Electromyographic validation of the pectoralis major and deltoideus anterior muscles in inverted "flying" exercises with loads.

Inverted 'flying" exercise with external loads of 25, 50, 75 and 100% of each individual maximum load in the pectoralis major and deltoideus anterior muscles was electromyographically analyzed in eleven male volunteers, using surface electrodes MEDI-TRACE-200 connected to a biological signals acquisition module coupled to a PC/AT computer. Electromyographic signals were processed and the effective values obtained were standardized through maximum voluntary isometric contraction. When the concentric phase of each muscle with the same load was statistically compared with the eccentric phase, it was observed that for all loads all the muscles presented significant electromyographic difference, and that the concentric phase was always higher. By analyzing the different loads for each muscle, it was noticed that in the concentric phase all the muscles presented significant electromyographic activity, being it higher with maximum load. When the effect of each load on different muscle in the concentric and eccentric phases was analyzed, the muscles presented a distinct activity profile.

Synthesis of sterically stabilized polystyrene latex particles using cationic block copolymers and macromonomers and their application as stimulus-responsive particulate emulsifiers for oil-in-water emulsions.

2-(Dimethylamino)ethyl methacrylate (DMA) was block copolymerized with methyl methacrylate (MMA) using group transfer polymerization to give four AB diblock, ABA triblock, and BAB triblock copolymers of low polydispersity (Mw/Mn < 1.20). In addition, a near-monodisperse styrene-functionalized DMA-based macromonomer was synthesized via oxyanionic polymerization using a potassium 4-vinylbenzyl alcoholate initiator. These five well-defined, tertiary amine methacrylate-based copolymers were evaluated as steric stabilizers for the synthesis of polystyrene latexes via emulsion and dispersion polymerization. The most efficient steric stabilizers proved to be the DMA-MMA diblock copolymer and the DMA-based macromonomer. The polystyrene latexes were characterized in terms of their particle size and morphology, stabilizer content, surface charge, and surface activity using dynamic light scattering, scanning electron microscopy, 1H NMR spectroscopy, aqueous electrophoresis measurements, and surface tensiometry, respectively. The pH-dependent surface activity exhibited by selected latexes suggests potential applications as stimulus-responsive particulate emulsifiers for oil-in-water emulsions.

Simultaneous determination of E-2-nonenal and beta-damascenone in beer by reversed-phase liquid chromatography with UV detection.

A method for the simultaneous determination of E-2-nonenal and beta-damascenone in beer by reversed-phase liquid chromatography using UV detection is presented. The method consists of beer steam distillation, followed by an extraction/concentration step using Sep-Pak Plus C18 RP cartridges and determination by HPLC at 226 nm UV-absorption maximum. The identity of the compounds was confirmed by GC analysis with MS detection of the isolated fractions. A recovery factor of approximately 80% was obtained for beta-damascenone with a R.S.D. of 3%. E-2-Nonenal and beta-damascenone were monitored in a comparative study of fresh and either naturally and forced aged beer. The results obtained show that both compounds have a similar behaviour through an extended storage of beer and consequently can be used as good analytical markers of beer ageing. Nevertheless, the use of beta-damascenone seems to be more convenient because this compound appears in beer in higher concentrations than E-2-nonenal, thus making it easier to measure.

Behavioral modulation by mutilation pictures in women.

Previous studies have shown that women are more emotionally expressive than men. It is unclear, however, if women are also more susceptible to the emotional modulation of behavior imposed by an affective stimulus. To investigate this issue, we devised a task in which female subjects performed six sequential trials of visual target detection following the presentation of emotional (mutilation and erotic) or neutral pictures (domestic utensils and objects) and compared the data obtained in the present study with those described in a previous study with male subjects. The experiment consisted of three blocks of 24 pictures and each block had an approximate duration of 4 min. Our sample consisted of 36 subjects (age range: 18 to 26 years) and each subject performed all blocks. Trials following the presentation of mutilation pictures (283 ms) had significantly slower reaction times than those following neutral (270 ms) pictures. None of the trials in the "pleasant block" (271 ms) was significantly different from those in the "neutral block". The increase in reaction time observed in the unpleasant block may be related in part to the activation of motivational systems leading to an avoidance behavior. The interference effect observed in this study was similar to the pattern previously described for men. Thus, although women may be more emotionally expressive, they were not more reactive to aversive stimuli than men, as measured by emotional interference in a simple reaction time task.

