Quantification of whole virus particles (146S) of foot-and-mouth disease virus in the presence of virus subunits (12S), using monoclonal antibodies in a sandwich ELISA.

This paper describes a method for the specific quantification of whole virions of foot-and-mouth disease (146S) in the presence of virus subunits (12S). The method involves the use of virus neutralising monoclonal antibodies directed against a linear epitope of the VP1 loop region of a type O virus. The monoclonal antibodies were used as both capture and detecting reagents (labelled with horse radish peroxidase) in a sandwich ELISA. Such monoclonal antibodies also have the advantage that they do not detect viruses containing proteolytically cleaved VP1, thus the assay system is ideal for estimation of whole particles in vaccine manufacture where the immunogenicity of the vaccine depends on virus integrity (whole virions being present) and uncleaved capsid protein. VP1. Other combinations of different anti-type O FMD virus monoclonal antibodies used as capture and detecting reagents were also examined. The system could be adapted to on-line continuous testing of virus being produced during a manufacturing run allowing maximisation of virus yield and quality control.

Fertilization rate in couples with unexplained infertility.

A group of 24 couples with unexplained infertility was scheduled for in-vitro fertilization and tubal embryo transfer between May 1989 and September 1990. In the same period, in-vitro fertilization and intrauterine transfer of embryos was planned in a control group of 44 women with tubal infertility. The mean age and duration of infertility were similar in both groups and the same scheme of ovarian stimulation was used. No statistically significant difference was obtained comparing oestradiol levels and numbers of mature oocytes retrieved between the group of patients with unexplained infertility and those with tubal infertility. The fertilization rate of the oocytes obtained from women with unexplained infertility (60.4%) was significantly lower (P less than 0.001) than that of the oocytes obtained from patients with tubal infertility (87.3%). There was no statistically significant difference in the cleavage rates between patients with unexplained infertility and those with tubal infertility. It is concluded that lack of fertilization is an unexplored cause of infertility in couples with unexplained infertility.

Primary intraosseous calvarial meningioma.

A case of primary intraosseous calvarial meningioma is documented by computed tomography and conventional radiography of the resected specimen. The lesion involved the frontal bone and expanded the diploic space. The process was radiodense with an internal radiating pattern that was perpendicular to the plane of the calvarium.
