH2 on analgesia induced by electrical stimulation of the central gray. It was shown that subanalgesic doses of enkephalins and their synthetic analog facilitated the appearance of analgesic action on subthreshold antinociceptive-brain stimulation and potentiated the analgesic effect of threshold central gray stimulation. Subanalgesic and low analgesic doses of the peptides increased antinociceptive effect of electroacupuncture. The data obtained are discussed from the standpoint of the implication of the peptidergic mechanisms in the realization of acupuncture and stimulation analgesia.

[Effect of stimulation of antinociceptive brain zones on the analgesic effect of electroacupuncture in rats].

It was shown in experiments on freely moving rats that electroacupuncture (EA) depresses the emotional-behavioral response to the nociceptive stimulation of the tail. Analogous changes in the pain reaction pattern arose during electric stimulation of the central grey matter and posterior parts of the lateral hypothalamus. In the presence of antinociceptive brain stimulation. EA was followed by strong potentiation of analgesia. Naloxon in a dose of 5 mg/kg removed the antinociceptive effect produced by stimulation of the deep brain structures and the analgetic effect of electroacupuncture. Potential mechanisms of the realization of the acupuncture effect are discussed.

[The combined use of acupuncture and antidepressants for managing the spondylogenic lumbosacral pain syndrome].

The effects of antidepressants amitriptyline and pyrazidol on the analgesic action of acupuncture in patients with lumbosacral radiculitis was studied by the double blind control method. The combined use of acupuncture with antidepressants was shown to enhance the effectiveness of the treatment, to increase on the average by 5-6 hours the duration of analgesia within 24 hours after each procedure, to reduce the duration of the treatment on the average by 4-5 days, that indicates the expediency of clinical use of the proposed combination.

[The analgetic activity of antidepressants and their influence on the pain-relieving effect of acupuncture].

The analgesic properties of new Soviet-made antidepressants pyrasidol, incazan, tetrindol as compared with amitriptyline and their influence on the analgesic effect of acupuncture were studied in experiments on rats by using the method of electrical stimulation of the tail root with stimuli of the increasing intensity. All the studied drugs at single and subchronic (6 days) administration possessed the analgesic actions. Pyrasidol exerted the most pronounced effect. The antidepressants enhanced the analgesic effect of electroacupuncture, in particular during activation of the points of general action and this effect showed up to the greatest degree under medication with pyrasidol.

[Analgesic activity of probon compared to morphine].

The analgetic action of probone as compared to that of morphine was studied in chronic tests on cats. Changes in individual signs of a complex reaction in response to a graded stimulation of the cutaneous nerve were assessed. Probone in doses of 50--150 mg/kg and morphine in doses of 2--4 mg/kg raised the threshold of the flexor reflex and reduced manifestations of the emotional-behavioral and vegeto-motor reactions of the animals. But unlike morphine probone did not suppress the vocal reaction, this being a specific component of the response to pain. It is assumed that the change in the response reaction occurring under the effect of probone is due not so much to its analgetic action, but rather to the myorelax and tranquilizing effects.

[Possibility of modulating morphine analgesia against a background of activation of serotonin- and GABA-ergic processes].

The analgesic effect of morphine in conditions of serotonin- and GABA-ergic activation was studied in experiments on male albino rats. The serotonin re-uptake blockers fluoxetin and citalopram (10 and 20 mg/kg) did not exert clear analgesic effect or did not potentiate morphine action in doses of 5 and 10 mg/kg. The GABA agonist muscimol (0.5--2 mg/kg) did not influence the pain reaction structure and did not enhance morphine-induced analgesia. The role of serotonin- and GABA-ergic processes in realization of the analgesic action of morphine is discussed.