Determination of E-2-nonenal by high-performance liquid chromatography with UV detection assay for the evaluation of beer ageing.

The analysis of E-2-nonenal is of considerable interest for the brewery industry as this compound is claimed to be responsible for a paper/cardboard unpleasant flavour. Usually, the presence of E-2-nonenal can be noticed in aged beers at levels higher than 0.1 microg/l. In this work, an analytical method was developed to determine E-2-nonenal in beer involving steam distillation of beer followed by an extraction/concentration step using solid-phase extraction and determination of E-2-nonenal by HPLC with UV detection. Fastness and simplicity are the main advantages of the proposed method, when compared with other existing methodologies for the determination of E-2-nonenal in beer. Using the developed conditions, the interference of E-2-nonenal formed by degradation of its precursors during steam distillation is almost negligible. The presence of sulphur dioxide at legal levels does not interfere with the assay. The method was used in a comparative study of fresh and either naturally or forced aged beers. A much larger chromatographic peak was found near the peak of E-2-nonenal that correlates well with the peak of E-2-nonenal. Identification of the corresponding compound is currently under investigation, considering its future application on the evaluation of beer ageing.

Evaluation of motivation methods used to obtain appropriate oral hygiene levels in schoolchildren.

The aim of the present study is to assess the behaviour of different motivation methods on levels of oral hygiene among schoolchildren aged from 7 to 9 years in Araraquara, SP, Brazil. The methods tested were: indirect instruction using 'The Smiling Robot' (group I), indirect instruction through class presentation (group II) and direct instruction with macromodels (group III). A control group was also constituted, which received no kind of motivation (group IV). The O'Leary Plaque Index was used as the evaluation method, applied before the instruction and 30 days after application of the different methods. It was noted that the plaque index had not decreased in group IV only. In conclusion, all the motivation methods promoted significant decrease of plaque index and among these methods, the 'The Smiling Robot' was the one that provided the best results.

Electromyographic analysis of the arm muscles in "front support" exercises.

The electromyographic activity of the biceps brachii--BB (long head), triceps brachii--TB (long head) and deltoideus--DA (clavicular portion) muscles, during the going (G) and return (R) phases in "front support" exercise, as well the efficacy of this exercise for the development of these muscles strength were studied in 10 male volunteers. The values were normalized through maximum voluntary isometric contraction (MVIC = 100%) and statistically analyzed using the Friedman, DMS and Wilcoxon non-parametric test. A value of p < or = 0.05 indicated significance (Campos, 1983). All the muscles presented higher electromyographic activity in the return phase of the movement. The triceps brachii was the muscle which had higher activity in both phases of the movement. It was concluded that the "front support" exercise is efficient for strength development mainly in the triceps brachii muscle.

Differentiation and numerical analysis of oral yeasts based on SDS-Page profiles. Influence of the culture media on the whole-cell protein extracts.

