Effect of boar seminal immunosuppressive fraction on B lymphocytes and on primary antibody response.

Repeated i.p. or rectal treatment of male and female mice with an immunosuppressive component isolated from boar seminal vesicle secretion reduced responses of B lymphocytes to mitogen as evaluated by [3H]thymidine or bromo-deoxyuridine incorporation. The proliferative activity of T lymphocytes was not affected. By means of the immunofluorescence method, the seminal immunosuppressive component was detected on the membranes of B lymphocytes separated from the spleens of mice treated in vivo with immunosuppressor. An i.p. injection or rectal infusion of the immunosuppressive component also led to a suppression of primary antibody response to soluble and particulate antigens. These findings indicate that in vivo deposition of semen may compromise some aspects of the immune system and may be an important cofactor in the development of viral and bacterial infections in homosexual men.

[Infectious agents and treatment of infertility with IVF and ET].

An infectious agent is a confirmed etiological factor of female tubal sterility. In conjunction with the recorded decline of fertility of the male population the participation of infection in this trend can be taken into consideration. In the submitted paper the authors are concerned with the detection of an infectious agent in spermatozoa in the sterile population and its role when using assisted reproduction techniques (ART). From the results ensues that cultivation examinations of spermatozoa before a programme of in vitro fertilization and embryo transfer (IVF + ET) has its place in the screening of the treated couple. Detection of virulent types of microorganisms has a negative effect on results of IVF + ET. Groups of men with this finding should be treated with antibiotics with regard to sensitivity and at the same time cultivation examinations of their partners should be made an necessary treatment and provided.

Immunosuppressive effect induced by intraperitoneal and rectal administration of boar seminal immunosuppressive factor.

The immunosuppressive component was isolated from boar seminal vesicle secretion and administered i.p. or rectally to male mice. By means of the immunofluorescent method, the seminal immunosuppressive component was found on the membranes of 50-70% of white blood cells of treated mice the first day after i.p. and the third day after rectal administration. The immunosuppressive component was observed on the membranes of 10-20% of white cells even at the 17th day after treatment. Intraperitoneal or rectal administration of the immunosuppressive component led to a decrease in the white cell concentration in blood of treated mice. These findings indicate that rectal deposition of semen may compromise some aspects of the immune system and may be an important cofactor in the development of viral or bacterial infections among homosexual men.

[The greater celandine (Chelidonium majus L.)--review of present knowledge].

The greater celandine (Chelidonium majus L.) is a species of the Papaveraceae family. The principal secondary metabolites of the plant are isoquinoline alkaloids; more than 30 were found here. The richest organ is the root, the total content of alkaloids in it reaching 2-3%. The dominant alkaloids of the root are chelidonine and coptisine. The aerial parts contain about 0.5-1.5% alkaloids. The principal base for the whole period of vegetation is coptisine, the representation of other alkaloids substantially changing during the development of the plant. Of nonalkaloidal secondary metabolites, esters of caffeic acid have recently been demonstrated in the plant drug. In the past, the drug was used to treat tumours. At present, the extract of alkaloids from the plant is the base of the preparation Ukrain showing immunomodulating activity and being employed in the therapy of different types of carcinomata. Chelidonium majus is also a component of some preparations employed in the diseases of the biliary tract and liver. Biological effect of the principal constituents of the drug are different and often quite antagonistic. As their representation varies in the course of vegetation, the efficacy of the summary preparations changes in dependence on the prevailing substance in the preparation.

Alkaloids

[Pharmacologic control of reperfusion dysrhythmias].

The present study aims to investigate the effects of the lipophilic antioxidant Trolox C (a vitamin E analogue) and stobadine, a scavenger of free oxygen radicals, on reperfusion dysrhythmias. Experiments were performed on isolated perfused rat hearts subjected to global stop-flow ischaemia followed by reperfusion. Trolox C (10(-4 mol.l-1) and stobadine (10(-5) mol.l-1) were infused immediately prior to ischaemia. Trolox C (10(-4) mol.l-1) and stobadine (10(-5) mol.l-1) decreased the incidence and duration of reperfusion-induced dysrhythmias (quantified by the dysrhythmia score) in comparison to the ischaemic-reperfusion damaged hearts. There was an improvement in the recovery of contraction force and left ventricular diastolic pressure in Trolox or stobadine pretreated hearts. No significant changes in coronary flow resistance were observed. The results suggest that both substances protect the myocardium during ischaemic-reperfusion injury probably by affecting the generation and activity of reactive oxygen species.

Effect of intra-rectal administration of boar seminal immunosuppressive fraction on mouse lymphocytes.

The repeated deposition of an immunosuppressive fraction isolated from boar vesicular gland secretion into the rectum of healthy male and female mice reduced responses of lymphocytes to mitogens in vitro. Rectal deposition of the immunosuppressive component also led to a decrease in the activity of plaque-forming cells. These findings indicate that repeated rectal deposition of semen may compromise some aspects of the immune system and may be an important cofactor in the development of viral or bacterial infections among homosexual men.

Reactivities of a monoclonal antibody directed against an immunosuppressive molecule in boar seminal plasma.

