The influence of Eleuterococcus senticosus on cellular and humoral immunological response of mice.

The influence of Eleuterococcus senticosus on cellular and humoral immune response was evaluated. The experiments were performed on animal models (Balb/c mice and F1 crossbreeds Balb/cxC3H). It was shown that Eleuterococcus has immunomodulatory properties. This substance enhanced the cellular response of the mouse immunological system (chemokinetic activity of mice spleen cells, GvH reaction). A stimulatory effect of Eleuterococcus on the humoral response (antibody production) was also observed. Eleuterococcus did not augment the angiogenic activity of human renal carcinoma cells.

Development and validation of a HPLC method for the determination of cetirizine in pharmaceutical dosage forms.

A rapid, simple and accurate HPLC method is described for the assay of cetirizine in commercial dosage forms. Methanol was found to be a suitable extraction solvent for tablets and for preparing solutions from drops and oral liquids. The samples were chromatographed on a Nova-Pak C18 column and UV detected at 227 nm. The elution was achieved isocratically with a mobile phase of 0.067 M phosphate buffer pH 3.40/acetonitrile (1:1, v/v). Ketotifen was applied as an internal standard. The method was validated for linearity, precision, accuracy and limit of detection. The recovery (mean +/- SD) for tablets was 100.88% +/- 0.8967, for drops 100.35% +/- 0.4062 and for solutions 101.20% +/- 1.1698.

Electron paramagnetic resonance (EPR) studies on stable and transient radicals in humic acids from compost, soil, peat and brown coal.

Quantitative EPR method was applied to characterise four types of humic acids (HA) derived from composts, soil, peat and soft brown coal. For each sample of HA the level of native (indigenous) radicals was estimated. Interactions of the HA with various gaseous agents and organic solvents were investigated. Strong effects of gaseous ammonia and aliphatic amines on spin concentration enhancement were observed; additionally, higher values of g-value were found to be associated with the formed 'transient' radicals. Correlation of copper(II) ions uptake by different HA with effect diminishing primary spin concentration was established. It was recognised that the radical centres, which are enhanced by 'ammonia effect' are quenched in the formed HA-Cu(II) complexes. The both opposite effects are competitive from each other, where 'copper(II) quenching effect' prevails. Reaction of nitrogen dioxide with the humic acids was also examined. The presence of diketones and/or other compounds with active methylene group results in formation of the iminoxy radicals; these radicals are immobilized in the solid (macromolecular) matrix of the humic acids.

Non-invasive assessment of cardiac physiology by tissue Doppler echocardiography. A comparison with invasive haemodynamics.

BACKGROUND: Tissue Doppler echocardiography reveals characteristic patterns of myocardial velocities within systole and diastole which are not well understood. AIM: The purpose of this study was to determine the relationship of myocardial velocity patterns, as assessed by tissue Doppler echocardiography, to the contraction and relaxation phases of the cardiac cycle, as determined during cardiac catheterization. METHODS: Recordings of left ventricular/aortic and left ventricular/pulmonary wedge pressures were obtained simultaneously with apical tissue Doppler echocardiographic images of the left ventricle. A total of 210 cardiac cycles from 22 patients (mean age 58 years, 18 male) undergoing cardiac catheterization were analysed. The time intervals of the different phases of the cardiac cycle were measured from the pressure tracings. These time intervals were correlated to the interfaces of colour myocardial velocity patterns obtained by M-mode tissue Doppler echocardiography. RESULTS: There was a good correlation between the time intervals assessed haemodynamically and those based on the different velocity interfaces obtained with M-mode tissue Doppler echocardiography. Comparable time intervals (from the R wave) obtained by pressure recordings and tissue Doppler echocardiography were, respectively: isovolumic contraction (70 +/- 14 vs 67 +/- 9 ms, r = 0.79); rapid ejection (206 +/- 54 vs 202 +/- 49 ms; r = 0.95); late ejection (357 +/- 36 vs 346 +/- 42 ms. r = 0.93); isovolumic relaxation (405 +/- 43 vs 409 +/- 56 ms; r = 0.95); rapid filling (514 +/- 67 vs 523 +/- 64 ms, r = 0.91); diastasis (697 +/- 153 vs 709 +/- 146 ms, r = 0.98); atrial contraction (890 +/- 128 vs 899 +/- 132 ms, r = 0.96). CONCLUSION: Tissue Doppler echocardiography has the potential to accurately measure the different phases of the cardiac cycle which until now could only be determined invasively. It may provide a sensitive method for the assessment of changes in both cardiac contraction and relaxation in different clinical settings.

[Pregnancy in women with Marfan's syndrome].

The authors discuss current state of knowledge contained in medical literature, concerning procedures with pregnant women with Marfan syndrome and give proposes directives of procedure in this causes. The most important parameters that distinguish with regard of risk of falling into layers of aorta or increase insufficiency aortic valvae during pregnancy is dimension of aortic bulb. If its is more than 4 cm the pregnancy should be avoided limitation of activity is recommended in treatment pregnant women with Marfan's syndrome. The blood pressure should be kept below 130 mm Hg eventually using beta-receptors blockers is recommended. It aortic bulb is wided to 5.5 cm the possibility of cardiosurgical intervention should be taken into consideration. Systematically evaluation of CTG after 6-8 weeks during pregnancy and to two months after delivery for evaluation the dimension of aorta and function of vulvae. We prefer the spontaneous delivery when the aortic dimension is less than 4 cm with collaboration with anaesthesiologist. It is reasonable to perform cesarean section if the dimension of aortic bulb is more than 4 cm.

Comparison of the absolute calibration method with the method of standard addition for the determination of halothane in blood by gas chromatographic headspace analysis.

As long-term exposure to exhaled halothane can be a cause of hepatitis and/or damage to the liver, the determination of halothane in blood is important in clinical practice. Gas chromatographic headspace analysis appears to be the most successful method. In the present study, two methods of quantitative evaluation of the analysis were compared by statistical treatment and direct comparison. The absolute calibration method was found to be unsuitable since it yields entirely unreliable results. This is due to different contents of lipoid and other components in the blood of the normal population, and this influences the vapour phase concentration of halothane and results in different values for the slopes of calibration curves for different blood samples. The standard addition method gives reliable results.

The influence of lymphocytes on blood proteins in vitro.

Autologous IgG, IgM and IgA globulins were assayed quantitatively by the immunodiffusion method in cultures of white blood cells from 14 patients with chronic lymphocytic leukemia. In cases in which levels of IgG in the patient's blood were lower than 1,000 mg/100 ml, the IgG globulin was statistically significantly deficient in the cultures. Moreover, concentrations of IgG in the culture supernatant and absolute lymphocytosis were moderately strongly correlated. These observations indicate that the white blood cells of leukemia patients combine with IgG in their environment, and that this may be one of the mechanisms leading to hypogammaglobulinemia.
