[Erythrocyte phenotypes in Gabon. I. Hemotypologic data].

Blood group antigens were investigated among 400 gabonese individuals subdivided in 6 ethnic groups. The frequencies were calculated and compared to those reported by MOURANT and al. on geographically close African populations. Our data enter the limits already known for a few antigens of ABO, Lewis, Rhesus, Kell, Duffy, Kidd, MNSs, P and Lutheran systems. In addition, we have observed some rare phenotypes: 12 S--s U--, 1 Lu(a--b--), 2 I negative and 7 Sc: --1.

[Fucosidosis and blood group substances in the urine].

Glycopeptide and oligosaccharide fractions obtained from fucosidosis urine contains more Lea activity (4-8 fold) than control urine. Both fucosidosis fractions also contained Leb and H activities, no A activity in contrast to the salivary and erythrocyte phenotypes (A, Le a+ b-). The amount of Leb activity is lower in both fractions (1) and (2) than that of Leb control children (4-16 fold decrease). Blood group A activity was not detected at any concentration used (less than or equal to 50 fold-concentrated urine) whereas A activity was found in (A, Le a- b+) control urine. On the contrary, both fucosidosis fractions contained H activity, whereas A (Le a- b+) control children fractions had none (less than or equal to 50-fold concentrated urine). H and Leb activity might originate from Lea precursor apart from the "non-secretory" type of the patient.

[K-Lymphocyte cytotoxicity: role of human blood group ABO, rhesus and P alloantibodies].

Using human erythrocytes of known antigenic density, sensitized by ABO and Rhesus (D) alloantibodies, it is shown that K cell cytotoxicity mediated by peripheral blood lymphocytes is directly correlated with the number of IgG molecules specifically bound to the target cell surface. The lytic sensitivity of P1k but not P2 or p erythrocytes coated with anti-Tja (anti-P + P1 + Pk) antibodies, demonstrates that effector K cells are mainly triggered through the IgG ANTI-Pk component of such sera.

The Lewis system: New histocompatibility antigens in renal transplantation.

Lewis antigen types (Le, le) were retrospectively determined in 255 first-kidney-transplant recipients. Actuarial survival of grafts at two years was significantly lower in the le/le recipients than in the Le recipients. This indicates that mismatching of these antigens contributes to rejection of kidney transplants. The effects of mismatching for the Lewis and HLA antigen systems seemed to be additive.

[Role of the Lewis (Le-le) and secretor (Se-se) systems in the rejection of kidney grafts].

ABH and Lewis antigens are secreted in distal convoluted and collector tubes of the kidney. The renal ABH secretion is genetically controlled by the Se-se system, while the Lewis specificities are controlled by the Le-le system. The secretory status of the recipient does not modify the probability of graft survival, while the Lewis phenotype seems to play a major role in rejection. The probability of graft survival at 2 years in le-le homozygous recipients (29%) is much lower than that of Le recipients (58%) p less than 0.01.

Labelling and metabolism of methionine-methyl-11 C.

Carbon 11 which is a 20.4 minutes half life isotope emitting positrons was used for methionine labelling on the methyl group by action of 11C-methyl-iodide on DL or L homocysteine. With the method described, 20 to 30 mCi of 11C-methyl-methionine (Specific activity 50 mCi/muM) may be obtained within 25 minutes. The 20 Mev proton beam used for 11C production was 10 muA. The metabolism of 11C methionine studied on mice shows a high uptake in pancreas and to a smaller extent in brain. On humans sequential images were obtained at the head level which allowed measurement of the uptake of the radio-labelled compound in the brain so as to study the radioactivity curve in different parts of the organ.

Lymphocyte transformation test in uveal pathology.

Comparative studies of skin tests and in vitro lymphocyte transformation tests were found to be fruitful with the following antigens: tuberculin and PPD, Streptococcus, vaccine and protein M of Streptococcus, candidin, herpes antigen, Brucella antigen, antigen from autologous saliva. Three types of response occur: (1) positive skin test with syndromic ocular reaction and positive LTT: the diagnosis of hypersensitivity is certain; (2) positive skin test without syndromic reaction but positive LTT in vitro; less sensitive than the in vivo test but enhancing the value of the latter; the diagnosis is presumptive, and (3) skin test positive or negative and LTT negative: indicates that the in vitro test is less sensitive than the in vivo one. Practical interest of this comparison stems from those cases in whom skin testings is contra-indicated because it is considered to be dangerous (macular choroiditis, Eales' disease). Specific desensitization treatment was carried out on most of the cases and seemed to be effective in several of them.