Using a monoclonal antibody (LIS-4) to an immunosuppressive factor isolated from boar vesicular gland secretion it was determined that this gland secretes a tissue-specific immunosuppressive molecule that is absorbed onto the acrosome of spermatozoa during ejaculation. Absorption of the immunosuppressive molecule onto murine embryos at the 2-, 4-, 8-cell, morula and blastocyst stages in vitro was evaluated by indirect immunofluorescence. In vivo absorption was detected on the zona pellucida of murine embryos obtained from oviducts injected with the immunosuppressive molecule. Immunofluorescence revealed that the immunosuppressive molecule was not absorbed onto murine embryos after solubilization of the zona pellucida. There was no effect of the antibody to the immunosuppressive molecule on the ability of boar spermatozoa to penetrate the porcine zona pellucida.

[Quantitative ultrastructure of the myocardium of Japanese quail after long-term hypodynamia].

Quantitative ultrastructural myocardial changes of myocardium of the japanese quail (Coturnix coturnix japonica) were studied during an immobilization stress (hypodynamia) lasting 9, 30, 60 and 90 days. Hypodynamic quails were fixed by waistcoats to the feeding place simulating conditions of long lasting space flight which the quails were determined for. The surveyed morphological parametres were: mitochondrial rate, mitochondrial volume in % of cytoplasm, volume of myofibres in % of cytoplasm, volume of single mitochondria, extent of cristolysis. Progressive mitochondrial hydratation and even local oedema were observed during the experiment. It led to a progressive increase of mitochondrial volume and simultaneous decrease of volume of myofilaments as a sign of intracellular atrophy of myofibres. The results correlated well with a decreased mass of myofibres observed in experimental quails from space flight on the orbital station MIR.

[Quantitative changes in the ultrastructure of myocardial cells in Japanese quail during hypergravity, hypodynamia and space flight].

The experimental work aimed at the quantitative ultrastructure of the myocardial cells of the Japanese quail Coturnix coturnix japonica during hypergravitation, hypodynamism and space flight in a Soviet satellite. For the determination of quantitative changes of the myocardial ultrastructure a morphometrical method was used with parameters like the number of mitochondria, average mitochondrial size, relative mitochondrial volume, deficiency of cristae and relative volume of myofibrils. The quails were observed in 3 groups. The absolute control consisted of quails living in normal Earth conditions, in the laboratory group the quails were exposed to conditions of hypergravitation and hypodynamism in a specially constructed centrifuge, and in the flying group the quails were exposed to space flight in a Soviet orbital station MIR. In the group of absolute controls no pathological changes of the myocardial ultrastructure were found. In the flying group there were no significant changes, with the exception of decreased relative volume of myofibrils, which however agrees with the findings on symptoms corresponding to human and animal heart weakness during space flights. In the laboratory group, pathological changes were observed in each of the fractions. The most significant pathological findings were found in the group controls in the center and in hypergravitation combined with hypodynamism. It can be concluded that the laboratories can simulate conditions induced by the start and flight of space ships. (Fig. 2, Ref, 8.)

Lack of effect of boar seminal vesicle fluid immunosuppressor factor on embryo development.

The effects of an immunosuppressive factor (ISF) isolated from boar seminal vesicle fluid on in vitro and in vivo mouse development were investigated. It was found that the zona pellucida of porcine and mouse oocytes pre-incubated with ISF reacted in indirect immunofluorescence with antibodies against ISF. Further results indicated that the boar ISF had no effect on embryo development.

Immunosuppressive factor in cow follicular fluid.

The activity of the immunosuppressive fraction and proteinase inhibitor isolated from cow follicular fluid was investigated. The immunosuppressive factor was separated from the accompanying proteinase inhibitor by Sepharose 6B and Sephacryl S-200 column chromatography. In vitro, the fraction significantly reduced mitogen-induced lymphocyte proliferation. In vivo, the fraction inhibited mouse plaque formation. However, the factor had no effect on the development of intact or zona free embryos. The active components have a molecular mass of about 110,000.

[Practical output of the results of computerized EEG frequency analysis].

For the practical application of EEG frequency analysis it is necessary to ensure an output of results which contains only clinically relevant data. The article describes a new solution where, based on adequate statistical evaluation of parameters of frequency spectra, only important facts are topographically visualized. Marked reduction of data thus makes it possible to concentrate useful information. Contrary to hitheric common "mapping" of electric activity of the brain, it is thus easier to detect pathological changes in the EEG frequency spectra.

[Cardio-pulmo-cerebral resuscitation in myocardial infarct in the emergency medical service].

Of 1234 patients with confirmed myocardial infarction who were treated at the emergency medical service station in Ostrava in 95 (8%) circulatory arrest developed and cardiopulmocerebral resuscitation was applied. In 60 (63%) it was not possible to restore spontaneous circulation and they were subjected to necropsy after resuscitation efforts. A total of 35 (37%) of the resuscitated patients were taken to hospital. Resuscitation was permanently successful in 19 (20%) of patients with myocardial infarction. The authors discuss the most frequent shortcomings of urgent resuscitation as well as other possible ways how to improve the final results.
