Isolation and rapid sequence characterization of two novel bovine beta-lactoglobulins I and J.

Two novel bovine beta-lactoglobulins I and J have been isolated from bovine milk and characterized by isoelectric focusing. Their primary structure was determined by a very rapid method consisting of a combination of Edman sequencing, mass analysis, and ladder sequencing by mass spectrometry. We found that both new beta-lactoglobulins are of the bovine beta-lactoglobulin B-variant type. beta-lactoglobulin I shows Gly instead of Glu at position 108, whereas beta-lactoglobulin J shows a Pro-to-Leu exchange at position 126.

[Working conditions and health status from the workers' viewpoint].

In the present study details about disruptive factors in working conditions, elements injurious to health or general well-being, personal attitudes to work etc. have been obtained by means of a representative survey of about 2000 employed persons in Switzerland. The most frequent disruptive factors in working conditions (> 30%) are noise and too little time for family and friends. A relatively large number of employed persons (20-25%) complain about unsatisfactory ventilation, air pollution due to dust, smoke etc., draughts, uncomfortable temperature, dry air, lifting and carrying heavy weights, uncomfortable body positions at work, unsuitable working hours, as well as pressure on time, too much concentration required, pressure of responsibility, insufficient recognition and lack of information about the planning and results of work being done. In the field of health complaints, back problems and nervous tension are at the top of the list (39%), followed by fatigue, sleeping problems, neck or leg problems, burning eyes, and stomach and intestinal problems (25-34%). The analysis shows that interrelationships exist--sometimes in the sense of a vicious circle--between complaints about disruptive factors in work conditions, professional groups, health problems and attitudes to professional live. The results provide a comprehensive survey of the incidence of problems in the fields of worker protection.

Genetic variants of bovine beta-lactoglobulin. A novel wild-type beta-lactoglobulin W and its primary sequence.

A novel bovine beta-lactoglobulin W has been isolated and its complete primary structure is presented. It was isolated by chromatofocusing of a beta-lactoglobulin AW heterozygote and purified by recrystallization. During sequencing of the oxidized protein, it became evident that the new beta-lactoglobulin W is a subtype of variant B with a single difference at position 56. This Ile----Leu substitution, which means a shift of a methyl group from C-beta to C-gamma of the amino-acid side chain causes a change of pI of 0.007 units, which can be detected by high resolution electrophoresis. This Ile56 amino-acid residue is among the most conserved residues with the exception of kangaroo beta-LG. The structures of other bovine beta-lactoglobulins and their relationships are discussed.

Isoelectric focusing in immobilized pH gradients with carrier ampholytes added for high-resolution phenotyping of bovine beta-lactoglobulins: characterization of a new genetic variant.

The genetic variants of bovine beta-lactoglobulin (beta-lg) from the "Murnau-Werdenfelser" breed were analyzed in three different isoelectric focusing (IEF) systems. While carrier ampholyte IEF with a pH gradient of 0.2 pH/cm did not resolve the new variant W from the B variant and IEF in immobobilized pH gradients (IPG) with 0.1 pH/cm only partially resolved it, adequate separation was achieved with IPG-IEF in a pH 5.25-pH 5.7 gradient, in presence of 0.8 % w/v carrier ampholytes, both over a 10 and 17 cm separation distance. Apparent isoelectric points (pI's) and genetic frequencies (f) were as follows: beta-lg A, pI = 5.370, f = 0.364; beta-lg B, pI = 5.485, f = 0.480; beta-lg W, pI = 5.492, f = 0.076; and beta-lg D, pI = 5.610, f = 0.080. The small difference of delta pI = 0.007 between beta-lg B and beta-lg W respectively, seems to originate from a "silent" substitution of neutral amino acid residues as compared to the larger delta pI's of the other genetic variants of beta-lg, which result from substitution of charged amino acids.

[Occupational health-related organization of work in large laundry facilities].

In the ergonomic assessment of work in 14 large laundries, it is primarily constrained posture due to working conditions and uniform overstrain that are highlighted objectively. Accordingly common are complaints about back problems, leg pain, severe fatigue, headache and nervousness (42-70%). The added work difficulties most objected to include heat (rate of mention in flatwork operations 80%), dust, dirt, odors (in sorting operations 75%), constant standing and walking (in several areas of work 70-80%), too great exertion of the arms (in sorting and folding operations 50-67%), and noise (depending on area of work 33-73%). On the basis of these results, attempts at resolving these problems to improve the organization of work in large laundries were developed. These include for example, an adequate capacity to adjust the working surfaces, suitable seats and aids for standing, installation of platforms, and further measures to reduce overstrain.

[Occupational medicine insight into the problem of occupational stress in the construction industry].

