Electron spin echo envelope modulation studies of pyruvate kinase active-site complexes.

Electron spin echo envelope modulation (ESEEM) spectroscopy, with Mn2+ and VO2+ as paramagnetic probes, was used to examine active-site structures at the protein-based divalent cation site of rabbit muscle pyruvate kinase in the presence of substrates, products, and the requisite inorganic cofactors. Two different VO.protein complexes were clearly distinguished, which differed with respect to coordination of the active-site lysine to VO2+. Lysine coordination was sensitive to the presence of pyruvate and phosphoenolpyruvate (PEP) and to the nature of the monovalent cation. In the presence of MgATP and oxalate, a 4-MHz 31P contact interaction was observed, which indicates that the ATP is directly coordinated to Mn2+ at the protein-based site. No 31P contact interactions were observed, however, in the presence of PEP. Pyruvate was determined to be a bidentate ligand of VO2+, on the basis of the observation of 2.2- and 5.4-MHz 13C contact interactions between VO2+ and [1-13C]pyruvate and [2-13C]pyruvate, respectively. Magnetic coupling between VO2+ or Mn2+ and 23Na, 39K, and 133Cs was observed, demonstrating the close proximity of the monovalent cation and the protein-based divalent cation.

Electron spin-echo spectroscopic studies of Escherichia coli fumarate reductase.

Electron spin-echo envelope modulation (ESEEM) spectroscopy was applied to the study of reduced Centre 1 of Escherichia coli fumarate reductase (succinate:(acceptor) oxidoreductase, EC 1.3.99.1). The ESEEM spectrum derived from stimulated (3-pulse) echo envelopes obtained at 8.8 GHz contained lines at 0.9, 2.1, 3.0 and 4.2 MHz in the g = 1.94 region. When studied at 11.4 GHz, these low-frequency components scale with magnetic field in a manner indicating interaction between the unpaired electron spin of the Fe-S cluster and a weakly coupled 14N nucleus. Spectral simulations of these ESEEM data yield nuclear quadrupole interaction parameters indicative of peptide nitrogen. For oxidized protein, the magnetic-field dependence of the linear electric-field effect (LEFE) for Centre 3 was measured, and the results confirm the presence of a [3Fe-4S] cluster in the protein.

Canine rhinosporidiosis in Arkansas.

Rhinosporidiosis is a fungal infection caused by Rhinosporidium seeberi, a microorganism of complicated, undetermined life cycle. Many species of animals are susceptible to rhinosporidiosis, including human beings. The frequency of occurrence of the disease in domestic animals is unknown. A one-year-old male Collie dog from Northeast Arkansas was found to have rhinosporidiosis presenting as an intranasal polypoid mass. We believe this is the first documented report of rhinosporidiosis from the State of Arkansas.

The meaning of AIDS: African American nursing students' perceptions of providing care to AIDS patients.

A qualitative study was conducted to explore the perceptions of African American nursing students as they provided care to acquired immunodeficiency syndrome (AIDS) patients. This research addressed the following question: What are the lived experiences of African American nursing students when they provide care to AIDS patients? The participants in this study were a convenience sample of 12 associate degree nursing students enrolled full-time at a historically Black college and university located in Virginia. Data were collected through audio taped interviews. Five dominant themes emerged: 1) fear of exposure and transmission; 2) hopelessness about the situation; 3) self-identification with the individual; 4) empathy toward persons with AIDS (PWAs); 5) and spirituality and belief in an afterlife. Female nursing students were able to identify with their AIDS patients while male nursing students were not. Many of the nursing students felt ostracized by their peers, and a lack of knowledge hindered the nursing students' deliverance of compassionate nursing care.
