Angular distributions for (3,4)(Lambda)H bound states in the (3,4)He(e,e(')K+) reaction.

The (3,4)(Lambda)H and (4)(Lambda)H hypernuclear bound states have been observed for the first time in kaon electroproduction on (3,4)He targets. The production cross sections have been determined at Q(2)=0.35 GeV2 and W=1.91 GeV. For either hypernucleus the nuclear form factor is determined by comparing the angular distribution of the (3,4)He(e,e(')K+)(3,4)(Lambda)H processes to the elementary cross section 1H(e,e K+)Lambda on the free proton, measured during the same experiment.

[HLA class I antigen expression correlates with histological cell type in uveal melanoma].

PURPOSE: The prognosis of uveal melanoma is correlated with its histologic cell type. The epithelioid cell type is associated with a higher metastatic rate than the spindle cell type. The Human Leucocyte Antigen Class I (HLA-I) expression of the melanoma also correlates with the prognosis. In this study, we analyzed HLA-I antigen expression of uveal melanomas to determine whether a relationship exist between antigenic expression and melanoma cell type. METHODS: Formalin-fixed, paraffin-embedded spindle cell type (n = 11) and epithelioid cell type (n = 11) uveal melanomas were immunostained with the HC10 antibody (1:80) for HLA-I antigen expression with appropriate positive and negative controls. Sections were assessed semiquantitatively according to the percentage of stained cells. RESULTS: Among the spindle cell type melanomas, 2 out of 11 (18%) stained with HC10 antibodies. The staining intensity was less than 25% of the cells in these two melanomas. Among the epithelioid cell type melanomas, 9 out of 11 (82%) stained with HC10. The staining intensity was more than 25% of the cells in 5 of these 9 melanomas. CONCLUSIONS: It is unknown why spindle and epithelioid cell type uveal melanomas have different prognoses. Human uveal melanoma cell lines with low HLA-I expression are susceptible to NK cell-mediated lysis in vitro and in murine studies. The prognostically more favorable spindle cell type melanoma expresses less HLA-I than the epithelioid cell type melanoma. These results stress the role of NK cells in the rejection of uveal melanoma.

Subtilisins of Bacillus spp. hydrolyze keratin and allow growth on feathers.

Keratinase is a serine protease produced by Bacillus licheniformis PWD-1 that effectively degrades keratin and confers the ability to grow on feathers to a protease-deficient B. subtilis strain. Studies presented herein demonstrate that B. licheniformis Carlsberg strain NCIMB 6816, which produces the well-characterized serine protease subtilisin Carlsberg, also degrades and grows on feathers. The PWD-1 and Carlsberg strains showed a similar time-course of enzyme production, and the purified serine proteases have similar enzymatic properties on insoluble azokeratin and soluble FITC-casein. Kinetic analysis of both enzymes demonstrated that they have high specificity for aromatic and hydrophobic amino acids in the P1 substrate position, although keratinase discriminates more than subtilisin Carlsberg against charged residues at this site. Nucleotide sequence analysis of the serine protease genes from B. licheniformis strains PWD-1, Carlsberg NCIMB 6816, ATCC 12759, and NCIMB 10689 showed that the kerA-encoded protease of PWD-1 differs from the others only by having V222, rather than A222, near the active site serine S220. Further, high-level expression of subE-encoded subtilisin from B. subtilis (78% similar to subtilisin Carlsberg) also confers growth on feathers on a protease-deficient B. subtilis strain. While strain PWD-1 and the kerA protease efficiently degrade keratin, keratin hydrolysis and growth on feathers is a property that can be conferred by appropriate expression of the major subtilisins, including the industrially produced enzymes.

Sexual and seasonal differences in the vomeronasal epithelium of the red-backed salamander (Plethodon cinereus).

Sexually dimorphic behaviors often are associated with sexually dimorphic neural structures. Perinatal hormonal levels influence structural sexual dimorphism, and seasonal structural changes also can be the result of seasonal hormonal fluctuations. We compared the volume of vomeronasal organs of male and female red-backed salamanders (Plethodon cinereus) throughout the year. Odorants are delivered to vomeronasal receptors through nasolabial grooves when salamanders touch the bases of these grooves to objects (nose tapping). Males may locate and identify potential mates through nose tapping during the prolonged breeding season that lasts from October through May. We compared vomeronasal organ data through multiple regression by using total body size, sex, and season as variables that may influence vomeronasal organ volume. Gonads also were examined as an indicator of reproductive status. Total body size and sex significantly affect vomeronasal organ volume; as body size increases, so does vomeronasal organ volume, and males have significantly larger vomeronasal organs than females at all times of the year. During the summer, both males and females have larger vomeronasal organs than at any other time of the year. Summer also is a period of intense foraging and the initiation of a new cycle of gametogenesis. Previous studies of circulating hormone levels in amphibians have shown that the initial stages of gametogenesis correspond to a period of low estradiol and testosterone levels but high gonadotropin-releasing hormone levels. The functional significance of sexual and seasonal differences in the vomeronasal organs of P. cinereus may relate to the neurogenesis of specialized receptors for courtship and mating.