The application of gel electrophoresis and numerical analysis of yeast soluble proteins analysis to the investigation of 12 oral yeast strains belonging to five species is described. It involves one-dimensional electrophoresis of SDS-solubilized whole-cell proteins using different culture media for the cultivation of the cells, integration densitometries in the areas of the gels and percentages of the proteins extraction. These extracts were prepared from four isolates of Candida albicans, two of C. tropicalis, C. guilliermondii, C. parapsilosis and C. krusei. The extracts from whole-cells proteins using different culture media for the cultivation of the cells were fractionated by slab electrophoresis using a discontinuous buffer system. The corresponding patterns showed at least 36 polypeptides in the range of 14.4-200 kDa. Different isolates of each species were clearly different in each of the five species. The data obtained suggest that different nutritional compositions led to the expression of different proteins derived from alternatives metabolic pathways expressed by the electrophoretic profiles. The construction of a database of protein fingerprints and numerical analysis based on such data, may have some implications in the classification and identification of such species with epidemiological, ecological and taxonomic purposes. A well defined or synthetic culture media seems to be much properly.

Anaerobic sequencing batch reactors for wastewater treatment: a developing technology.

This paper describes and discusses the main problems related to anaerobic batch and fed-batch processes for wastewater treatment. A critical analysis of the literature evaluated the industrial application viability and proposed alternatives to improve operation and control of this system. Two approaches were presented in order to make this anaerobic discontinuous process feasible for industrial application: (1) optimization of the operating procedures in reactors containing self-immobilized sludge as granules, and (2) design of bioreactors with inert support media for biomass immobilization.

Continuous fermentation of sugar cane syrup using immobilized yeast cells.

Several strains of Saccharomyces sp. and commercial Baker's yeast were immobilized by adhesion onto chrysotile, a fibrous magnesium silicate (Mg6Si4O10 (OH)8). The activity of the cells is higher when immobilized, mainly for fermentation of 30 to 50% w/v glucose solutions. In the medium containing 30% w/v glucose, the initial fermentation rate increased 1.2-2.5 times. Yields were in the range of 80.4 to 97.3% for the immobilized cells and 72.7 to 84.5% for the free cells. A packed bed reactor for continuous fermentation was set up using one of the tested strains immobilized onto chrysotile. An average productivity of 20 to 25 g.l(-1).h(-1) was obtained in the first 20 d, and an average of 16 g.l(-1).h(-1) was obtained after 50 d of operation.

In vitro microleakage of composite restorations after nonvital bleaching.

OBJECTIVE: After bleaching treatment, esthetic restorations often need to be replaced due to color changes. Some papers have shown alterations in the bond of adhesive restorations to bleached teeth. The purpose of this study was to evaluate tooth and resin composite adhesion when submitted to nonvital dental bleaching. METHOD AND MATERIALS: One hundred and twenty bovine teeth were assigned to 3 groups (n = 40); paste of sodium perborate and water; 37% carbamide peroxide gel; and no bleaching (control). After 3 weeks of continuous bleaching treatment, standardized Class V cavities were prepared at the cementoenamel junction and restored with Single Bond adhesive system and Z100 resin composite. The samples were thermocycled 1,500 times (5 +/- 1/55 +/- 1 degrees C) with a 1-minute dwell time. Then, they were immersed in a 2% methylene blue solution (pH 7) for 4 hours, sectioned, and analyzed by stereomicroscopy. Microleakage analyses were done, using scores from 0 to 4, considering leakage on the incisal wall (enamel) and the cervical wall (dentin). Data were analyzed by Kruskal-Wallis and Mann-Whitney tests (alpha = 0.05). RESULTS: The results showed that sodium perborate and carbamide peroxide gel significantly increase the microleakage in Class V resin composite restorations to dentin but not to enamel margins. CONCLUSION: The risk of microleakage in dentin margins is increased soon after bleaching treatment.

Influence of agitation rate on the performance of a stirred anaerobic sequencing batch reactor containing immobilized biomass.

The present work reports on the influence of the mechanical agitation rates on the performance of a stirred anaerobic sequencing batch reactor containing immobilized biomass on polyurethane foam, as inert support, treating synthetic domestic wastewater. The reactor was operated at 30 degrees C and an 8-hour cycle was used to treat approximately 0.5 L of the synthetic substrate with a COD concentration of nearly 500 mg/L. The studied agitation rates ranged from no agitation to 750 rpm. The system attained non-filtered substrate removal efficiency greater than 83% when agitation was employed. A very short start-up period and good solid retention could be observed. The use of agitation increased the efficiency of the reactor and enabled reduction of the total cycle time. An empirical equation and a first-order kinetic model are proposed to analyze the influence of agitation rates on the reactor's performance.