According to the present study, in addition to the high accident rate, work in the building trade is characterized by intense muscle strain and constrained physical posture. In bricklaying, for example, a worker moves by hand up to seven tons of stone per shift. Consequently, various health complaints occur far more frequently in this occupation than in the other occupational groups, for instance, back complaints (71%), severe fatigue (58%), pain in the arms and hands (42%), headache (40%) and leg pain (34%). The most complained of factors include effects of the weather (74%), draft (50%), noise (55%), air pollution via dust (53%), skin contact with dirt and harmful substances (47%), moving heavy objects by hand (53%), adverse posture during work (47%), and a high risk of accidents (47%). Besides technical and organizational measures, the prevention of health impairments in employees in the building trade should be promoted by purposive care by works services of occupational medicine.

[Ergonomic problems in cash register jobs at self-service stores].

By means of inquiry and work place inspection health and ergonomic problems of jobs on diverse cash register systems were investigated, taking into account larger shopping centers as well as smaller self-service shops in various parts of Switzerland. A lot of work place inconveniences, e.g. current of air = 62%, tiresome goods handling = 31%, limited scope for movement = 30%, unsuitable seat = 23%, etc., as also construction and arrangement short comings related to definite types of cash register boxes, obviously resulting in an increased incidence of bodily ailments (back pain = 55%, arm pain = 43% visual troubles = 42%, etc.) were identified. Based on this investigation and on ergonomic analysis of different types of work place arrangements, recommendations were made on the planning and construction of cash register set-ups and on the organisation of work.

Biochemical polymorphism in Egyptian Baladi cattle and their relationship with other breeds.

Gene frequencies were estimated in a sample of Baladi cattle for milk proteins, blood proteins and blood groups. Gene frequency estimates of Bos taurus, Bos indicus and Sanga breeds were assembled from the literature. The gene frequencies were utilized for estimating the genetic distance between the breeds and breed groups. The Egyptian Baladi cattle appeared to be closer to Bos taurus breeds than to the Sanga. They are far removed from Zebus.

[Occupational diseases in forestry workers].

Inquiries and medical examinations provided a considerable amount of informations on the state of health of forest workers in Switzerland. A large number of forest workers suffers from back troubles (55%) and other health problems concerning the muscular-skeletal system. These are obvious reasons for giving up forestry work before reaching the retiring age limit. Complaints about overfatigue and overall body degeneration are frequent and point to the necessity of further investigations. Vibration induced "White Fingers" are frequent amount forest workers (46%). They are closely related to total exposure time. During recent years among professional forest workers there has been a change-over to chain-saws with vibration damping devices (97%) and, to some extend, to chain-saws with heated handgrips. This seems to lead to a reduction of White Finger symptoms. Chain-saw exhausted gas continues to be a problem requiring solution.

[Medical aspects of stress measurement in the workplace].

To protect workers from overstress by occupational work, there are some methods to measure their working stress by recording different parameters continually during a period of several hours or days. The following physiological parameters are taken into consideration: heart rate, ECG, EMG, movement activities, breathing rate, oxygen consumption, body temperature, resistance of the skin and EEG. Periodically repeated measurements of the lactic acid in capillary blood as well of the catecholamines in the urine are also suitable. The heart rate is till now the most important parameter for practical use to estimate the physical and psychical strain. For values which can be registered by an electrical signal, three modern pickup systems are available: telemetry, magnetic tape recording, solid state memory. The most important advantage of the solid state memory used for the long time registration of bioelectrical signals (except those of high frequency) is actually evident. As the Laboratory of Occupational Medicine of the Federal Office for Industry and Labour is setting up a system to evaluate the individual psychical and physical stress, there has been developed a small memocardiometer which is able to record the heart rate during about 35 hours. The stored datas are transmitted via an interface to the computer HP-85, to be immediately available for calculations, tabulating and plotting.

[Development of occupational medicine in industrialized countries].

The quality of the occupational medical care could be improved by considering the following points: -- the participation of occupational physicians on inspections of enterprises should be increased, -- the medical supervision instances of work protection should have efficient laboratories, -- the occupational health services in the enterprises should be established; they should have a sufficient number of trained personnel and be sufficiently equipped with instruments, -- the institutes of occupational medicine at the universities should be developed, -- following the circumstances, there should also be established central institutes for occupational health.

[Physiological characteristics during exertion of Swiss Alpine and urban pubertal children].

On account of standardized examinations of circulatory and respiratory responses during the ride on the bicycle ergometer the physiological characteristic of Swiss children population from 12 to 15 years of age living in alpin and urban habitats is reported. The average maximum oxygen intake per kg body weight was 49-50 ml/min for alpin boys, 45-47 for urban boys, 39-41 for alpin girls and 37-39 for urban girls. Except for somatometric values, there is no other significant difference between alpin and urban children from the physiological point of view. A comparison of presented results with data from abroad does not show any disadvantageous properties of cardiorespiratory functions for Swiss boys and girls.