Surface-modified nanocrystalline ceramics for drug delivery applications.

Drug delivery systems comprised of various types of carriers have long been the object of pharmacological investigation. The search has been stimulated by the belief that carriers will lead to reduced drug toxicity, dosage requirements, enhanced cellular targeting and improved shelf-life. Among the carriers investigated are complex polymeric carbohydrates, synthetic proteins and liposomal structures. For the past four years, we have been experimenting with a radically new class of carriers comprised of surface-modified nanocrystalline ceramics. While the ceramics provide the structural stability of a largely immutable solid, the surface modification creates a glassy molecular stabilization film to which pharmacological agents may be bound non-covalently from an aqueous phase with minimal structural denaturation. As a consequence of maintained structural integrity and owing to concentration effects afforded by the surfaces of the nanocrystalline materials, drug activity following surface immobilization is preserved. We have used successfully surface-modified nanocrystalline ceramics to deliver viral antigens for the purpose of evoking an immune response, oxygenated haemoglobin for cell respiration and insulin for carbohydrate metabolism. The theoretical principles, technical details and experimental results are reviewed. Surface-modified nanocrystalline materials offer an exciting new approach to the well-recognized challenges of drug delivery.

Long-term safety and clinical acceptability of venlafaxine and imipramine in outpatients with major depression.

The antidepressant efficacy and safety of venlafaxine was shown previously in 6-week, placebo-controlled trials. We evaluated the long-term safety and clinical acceptability of venlafaxine and imipramine in a double-blind, parallel-group, comparative study. Two hundred ninety depressed outpatients were treated with venlafaxine, and an additional 91 received imipramine for as long as clinically necessary, up to 1 year. The total daily dose of each drug could vary from 75 to 225 mg. The Clinical Global Impressions Scale and a therapeutic response rate that was based on Clinical Global Impressions Scale-Improvement and incorporated discontinuation information were used to evaluate efficacy. Safety determinations and patient subjective ratings were used to evaluate safety and clinical acceptability. During the study, the adverse events were generally mild to moderate and most subsided with continued treatment; the most frequent were nausea for venlafaxine and dry mouth for imipramine. The anticholinergic side effect burden was significantly higher in the imipramine group than in the venlafaxine group. Venlafaxine was judged significantly more acceptable than imipramine, on the basis of the subjective ratings by patients. Fewer venlafaxine-treated patients than imipramine-treated patients withdrew because of adverse events and unsatisfactory response. There was a consistent trend in the therapeutic response rates in favor of venlafaxine that reached statistical significance at months 2, 6, and 12. In this long-term study, patient acceptability was greater for venlafaxine than for imipramine, suggesting therapeutic advantages for venlafaxine in the long-term treatment of depression. Additional studies with other active comparators are underway to confirm and extend these encouraging results.

High-risk subgroups of patients with non-Q wave myocardial infarction based on direction and severity of ST segment deviation.

To determine the significance of the direction of ST segment deviation on admission of patients who evolved non-Q wave myocardial infarction (MI), 97 patients with initial ST segment depression were compared to 207 patients with initial ST segment elevation. Patients with ST segment depression developed smaller infarcts than those with ST segment elevation (creatine kinase MB isoenzyme 8.2 vs 13.3 gmEq/m2, p less than 0.002), but had a lower left ventricular ejection fraction on admission (44% vs 51%, p less than 0.001), more in-hospital complications, and a higher cumulative 1-year mortality (29% vs 11%, p less than 0.001) that could be accounted for by an excess of adverse baseline characteristics. Although a severity index (combining magnitude and extent of the initial ST segment deviation) was not useful for discriminating prognosis of patients with non-Q wave MI who presented with ST segment depression, it was useful in identifying a subgroup of patients with ST segment elevation with an adverse prognosis. The poor outcome of patients with non-Q wave MI presenting with either ST segment depression or severe ST segment elevation on admission suggests that patients in these subgroups should receive close surveillance and should possibly be considered for aggressive therapy.

Compartments and barriers in the sciatic nerve of the rabbit.

Near constant concentration of various radioactively labelled polar nonelectrolytes were maintained in the blood plasma of renal ligated rabbits. The uptake of [14C]urea, [51Cr]EDTA and [14C]inulin into sciatic nerve was interpreted in terms of a two-compartment model. There was a rapidly equilibrating space representing about 20% of the tissue weight which is almost certainly the interstitial fluid of the epineurium and outer layers of the perineurium. Urea penetrated the blood-nerve barrier at a similar rate to the entry of this compound into brain. Sucrose, chromium-EDTA and sulphate passed into the true interstitial fluid approaching a total volume of distribution of about 36% of the tissue weight at 6-8 h. Since movement of these 3 solutes is severely restricted by the blood-barrier, the blood-nerve barrier in the rabbit must contain channels of greater than 1 nm which are not present in the developed blood-brain barrier. Uptake of [3H]3-O-methylglucose into nerve was examined but the results did not give a conclusive answer to the question of whether there is specific transport of sugars at the blood-nerve barrier. The wet nerve contained (in mmole/kg) sodium 74.3; potassium 42.6 and chloride 48.8. Water was 64.6 ml/100 g. Intracellular electrolyte concentrations were estimated. Approximately 30% of the total sodium content cannot be accounted for as being free in the interstitial fluid at a Gibbs-Donnan distribution with plasma. Analysis of metabolically poisoned nerves which have equilibrated with salt solution in vitro suggests that negatively charged polyelectrolyte molecules retain about 14 mmole of sodium/kg wet normal nerve.

