A catalogue of molecular, physiological and symbiotic properties of soybean-nodulating rhizobial strains from different soybean cropping areas of China.

We have analysed 198 fast-growing soybean-nodulating rhizobial strains from four different regions of China for the following characteristics: generation time; number of plasmids; lipopolysaccharide (LPS), nodulation factors (LCOs) and PCR profiles; acidification of growth medium; capacity to grow at acid, neutral, and alkaline pH; growth on LC medium; growth at 28 and 37 degrees C; melanin production capacity; Congo red absorption and symbiotic characteristics. These unbiased analyses of a total subset of strains isolated from specific soybean-cropping areas (an approach which could be called "strainomics") can be used to answer various biological questions. We illustrate this by a comparison of the molecular characteristics of five strains with interesting symbiotic properties. From this comparison we conclude, for instance, that differences in the efficiency of nitrogen fixation or competitiveness for nodulation of these strains are not apparently related to differences in Nod factor structure.

[Cloning and characterization of three novel genes encoding transmembrane proteins of Schistosoma japonicum].

OBJECTIVE: To clone and analyze novel antigen molecules of Schistosoma japonicum (Sj), and to provide effective vaccine candidate antigens against schistosomiasis japonica. METHODS: Sj adult cDNA library was screened using sera of mice infected with Trichinella spiralis (Ts) and the inserts of positive clones were specifically amplified by PCR. The positive clones were sequenced and the sequence data were analyzed using Nucleotide BLAST software of NCBI and Expert Protein Analysis System of Swiss Institute of Bioinformatics. RESULTS: Nine positive clones were obtained after three rounds of immunoscreening. The size of these inserts ranged from 0.6 kb to 2.1 kb. Among five novel genes, Sj-Ts1, Sj-Ts3 and Sj-Ts5 (GenBank accession number: AY005816, AF299080 and AY024352, respectively) encode trans-membrane proteins with 83, 83 and 233 amino acids, respectively. Sj-Ts1 protein predicted contains one possible trans-membrance helix, one N-myristoylation site, two phosphorylation sites for protein kinase C and one for tyrosine kinase, Sj-Ts3 protein contains two possible transmembrance helices and one casein kinase II phosphorylation site, whereas Sj-Ts5 protein has five possible transmembrance helices, one N-glycosylation site, one N-myristoylation site, two phosphorylation sites for cAMP- and cGMP-dependent protein kinase and four for protein kinase C and one for casein kinase II. CONCLUSION: Three novel genes encoding three transmembrane proteins might be developed as new vaccine candidates against Sj infection.

[Immunoscreening of Schistosoma japonicum adult worm cDNA library by sera from rabbits vaccinated with ultraviolet-attenuated cercariae].

OBJECTIVE: To find out the major antigenic molecules of radiation-attenuated cercariae, and provide some useful candidate antigens for developing schistosomiasis vaccine. METHODS: Schistosoma japonicum (Sj) adult worm cDNA library was screened by sera of rabbits vaccinated with ultraviolet-attenuated cercariae, and the inserts of positive clones were specifically amplified by PCR and sequenced. RESULTS: Ten positive clones were obtained after three rounds of screening, and the size of Sj cDNA fragments of the positive clones ranged from 1.5 kb to 1.8 kb. Five partial sequences were got after preliminary sequencing. Two of them had significantly homology with Sj dynein light chain 5(DLC 5) gene and Sj mitochondrial gene, respectively, and the others were identified as partial sequences of novel genes for they showed only partial homology with non-schistosome genes or other organism in the database. CONCLUSIONS: The positive clones may be the genes encoding the antigens that can elicit protective immunity against Sj.

[Experimental study on immunization of mice with nucleic acid vaccine Sj31BIN combining IL-12 against Schistosoma japonicum].

Based on the anti-fecundity effect of Schistosoma japonicum nucleic acid vaccine Sj31BIN, we combined Sj31BIN with IL-12 in vaccination of mice to explore the role of IL-12 as an adjuvant. The result showed that immunization of the mice with Sj31BIN + IL-12 led to a significant decrease in adult worm recovery, a liver egg count reduction of 59.74%, an intestine egg count reduction of 59.60% and a liver surface egg granuloma reduction of 71.3%. In addition, vaccination of the mice with IL-12 alone also led to some but not significant decrease in adult worm recovery and the egg counts. It is concluded that Sj31BIN plus IL-12 induces significant anti-fecundity immunity.

[Expression and purification of recombinant Schistosoma japonicum paramyosin].