Differential pulse polarographic determination of alpha-dicarbonyl compounds in foodstuffs after derivatization with o-phenylenediamine.

A differential pulse polarographic method has been developed for the indirect determination of diacetyl and other vicinal diketones in foodstuffs, based on the quantitative derivatization of the diketones with o-phenylenediamine to give quinoxalines. The method allows the identification of diacetyl and methylglyoxal in brandy, vinegar, wine, and butter samples. The polarographic reduction peaks of the quinoxalines formed are sufficiently separate, in some cases, to allow the simultaneous determination of these two compounds. The polarographic method is not affected by the presence of sulfite ion, probably because the o-phenylenediamine is capable of dislodging the alpha-diketones from their sulfite complexes.

Isolation and amino acid sequence analysis reveal an ancient evolutionary origin of the cleavage stage (CS) histones of the sea urchin.

The cleavage stage (CS) H1, H2A, and H2B histones of the sea urchin, which have previously been identified by their distinct electrophoretic mobility on Triton/acid/urea gels, are known to be maternally expressed during oogenesis and have been implicated in chromatin remodeling of the male pronucleus following fertilization. Here, we describe the isolation of these three CS histones by reverse-phase HPLC chromatography. Moreover, a novel CS H3 protein was identified by the same purification procedure. A low incorporation of radioactive amino acids into the CS histones during early development revealed that the bulk of these proteins in the blastula embryo are derived from the maternal pool of the egg. Amino acid analysis, together with the previously described electrophoretic mobilities, unequivocally identified the purified proteins as CS histones. Peptide sequence analysis confirmed the novel nature of the CS variants as they are distantly related to the early, late, and sperm histone subtypes of the sea urchin. The CS H1 protein displays highest sequence similarity with the H1M (B4) histone of Xenopus laevis, indicating that the frog H1M protein may be a vertebrate homologue of the CS H1 histone. These data suggest an ancient evolutionary origin and wide distribution of the CS histone variants.

Hypothyroidism affects pulsatile LH secretion in pubertal orchiectomized rats.

The present study evaluated the effect of hypothyroidism on the pituitary-testicular axis in rats rendered hypothyroid on the beginning of puberty. Rats were treated with propylthiouracil for 8 weeks and killed for determination of hormonal status and body parameters. For determination of pulsatile LH secretion other animals were orchiectomized two weeks before sampling. Analysis of the results led us to conclude that although the absolute weight of sex organs tended to decrease in hypothyroid animals, the relative weights were equal or higher than control, suggesting that the development of these organs were not affected by hypothyroidism; the androgenic activity of hypothyroid rat testes were preserved; basal plasma levels of pituitary hormones were similar in control and hypothyroid groups; the pulsatile LH secretion showed a decrease in the number of pulses, nadir mean and total LH secretion in hypothyroid animals. Our results demonstrate that although hypothalamic-pituitary axis of hypothyroid pubertal rats displays an abnormal pulsation LH release, no evidences of abnormalities in the reproductive system functions were found.

The bradykinin receptor of the rat uterus affects the ligand properties of charged mercurials.

Using monovalent organic mercurials to modify the response to the oxytocic action of bradykinin (BK) on the rat uterus, a charge effect, related to the reagent applied, was noted. With neutral, and positively charged mercurials, a bimodal pattern of activity, causing stimulation of BK contraction at lower, and inhibition at higher concentrations was observed. With negatively charged mercurials transient stimulation occurred, inhibition only appearing after very large doses and long periods of incubation. Anionic charges of the bradykinin receptor affect the ligand properties of mercurials, repelling negative charges. By themselves, mercurials did not cause any response.
