The possible role of the spleen in the reduction of nitroblue tetrazolium by neutrophils.

Patients with sickle cell anaemia (SCA) and individuals who have undergone splenectomy have an increased susceptibility to bacterial infections. Reports of variable capacity of reduction of nitroblue tetrazolium (NBT) by neutrophils from patients with SCA suggest that there may be a polymorphonuclear dysfunction in this disease. In order to assess the role of the spleen in this neutrophil abnormality we carried out the NBT test on neutrophils from normal or splenectomized individuals and from patients with SCA, sickle cell trait and sickle-cell-beta-thalassaemia (S-thal). Decreased NBT scores were observed in SCA and in splenectomized control individuals. In the S-thal group, most of the non-splenectomized patients had normal or near normal NBT scores while all splenectomized patients exhibited decreased values. These results indicate a possible contribution of the spleen in the pathogenesis of this neutrophil dysfunction. The mechanism of this participation is unknown.

Severe strongyloidiasis associated with hypogammaglobulinaemia.

The association of severe strongyloidiasis and hypogammaglobulinaemia in one patient is described. Severe strongyloidiasis was observed twice (March 1971 and June 1974), and the Strongyloides stercoralis infestation probably lasted for at least 58 months (from March 1971 to December 1975), in spite of successive thiabendazole courses of treatment. Immunological studies performed disclosed severe, steady hypogammaglobulinaemia but no abnormality in cell-mediated immunity. The respiratory infections also seen during the follow-up were ascribable more to humoral immunity impairment than to cell-mediated immunity impairment. This is the first report of the association of hypogammaglobulinaemia and severe strongyloidiasis, and it suggests that humoral immunity plays a relevant role in the human defences against S. stercoralis infestation.

Lymphocyte subpopulations and neutrophil function in chronic human Chagas' disease.

The absolute numbers of total leukocytes, lymphocytes. T cells, helper/inducer, suppressor/cytotoxic and B cells were decreased in the peripheral blood of patients with chronic Chagas' disease. Since antilymphocyte antibodies were present only in a minority of patients they probably cannot account for the abnormalities in lymphocyte subsets. Patient neutrophils stimulated with endotoxin-treated autologous plasma showed depressed chemotactic activity and this seems to be an intrinsic cellular defect rather than plasma inhibition. Random migration of neutrophils was normal. Reduction of nitroblue tetrazolium by endotoxin-stimulated neutrophils was also decreased. These findings further document the presence of immunosuppression in human Chagas' disease. They may be relevant to autoimmunity, defense against microorganisms and against tumor cells at least in a subset of patients with more severe abnormalities.

Applications of flow cytometry to hematopoietic stem cell transplantation.

Applications of flow cytometry to clinical and experimental hematopoietic stem cell transplantation (HSCT) are discussed in this review covering the following topics: diagnosis and classification of lymphohematologic disorders, quantitation of hematopoietic progenitors in the graft, lymphohematopoietic reconstitution following HSCT and animal models of human HSCT. At the end, the utilization of flow cytometry in clinical HSCT by Brazilian transplant centers is briefly reviewed.
