Reversal of alpha 1-receptor mediated relaxation in intestinal smooth muscle.

The alpha 1-receptor agonist phenylephrine relaxed longitudinal rabbit jejunal muscle contracted in vitro by low concentrations of barium ions (1 mM). When the Ba2+ concentration was increased to 10-15 mM the response to phenylephrine was a contraction, and at Ba2+ concentrations in between the high and low range this response was biphasic--a relaxation followed by a contractile phase. The alpha 2-receptor agonist clonidine did not affect the tone of the Ba2+ contracted preparation. When the muscle preparation was contracted by Sr2+ (1-20 mM) in the presence of Ca2+ (2.5 mM), phenylephrine relaxed it, and no contractile response to phenylephrine was observed. In the absence of extracellular Ca2+, 5 mM Ba2+ caused a contraction. Under these conditions phenylephrine had no effect on the tissue tone. When Ca2+ was added in a low concentration (0.2-2 mM), phenylephrine elicited a gradually increasing contractile response. At 5 mM Ca2+ the contractile response was replaced by the normal relaxation. The contractile response to phenylephrine in the presence of 5 mM Ba2+ and 2.5 mM Ca2+ was partially blocked by low concentrations of verapamil. In higher concentrations verapamil abolished the tissue tonus completely. The contractile response to phenylephrine in the presence of 5 mM Ba2+ and 2.5 mM Ca2+ could be reverted to the normal relaxation by the addition of 20 mM Mg2+. Increasing the K+ concentration from the normal 5.9 to 62.9 mM blocked the phenylephrine-induced relaxation. No contractile response to phenylephrine occurred. It is concluded that Ba2+ could reverse the response of alpha 1 receptor stimulation in rabbit jejunum from a relaxation to a contraction and that this contractile response was dependent on the presence of Ca2+.

Effect of cGMP derivatives on contraction relaxation cycle, release of norepinephrine and protein kinase activity in guinea pig vas deferens.

Contractions of the guinea pigs vas deferens were induced by field stimulation and the effects of derivatives of cGMP were tested on the contraction-relaxation cycle. Relaxation was induced by 8-Br-cGMP and O2'-mb-cGMP. On the contrary, db-cGMP and N2-mb-cGMP potentiated the contraction. The relaxant effect of 8-Br-cGMP was not mimicked by 8-Br-5'GMP, instead the latter caused the opposite effect. Since neither 8-Br-cGMP nor db-cGMP affected the release of norepinephrine on field stimulation the cGMP derivatives induced their effects at a postsynaptic site. 8-Br-cGMP was found to be 100-fold more potent than cGMP to simulate protein kinase activity in crude extracts from guinea pig was deferens. N2-mb-cGMP and O2'-mb-cGMP were approximately equipotent to cGMP while db-cGMP was less active. 8-Br-5'-GMP did not stimulate the protein kinase. It is suggested that the results support the hypothesis that cGMP acts as a relaxant in the smooth muscle of the vas deferens. The potentiation caused by db-cGMP and N2-mb-cGMP on contractions could be dependent on the substitution in the guanine moiety of the molecules with the butyryl group. Their effect was thus probably unrelated to a specific cGMP effect.

A simple and efficient method for studying neurotransmitter release in vitro by a radiotracer technique.

A method which allows measurement of neurotransmitter release from isolated preparations has been developed. With this method release of 3H-acetylcholine from guinea pig ileum and rabbit jejunum could be studied after preincubation of the preparations with 3H-choline. The release of noradrenaline from guinea pig vas deferens could also be measured after preincubation with 3H-noradrenaline. The method is based on a device which allows experiments on 4 preparations at the same time, including incubation, field stimulation and collection of fractions for counting of radioactivity. With the purpose of obtaining a simple and inexpensive device for stimulating the tissue preparations electrically, a physiological stimulator distributor (PSD) was constructed. It is concluded that the described method constitutes an inexpensive simple and quick means of studying neurotransmitter release from isolated tissues.

Comparison of beta 1- and beta 2-receptor stimulation in oestrogen or progesterone dominated rat uterus.

Electrical stimulation of rat uterine preparations induced constant and reproducible contractions. The inhibitory effect of some adrenergic substances was tested on oestrogen- or progesterone-primed electrically stimulated rat uteri. The rank order of the relative potencies of some adrenergic agonists was shifted from Iso greater than E greater than Salb greater than Ne greater than H 133/22 greater than Phe greater than Clo in the oestrogen primed uterus to Iso greater than Salb greater than E greater than Ne greater than Phe greater than Clo greater than H 133/22 in progesterone primed uterus. The selective beta 2- agonist salbutamol was relatively more potent in progesterone treated rats than in oestrogen treated, while the beta 1-agonist H 133/22 was more potent in oestrogen dominated uterus. We therefore suggested that beta 2-receptors is dominating in the progesterone primed uterus, while both beta 1 and beta 2-receptors could be found in oestrogen primed uterus. No dissociation between the inhibitory effect on contractility and the cAMP increasing effect of isoprenaline could be demonstrated in rat uteri.

Synthesis of 3H-acetylcholine in the rabbit lacrimal gland and its release by electrical field stimulation.

Electrical field stimulation was applied to the rabbit lacrimal gland during incubation in vitro with 3H-choline, after which the efflux of tritiated metabolites was studied. In the presence of physostigmine 3H-acetylcholine was released into the incubation medium when the gland was stimulated electrically. The identity of the 3H-acetylcholine synthesized by the preparation was established by its identical ion exchange chromatographic properties to native acetylcholine. The tissue contents of 3H-acetylcholine and 3H-choline were 14.4 and 3.6%, respectively, of the total activities found in the gland. This study demonstrates that the rabbit lacrimal gland is innervated cholinergically. It is suggested that the method developed might be of value for studying effects of drugs on cholinergic neurotransmission in this organ.

A promoting action of cyclic GMP on contractions of guinea-pig vas deferens.

The effect of dbc-GMP has been studied in ductus deferens of the guinea-pig. The nucleotide potentiated the contractions induced by electrical field stimulation and by adrenergic agonists. The site of action was probably in the smooth muscle cell, since the release of excitatory transmittor was not influenced.