Paramyosin of Schistosoma japonicum was expressed at a high level in E. coli. The recombinant protein could be easily purified from bacteria lysate by fast protein liquid chromatography(FLPC) on a TALON resin column, due to the protein being expressed with a tag of six histidine residue fused to the N-terminus. The protein was completely soluble and could be eluted under non-denaturing condition using imidazole. To eliminate imidazole and residue of E. coli, the elution was further purified by ion-exchange chromatography. The purified protein will be used in water buffaloes in the study on protective immunity against Schistosoma japonicum.

[Preliminary report on immunoscreening of Schistosoma japonicum adult worm cDNA library by sera from infected Microtus fortis].

Microtus fortis (Mf) is resistant to Schistosoma japonicum (Sj) infection. To explore if Mf responses to special Sj antigens after infection, Sj adult worm cDNA library was screened with sera from infected Mf. Twelve positive clones were obtained after rescreening. After automatically excised with the helper phage, positive clones were further identified by PCR. The size of Sj cDNA fragments in positive clones ranged from 300 bp to 1.8 kb. Six of these fragments were 300 bp, one was 1 kb, and five were 1.8 kb in size. The results indicate that the infected Mf serum could recognize Sj specific antigens. It is valuable to further study the immune protection of these cloned molecules.

[Improvement of nitrogen fixation efficiency and plasmid stability in Bradyrhizobium japonicum by the introduction of dctABD and parCBA/DE genes].

A recombinant plasmid pHN207 containing C4-dicarboxylic acid transport genes (dctABD) from Sinorhizobium meliloti, parCBA/DE genes from pTR102 and reporter genes luxAB from pDB30 was constructed by using pLAFR3 as the vector. The pHN207 was then introduced into the Bradyrhizobium japonicum TA11 and CB1809 by bi-parental mating. It was confirmed that parCBA/DE genes could increase the stability of pLAFR3 in the transconjugants under both free-living and symbiotic condition. The results of plant pot experiment indicated that the introduction of dctABD genes could significantly improve the symbiotic nitrogen fixation efficiency of TA11 and CB1809 with soybean varieties of Heilong 33, Ningzhen No. 1 and Yudou No. 1. Compared with the control, the shoot dry weight (biomass) and total nitrogen content of the plants tested were significantly increased.

[Evaluation of Schistosoma japonicum tegumental membrane antigen (SjMAg) in immunodiagnosis and efficacy assessment].

OBJECTIVE: To evaluate the specificity and sensitivity of SjMAg in detecting specific antibodies in sera of patients with schistosomiasis japonica and in assessing therapeutic efficacy. METHODS: SjMAg-ELISA was used to determine the specific IgG and IgG4 in sera of patients with schistosomiasis japonica at different time points after chemotherapy. RESULTS: SjMAg-ELISA was as sensitive and specific as SEA-ELISA. The negative conversion rate of IgG detected by SjMAg-ELISA 12 months after treatment was 80.0%, being significantly higher than 43.3% by SEA-ELISA. The negative conversion rate of IgG4 detected by SjMAg-ELISA 12 months after treatment was 93.3%, being significantly higher than 60.0% by SEA-ELISA. The negative conversion rates of IgG and IgG4 2 yr after treatment were 92.9% and 97.6% respectively. CONCLUSION: SjMAg-ELISA is comparable to SEA-ELISA in diagnosing schistosomiasis japonica and is more effective in assessing therapeutic efficacy.

[High dose cisplatin combination chemotherapy for advanced breast cancer--a report on 50 cases].

Fifty patients with advanced breast cancer were treated by high dose cisplatin (DDP) combination chemotherapy during the period of August 1984 to June 1991. All of them had measurable disease. Among the fifty patients, 24 had previously received chemotherapy. DDP was given at a dose of 50-128 mg/m2 intravenously every 3 weeks. The regimen was PCMF (DDP + CTX + MTX + 5FU), PA (DDP+ADM) +/- other agents or PF (DDP + 5FU) +/- other agents. The overall response rate was 76% (38/50) with a CR rate of 20% (10/50). The median response duration and median survival time were 5.5 months and 15 months, respectively. Best response was obtained in soft tissue metastasis (83.3%, 40/48), and in lung metastasis (79.3%, 23/29). The response rate was lower in liver metastasis (40%, 2/5) and lowest in bone metastasis (0/9). Marked gastrointestinal reaction was observed in most patients (84%). These results indicate that high dose cisplatin combination chemotherapy is an effective regimen for advanced breast cancer.

[Effect of bone morphogenetic protein: experimental study of the implants-bone interface integration].

We investigated the effect of bone morphogenetic protein (BMP) on the implants--bone integration by light microscope, ABC technique, and EDXA. The results showed that there was difference between the BMP of foreign origin and internal origin in the time of inducing new bone formation and in the starting mechanism of new bone formation. The BMP composite implants could accelerate the osseointegration between implants and bone, and would be of value in clinical application.