A psychosocial kinship model for family therapy.

Family therapy has traditionally centered on the nuclear family and thus has been typically oriented toward urban white middle-class families. A variety of modifications in therapeutic technique has evolved for work with the many modern families whose structure consists of a functional psychosocial family kinship. The authors have developed a formal theoretical framework and model for family therapy that encompasses the total psychosocial network, i.e., the extended kinships of the nuclear family and the functional kin such as friends, neighbors, and associates.

Electrolytes and water in the brain and cerebrospinal fluid of the foetal sheep and guinea-pig.

1. Samples of cisternal cerebrospinal fluid (c.s.f.) and blood plasma have been obtained nearly simultaneously from foetal sheep of different ages, the foetus having been exteriorized and maintained in a normal state with respect to its blood gases and arterial pH. The brains were removed from these foetuses and also from foetal guinea-pigs after exsanguination.2. A comprehensive study has been made of the concentrations of water, chloride, sodium, potassium, calcium and magnesium in blood plasma, c.s.f. and brain from foetal sheep and in brain from foetal guinea-pigs during development in utero. Maternal arterial blood plasma, cisternal c.s.f. and brain from the sheep and brain from the guinea-pig have been analysed for comparison.3. Concentrations of the ions analysed in foetal blood plasma are similar to those found by others in the sheep and other species. In the case of calcium, the results suggest an active maintenance of the concentration of this ion in foetal plasma by the placenta.4. The chloride concentration in c.s.f. at ages between 65 days and term (147 days) averaged 1.19 times that in foetal plasma, but only 1.08 at 45-50 days; the sodium concentration in c.s.f. was also slightly reduced at this time. The increase in the c.s.f./plasma ratios for chloride and sodium appears to coincide with the first development of the blood-brain and blood-c.s.f. barriers to non-electrolytes.5. Magnesium was at a slightly higher concentration in c.s.f. than in plasma at all foetal ages and did not vary with age. The concentrations of potassium and calcium in c.s.f. were high at early ages and fell to reach adult concentrations after birth: the mechanisms determining the concentrations of the various ions in c.s.f. develop at very different, largely independent, rates.6. The water content of the cerebral hemispheres of the foetal sheep was stable at 90% of the wet weight till 105 days and fell thereafter. The contents of chloride, sodium and potassium followed paraboloid relations with age. Chloride and sodium reached a peak of 62 and 81 m-equiv/kg respectively between 89 and 105 days in the sheep. Potassium was at a minimum of 65 m-equiv/kg at the same time. The content of water and these electrolytes in the cerebral hemispheres of the foetal guinea-pig underwent similar changes, the maxima and minimum occurring as in the sheep at two-thirds of the total length of the pregnancy, namely 65 days in the guinea-pig. At 46 days in the guinea-pig, chloride in brain reached 53 m-equiv/kg, sodium was 68 m-equiv/kg and potassium was 75 m-equiv/kg. In contrast to the sheep, no reversal of the sodium-potassium ratio occurred. These changes in water and electrolytes probably represent a rise, a peak and a decrease in the volume of the extracellular space of cerebral cortex, but changes in the volume occupied by a cell-type, containing much intracellular chloride and sodium, could also contribute to this phenomenon.7. The calcium content in the cerebral hemispheres of the foetal sheep remained at about 3.0 m-equiv/kg throughout pregnancy. Magnesium in the cerebral hemispheres of both the foetal sheep and the guinea-pig showed a trough in concentration during pregnancy which corresponded approximately in position to the minimum in potassium content and the maxima in chloride and sodium contents. Lowest values were 9.6 m-equiv/kg at 105 days in the sheep and 9.5 m-equiv/kg at 51 days in the guinea-pig.

A portable device for group modification of classroom attending behavior.

There is a need for a portable means of signalling appropriate classroom behavior that the teacher can carry with her as she moves about the room. An inexpensive wireless switch, adapted for classroom use, was used to turn on a clock when all children were attending to their work. When a movie was made contingent on accumulating a specific number of minutes on the clock, class-attending increased. Throughout these conditions, the records of class attending made by an independent observer with a stopwatch correlated highly with the remote controlled clock.

When owned and governed by those who implement it, clinical supervision can become a living, breathing part of professional working life, and not just another ritual. A multidisciplinary team from Southampton explains.
