Cell cultures were established from the biopsies of lung, skin and kidney from each of nine human fetuses, and chromosome analyses were performed on material through the fifth subculture. Kidney cell cultures generally showed a higher level of polyploidy than lung or skin. The frequencies of hyperdiploid cells and those with structural abnormalities were consistent with the low levels found in cultures of human lymphocytes. The data provide a normal cytogenetic baseline for human fetal material which may be useful in a variety of studies.
Primary cultures of African green monkey kidney and rabbit kidney as well as diploid cell lines WI-38 and DBS-FRhL-2 were examined for evidence of tumorigenicity and latent RNA tumor viruses. Cells inoculated into immunosuppressed newborn hamsters and rhesus monkeys were not tumorigenic. Cells treated with 2'-deoxy-5-iodouridine to induce the production of latent viruses were examined by electron microscopy, density gradient centrifugation, and the reverse transcriptase enzyme assay. No evidence was found for RNA tumor viruses by the biochemical or biophysical methods used. The results indicated that each type of mammalian cell currently used in the production of virus vaccines would be acceptable for these parameters of safety if similar control procedures were applied at the time the vaccines were manufactured.
Use of immunoadsorbents to remove bacteriophages from tissue culture serum was investigated. Immune globulins from rabbit antiserum prepared against phi V-1 phage were immobilized by covalent linkage to activated porous silica glass derivatives of p-aminoarylamine and to Sepharose-4B. Chromatographic columns of each material were used to filter samples of a fetal bovine serum into which had been introduced 8100 plaque-forming units of the phage per ml. Efficiency of removal was determined by plaque assays of phi V-1 phage recovered in the effluent fluids. Activated but uncoupled matrices nonspecifically removed from 49 to 59% of the phages introduced into the experimental serum. A reduction of 35 to 37% in phage content occurred in the serum after filtration through columns coupled to nonantibody protein. With specific immune globulins attached, the Sepharose-4B matrix reduced the concentration of phage in the serum below a detectable quantity. Noapparent alterations occurred in the growth-promoting property of serum filtered through the Sepharose-4B immunoadsorbent as measured by cloning efficiency of BHK-21, WI-38, and FRhL-2 cells. These experiments serve as a model system for use of immunoadsorbents for selective removal of bacteriophages and perhaps other extraneous microbial agents from tissue culture serum.
The ultrastructural characteristics of galactosaemic human fibroblasts exposed to media with various sugars were compared. Considerations of known biochemical pathways suggested that cells without the galactokinase enzyme and exposed to galactose would be functionally equivalent to cells in a medium without any sugar source. We found the ultrastructural characteristics of such cells to be consistent with that hypothesis. In addition, we unexpectedly found that homozygous galactosaemic fibroblasts, exposed to medium with glucose as the energy source, showed evidence of degeneration, such as autophagic vacuoles. Degenerative changes were not found in fibroblasts heterozygous for the galactokinase enzyme exposed to glucose. The implications and uses of these findings are discussed.
Rhesus and African green monkeys were treated with dexamethasome, antithymocyte globulin, 60Co irradiation, or combinations of those immunosuppressive agents, and the leukocyte response was followed. The lymphocyte population could be significantly depressed with 60Co, but dexamethasone was required to maintain a low lymphocyte count. The ability of the immunosuppressed animals to support the growth of transplanted tumor cells was markedly enhanced by the addition of ATG to the treatment schedule, even though ATG had no effect on the numbers of leukocytes in the peripheral circulation.
Continuing follow-up observations and study of information as derived from a large cohort of fetal recipients of intrauterine transfusion (IUT) is utilized to provide estimates of maternal and fetal risks associated with the transfusion procedures, subsequent maternal complications of the pregnancy, and the health, intelligence, and behavior of the erythroblastotic recipient. The upper limit of risk of the procedure is estimated to be 7.2 per cent. Fourteen per cent of hydropic fetuses survived through the perinatal period as compared with 47 per cent of nonhydropic fetuses. One infant developed acute lymphocytic leukemia (ALL) at age 4 1/2 years. The IUT survivors showed a marked correlation among the frequencies of abnormalities in the three areas measured: physical, intellectual, and social maturity. Only about 50 per cent were considered normal in these parameters of health. Maternal complications were observed in only 2 per cent of the cases.

The ribonucleic acid-dependent deoxyribonucleic acid polymerase assay was used to detect type-C viruses in live virus vaccines. Conditions were first established to maximize the sensitivity of the assay. Vaccines tested included live poliomyelitis, rubella, measles, mumps, and yellow fever. Only yellow fever and measles vaccines known to have been produced in avian leukosis-contaminated cells showed evidence of type-C viruses using the assay. The result of the survey show that the assay has direct practical application to the problem of detecting latent agents in biological products intended for human use.

A bacteriophage, isolated from a live measles vaccine and designated varphiV-1(L), was intentionally introduced into cell culture systems. The bacterial virus persisted in the cell cultures after daily medium changes and trypsinization, and in cell lysates prepared by freezing and thawing. The results suggest that phages contaminating cell cultures may be carried over into the final product during the manufacture of viral vaccines.