[External pituitary irradiation therapy for acromegaly].

128 acromegaly patients were followed up for 1-23 years (mean 5.9 +/- 5.0 years) after external pituitary irradiation; among them 104 patients received fractional irradiation with an ultimate total dose of 4000-5000 rad. The remaining patients received a total dose of either < 4000 rad or > 5000 rad. Human growth hormone (hGH) level was measured after an oral load of 100g glucose to evaluate the effect of irradiation. After external pituitary irradiation, the serum level of hGH decreased progressively. The decrease was more rapid in the first three years with a decrement of 35.1% per year. 36.2% of all the patients has been judged as clinically inactive with low serum hGH level of < or = 5 ng/ml. 62.1% of the patients was considered as slightly active with hGH level < or = 10 ng/ml in the 15th year after therapy. The effect of irradiation was not influenced by the age of the patients, the course of acromegaly, the size of sella turcica and operation prior to irradiation. If the total dose of irradiation is lower than 4000 rad, the effect will not be good.

[The dynamics of antigen and antibody in Schistosoma japonicum egg granuloma and its relationship with the granuloma response].

Synchronous Schistosoma japonicum egg granuloma in the lung of mouse model and PAP (peroxidas-anti-peroxidase) technique were employed to study the dynamics of antigen and antibody in the egg granuloma of S. japouicum and its relationship with the granuloma formation. The granulomatous response began on the 3.5 day after egg injection and increased to the maximal size at the 4th week. Lymphocyte populations and macrophage comprised an important part of lesions during the time of acute granulomatous response (7-28 day). Using PAP staining, the SEA within egg could be detected at high level on the first day after egg injection and then declined gradually. On the contrary, the SEA around egg was minimal at the first week and increased to the peak at the 4th week, then decreased gradually. No antibody could be detected throughout the experimental period (35 days). The results suggested that 1) the antigen of Schistosoma egg is the essential factor for the granuloma formation. 2) S. japonicum egg granuloma could be formed in unsensitized mouse. 3) The mechanism of egg granuloma formation of S. japonicum is similar to that of S. mansoni.

[Randomized trial of combined chemotherapy including high dose cisplatin and radiotherapy for esophageal cancer].

A prospective randomized trial in 64 esophageal cancer patients was conducted from 1986 to 1989. The patients were randomized into two groups-PPF (DDP 100 mg/m2 + pingyangmycin + 5-FU) regimen plus radiotherapy (combined group) or radiotherapy alone (control group). The overall response rate and CR rate in the combined group were 87.5% (28/32) and 40.6% (13/32). Those of the control group were 81.3% (26/32) and 21.9% (7/32), respectively. The overall response rates were similar in these two groups, but the CR rate in the combined groups was higher than that of the control group. The 1-year survival rates in the combined and control groups were 77.4% (24/31) and 45.2% (14/31) (p less than 0.01). The 2-year survival rates were 56.3% (9/16) and 34.6% (9/26), respectively (p greater than 0.05). Our preliminary results show that combined chemotherapy including high dose cisplatin and radiotherapy may improve the therapeutic result of esophageal cancer.

[Serological cross reaction between schistosomiasis japonica and pagumogonimiasis skrjabini].

The sera from 25 patients with acute, 36 patients with chronic schistosomiasis japonica and 68 patients with pagumogonimiasis skrjabini were tested against SEA, schistosome egg, Paragonimus adult antigen (PAA) and Paragonimus metacercaria using different tests including biotin-avidin system (BAS), ELISA, circumoval precipitate reaction (COPT) and metacercaria membrane reaction (MCMR). Results showed: 1. The specific reactions using PAS and ELISA were (1) both 100% in acute schistosomiasis patients (ASP), (2) 100% and 97.2% respectively in chronic schistosomiasis patients (CSP) and (3) both were 98.5% in paragonimiasis patients (PP). 2. The cross reactions using BAS, ELISA and MCMR were (1) 76%, 72% and 20% respectively in ASP, (2) 27.8%, 22.2% and 19.4% respectively in CSP. 3. The cross reactions using BAS, ELISA and COPT were 26.5%, 30.9% and 8.8% respectively in PP. 4. The frequency of cross reaction was related to the sensitivity of the test used. 5. The frequency of cross reaction in ASP was remarkably lower with MCMR than with BAS or ELISA. 6. The frequency of cross reaction in CSP was related to the intensity of infection. 7. We suggest that more than one test should be carried out in patients who showed cross reaction to a single test, then the frequency of cross reaction would decline.
