Differential splicing of transcripts encoding the orphanin FQ/nociceptin precursor.

Orphanin FQ or nociceptin (OFQ/N), the heptadecapeptide agonist for the NOP receptor, is derived by proteolytic processing from a precursor protein, preproOFQ/N. Previous studies have reported alternative splicing between exons 3 and 4 of the mouse OFQ/N transcript, which, upon translation, would yield precursor proteins with different C-termini. Using RT-PCR, we identified similar alternative splicing of preproOFQ/N transcripts in humans and rats. In addition, we identified two novel human preproOFQ/N splice variants from which exon 2 has been excised and which also undergo alternative splicing between exons 3 and 4. Exon 2 contains the translational start site for preproOFQ/N and encodes the signal peptide sequence. In vitro translation of cRNAs lacking exon 2 yields shorter translation products which arise from an alternative initiator methionine located within exon 3. The resulting proteins would lack a signal peptide sequence, which would likely alter their cellular trafficking and processing.

Brain lesions affect penile reflexes.

Electrolytic lesions of several potential brain afferents to the spinal nucleus of the bulbocavernosus (SNB) affect the display of penile reflexes. Ablation of the median and pontine raphe areas significantly potentiates the expression of cups and flips. Animals with a bilateral lesion of the paraventricular nucleus of the hypothalamus have a shorter latency to the first erection but otherwise display normal reflex behavior. Although bilateral destruction of the lateral vestibular nucleus (LVN) completely eliminated penile reflex activity, it also caused significant motor impairment thus clouding conclusions concerning the normal role of the LVN in penile reflex behavior. These and other results support the hypothesis that these brain regions which project to the SNB region normally modulate spinal reflex behavior of the rat penis.

Intergenic suppression of stage II sporulation defects by a mutation in the major vegetative sigma factor gene (rpoD) of Bacillus subtilis.

The Bacillus subtilis intergenic suppressor mutations crsA and rvtA, previously shown to restore sporulation competence to a variety of strains containing stage 0 sporulation defects, also suppress lesions in the stage II sporulation genes spoIIF, spoIIN and spoIIJ. They do not rescue sporulation in other stage II through stage V sporulation mutations. Cells containing spoIIN, spoIIF96 and spoIIJ::Tn917 mutations fail to transcribe spoIID, a late stage II gene. Introduction of crsA47 into spoIINts279, spoIIF96, or spoIIJ::Tn917 mutant backgrounds circumvents the need for the spoIIF, IIN, and IIJ products, restoring both expression of spoIID, and sporulation competence.
