Human recombinant interferon-alpha-2 in the treatment of patients with hairy cell leukemia.

During the last 2 years, we have treated 14 hairy cell leukemia (HCL) patients with human recombinant interferon-2 alpha (Boehringer Ingelheim). The above group consisted of eight nonsplenectomized and six previously splenectomized progressive HCL patients. The patients received daily doses of 5 x 10(6) units of IFN for 3 months and two doses per week for the next 3 months thereafter by i.m. route. The therapy resulted in the complete (nine cases, = 64.3%) or partial (two cases = 14.3%) clinical and hematological remission (response rate 78.6%), with either disappearance or marked reduction in circulating and bone marrow hairy cells, decreased spleen size, and recovery of normal hemopoiesis. Apart from a transient flulike syndrome during the first 2 weeks of therapy, no other side effects were observed.

Lysosomal enzymes of neutrophils in women with breast cancer.

Activity of acid phosphatase, beta-glucuronidase and N-acetyl-beta-D-glucosaminidase in peripheral blood neutrophils of 30 women with breast cancer has been studied by means of semiquantitative cytochemical methods. In comparison to 20 healthy women the patients showed an increased activity of acid phosphatase and beta-glucuronidase whereas that of N-acetyl-beta-D-glucosaminidase was significantly lowered. It has been suggested that observed intracellular enzymatic deficiency is of importance with regard to lowered antitumor activity of neutrophils. Analogous deficiency of N-acetyl-beta-D-glucosaminidase within the neutrophils has previously been observed by the authors in women with malignant tumors of reproductive organs.

Acid phosphatase in peripheral blood lymphocyte lysosomes in patients with diabetes mellitus.

In 55 patients with diabetes mellitus metabolically balanced the activity and intracellular localization of acid phosphatase has been estimated by means of semiquantitative method of Barka and Anderson [3] in peripheral blood lymphocytes. It has been shown that patients in question, both insulin-dependent and those with overweight, in comparison to control group, exhibit more than twofold lower numbers of lymphocytes having intact enzyme positive lysosomes and more than twofold higher numbers of that cells with damaged lysosomes and extralysosomal localisation of the enzyme. According to authors suggestion the above changes may be associated with disturbed lymphocyte functions in the patients with diabetes mellitus.

Lithium carbonate effect on nitrotetrazolium blue (NBT) production in neutrophils of patients with granulocytopenia.

In 30 persons with essential granulocytopenia, lithium carbonate in doses of 12.5 mg/kg per body weight was applied during 10 days. This resulted in an increased absolute neutrophil count and total leukocyte count as well as increased percentage of cells reducing NBT in the spontaneous NBT test according to Park. Prior to the beginning of treatment, patients with granulocytopenia exhibited an elevated percentage of NBT-positive cells in comparison to the control group of 28 healthy persons. The significance of this observation for treating granulocytopenia by means of the agent mentioned above is discussed.

Enzymes of neutrophils in women with malignant tumors of reproductive organs.

Enzymatic equipment in peripheral blood neutrophils has been determined in 10 women with cervical carcinoma, 5 with carcinoma of the uterus body and 30 women with myomas of the uterus. Using cytochemical techniques the authors observed the intracellular deficiency of N-acetyl-beta-glucosaminidase accompanied by diminished absolute count of the enzyme-positive cells as well as the beta-glucuronidase- and the myeloperoxidase-positive neutrophils. Lowered activity of the myeloperoxidase was another pattern of cells in question. The acid phosphatase activity of neutrophils in the patients was significantly elevated. It has been suggested that observed intracellular deficiencies of selected enzymes within the neutrophils are of importance with regard to lowered antitumor activity of that cells operating mainly through the myeloperoxidase-H2O2-halide system.
