Enzymatically stable renin inhibitors containing statine and 6 aminohexanoic acid. Part IV.

Eight new peptide renin inhibitors: Boc-Phe/4-OMe/His-Sta-epsilonAhx-Iaa(13), Boc-Phe/4-OMe/-His-Sta-episilonAhx-OMe,(21),Boc-Phe/4-OMe/-MePhe-S ta-epsilonAhx-Iaa(27),Boc-Phe/4-OMe/-MePhe-Sta-epsilonAhx-++ +epsilonAhx-Iaa(32),Boc-Phe/4-OMe/-MePhe-Sta-Val-epsilonAhx- OMe (38),Boc-Phe/4-OMe/-MeVal-Sta-Val-Iaa(48),Boc-Phe/4-OMe/-Me Val-Sta-Iaa(51), Boc-Phe/4-OMe/-MeLeu-Sta-epsilonAhx-Iaa (57) have been synthesized in search after compounds of improved biological properties. All peptides were obtained by carbodimide method in solution by stepwise elongation of the peptide chain or by fragment condensation. Their potency was assayed in vitro by a spectrofluorometric method/assay of Leu-Val-Tyr-Ser released from N-acetyltetradecapeptide substrate by renin in the presence of an inhibitor/. Their resistance to enzymatic degradation was assayed by determination of stability to chymotrypsin activity. The most potent inhibitor was (13):IC50 = 7 x 10(-8)M/1. All inhibitors were stable to chymotrypsin.

[Comparative studies of 2 systems of early cyto-oncologic diagnosis of cervix erosion].

In the years 1979-1980 screening examinations (by the open method) were given to 51232 women, that is 85.3% of the female population of Bytom at risk for cervical cancer. Grade I and II cytological smears were found in 46320 (98.52%) of women. Papanicolau grade III was found in 201 (1.12%), and grade IV-V in 64 (0.12%). At the same time, in the years 1978-1984 in the closed health service institutions 19968 women were examined, and suspected smears (grade III-IV) were noted in 368 cases (1.8%). Histological verification showed cervical dysplasia in 184 (0.36%) and 106 (0.53%) respectively, and cervical carcinoma grade 0-IV 52 (10%) and 156 (0.94%). The author compared the results with similar ones obtained in various regions of Poland and in foreign countries. The author failed to find greater detectability of cervical dysplasia and carcinoma in Silesia in comparison with certain, less industrialized, regions of Poland. The author thinks that the stationary system of cervical cancer prevention has a greater chance of being introduced on condition of reorganization of this system, introduction of training of cytological technicians and organization of centres of cytooncological examinations.

[Measurement of angiotensin I convertase activity in the serum of the rat].

A fluorimetric method has been developed for the determination of activity of angiotensin I convertase in rat blood serum. The method proved highly sensitive. It enables determination of the activity of angiotensin I converting enzyme in 0.01 cm3 of blood serum. The method may be applied to in vitro and in vivo testing of inhibitors of angiotensin I convertase.

[Growth hormone, insulin and placental lactogen in the sera of mothers and fetal blood of infants from physiological pregnancy and pregnancy complicated by diabetes mellitus].

The study was aimed at investigating the changes in the blood serum concentrations of growth hormone (HGH), insulin and placental lactogen (HPL) in the newborns of diabetic mothers as compared to those born from physiological pregnancies. The concentration of hormones was determined by RIA both in the newborns and in mothers of the two groups. The mean insulin concentration was significantly higher, and that of HGH significantly lower in the newborns of diabetic mothers, as compared to those of healthy mothers. It seems that the occurrence of fetal macrosomy in diabetic pregnancies is associated with high insulin levels.

[Toxicity of volatile thermal degradation products of conveyer belts studied in guinea pigs].

The toxicity to guinea pigs of the thermal degradation products of conveyor belts, preliminarily filtered through mining gas-mask container, has been tested. The smoke from rubber belts was more toxic than that from polyvinyl chloride. Addition of coal during burning decreased the toxicity of smoke. The test products caused an increase in serum aspartate and alanine aminotransferase, and pulmonary emphysema and edema. In the animals that survived the early period post intoxication, bronchopneumonia as well as pneumonia of parenchymatous type and small degeneration lesions in the liver and kidney have been found.

[Further studies of the toxicity of volatile thermal degradation products of conveyor belts].

Toxic effects of the thermal degradation products of polivinyl chloride and rubber conveyor belts, preliminarily filtered through modified mining gas-mask container were studied in guinea pigs. The smoke concentration was approximate to the one found in mines near the fire during minimal ventilation (0.15 m/sec.) Within 60 min. exposure the test products showed slight irritative effects and caused a moderate lung injury and small degenerative lesions in the liver and kidney. All these changes were rather reversible and did not involve any risk to the animals' lives. The modified container reduced the toxicity of the test substances much more than did the hitherto used mining gas-mask container.

[Toxic effect of volatile products of thermooxidizing decomposition of conveyor belts].

Toxic effects of thermo-oxidizing decomposition products of conveying belts made of PVC and rubber have been tested. The resultant smokes were passed through a protective absorber. During 14-days' post-exposure observation, an increased activity of asparagine, aminotransferase and in some animal alanine aminotransferase was found. Moderately increased pathomorphological changes were found in lungs (emphysema, oedema interstitial and bronchogenic inflammations), along with slight degenerative changes in the liver and kidneys. In the peripheral blood, increased amounts of erythrocytes and leukocytes were found.

[Preventive gynecological examinations of women employed in the cement industry].

Prophylactic gynecologic examinations were carried out in 218 women-workers of a cement plant. The most frequent diseases were: retroflexion of the uterus (55.5%), inflammation of the ovary (50.45%), erosion of the vaginal part of the uterus (41.8%), prolapse of the vaginal walls (39.45%), incontinence (35.32%) and Trichomonas vaginalis (18.7%). The extensive prevalence of the diseases imposes a need to carry on preventive examinations of women employed in cement industry. The authors indicate that certain diseases are obviously due to deficient hygiene.
