Anisotropic dynamics in a shaken granular dimer gas experiment.

The dynamics, velocity fluctuations, and particle-plate interactions for a two-dimensional granular gas of shaken, nonspherical particles are studied experimentally. The experiment consists of a horizontal plate that is vertically oscillated to drive the dynamics of macroscopic dimers, spherical pairs that are loosely connected by a rod that couple the interaction each of the spheres has with the shaking plate. The extended nature of the particles results in more than one energy-momentum transfer between the plate and each dimer per shaking cycle. This complex interaction results in anisotropic behavior for the dimer that is a function of the shaking parameters.

A long-term follow-up of surgically treated vesicoureteric reflux in girls.

Ninety-six girls with surgically treated vesicoureteric reflux (VUR) operated on between 1970 and 1975 were studied to determine their post mature growth (height and weight); the incidence of hypertension; the incidence, progression, and development of new renal scars; the incidence of urinary infection; and the outcome of any pregnancies. The average age at follow-up was 20 years 2 months (range, 16 years to 25 years). The final "post mature" height was significantly greater than that of a group of controls from all regions of Great Britain; the incidence of hypertension was 4.5% and the incidence of urinary infections was 37.6%. The majority of subsequent pregnancies were uneventful but urinary infections occurred in 36.4% and 3 of the 20 infants born had VUR.

Role of oxygen free radical formation in the mechanism of menogaril resistance in multidrug resistant tumor cells.

The mechanisms of action and resistance to menogaril, a clinically active anthracycline antitumor drug, were evaluated in sensitive and doxorubicin-selected multidrug resistant human breast tumor (MCF-7) cell lines. While MCF-7/ADRR cells were highly resistant (250-500-fold) to doxorubicin, they displayed only marginal resistance (10-fold) to menogaril. In contrast to doxorubicin, the mechanism of resistance to menogaril in these cells does not involve differential inhibition of DNA synthesis as measured by thymidine incorporation. P-170-glycoprotein-dependent drug transport did not contribute to resistance as there was no difference in the accumulation and retention of menogaril by sensitive and resistant cell lines. However, there was a 2-fold decrease in oxygen free radical formation in the resistant cells, compared to sensitive cells, in the presence of menogaril. Since resistant cells contain 12-fold higher glutathione peroxidase activity than the parental sensitive cells, the detoxification of hydrogen peroxide may be responsible for the decreased free radical formation and thus, may play a role in the resistance to menogaril.

Differential oxygen radical susceptibility of adriamycin-sensitive and -resistant MCF-7 human breast tumor cells.

Recent evidence supports the concept that Adriamycin cytotoxicity may be mediated by drug semiquinone free radical and oxyradical generation. We tested this hypothesis further by exposing drug-sensitive (WT) and 500-fold Adriamycin-resistant MCF-7 human breast tumor cells (ADRR) to exogenous superoxide- and hydrogen peroxide-generating systems and subsequently monitored cell proliferation as a measure of cytotoxicity. The ADRR tumor cells tolerated a 4-fold greater exposure than sensitive cells to superoxide generated by the xanthine/xanthine oxidase system. Likewise, exposure to hydrogen peroxide produced by the action of glucose oxidase on glucose revealed a 4-fold diminished susceptibility of the drug-resistant cells to this reduced form of oxygen. Similar results were obtained by the direct application of hydrogen peroxide to cells. For both cell lines, cytotoxicity was dependent upon the magnitude and the duration of reactive oxygen exposure. When WT and ADRR cells were cultured under hyperoxia (95% O2:5% CO2), in order to stimulate the intracellular production of oxyradicals, proliferation was inhibited to a greater extent in the drug-sensitive cell line. Additionally, hyperoxia potentiated the cytotoxicity of Adriamycin to both sensitive and drug-resistant cells, but the effect depended upon the concentration of the drug. Under hyperoxic conditions, Adriamycin caused oxygen radical-dependent cytotoxicity to the WT tumor cells at clinically relevant drug concentrations as low as 2 to 3 nM. With ADRR tumor cells, hyperoxia increased the cytotoxicity of Adriamycin at concentrations above 5 microM. Paradoxically, both the WT and the ADRR tumor cells were equally susceptible to the cytotoxic effects of gamma irradiation. It is known that the Adriamycin-resistant MCF-7 cells greatly overexpress glutathione peroxidase and glutathione transferase activities; however, other biochemical defenses against reactive drug intermediates and oxygen radicals have been reported to be similar in the two cell lines. We have reexamined those observations in this report. The resistance of ADRR breast tumor cells to Adriamycin appears to be associated with a developed tolerance to superoxide, most likely because of a twofold increase in superoxide dismutase activity, and a decreased susceptibility to hydrogen peroxide, most likely because of 12-fold augmented selenium-dependent glutathione peroxidase activity. Acting in concert, these two enzymes would decrease the formation of hydroxyl radical from reduced molecular oxygen intermediates.(ABSTRACT TRUNCATED AT 400 WORDS)

The role of the children's nurse in the community.

In 1969 a pediatric home nursing service was started in Southampton to care for sick children at home. The primary role of the children's nurse in the community is discussed with particular reference to day case surgery, early discharge, care of the mentally and physically handicapped, pediatric medical care, prevention of a hospital admission, and care of the terminally ill. The secondary roles of teaching and liaison between the hospital and the primary health care team is discussed. After 10 yr experience with over 12,000 admissions to the day unit we believe that the majority of general surgical procedures such as circumcision, inguinal herniotomy, and orchidopexies can be safely undertaken on a day basis. The provision of adequate nursing care in the community which is closely linked with the hospital has been one of the main reasons for the success of the scheme.
