Nucleotide sequence of a human immunoglobulin C gamma 4 gene.

We report the nucleotide sequence of a gene encoding the constant region of a human immunoglobulin gamma 4 heavy chain (C gamma 4). These data represent the first complete sequence determination of a human CH gene. As expected from structural studies of mouse C gamma genes, the coding sequences for the CH domains and hinge segment are separated from one another by intervening DNA sequences. Comparison with genomic sequences of the mouse C gamma 1, C gamma 2a, and C gamma 2b genes shows conservation of the sequences in the constant region domains and the 3' untranslated region surrounding the presumed site of poly(A) addition.

Fluorescent staining of fungal nuclei with a benzimidazol derivative.

A direct staining procedure is described for fluorescence microscopy of fungal nuclei, chromosomes and mitochondria. The fluorochrome is a benzimidazol derivative (33258 Hoechst) known to bind selectively to deoxyribonucleic acid at neutral pH. The advantages of 33258 Hoechst relative to Feulgen compounds used previously to stain these structures include a greater intensity of fluorescence, the absence of fading or rapid quenching of the fluorescence, and the omission of acid hydrolysis from the procedure for removal of ribonucleic acid. 33258 Hoechst has been evaluated as a nuclear stain with a yeast (Saccharomyces cerevisiae) and a filamentous fungus (Agaricus bisporus) and appears to penetrate easily vegetative cells and spores of both fungi.

Quantitative histochemical evaluation of normal human skeletal muscle.

Skeletal muscle tissue was obtained by open biopsy from the vastus lateralis and peroneus brevis muscles from 12 and 16 healthy paid volunteers, respectively. Frozen sections were examined with standard histochemical methods. Central nuclei, small and large angular fibers, and small round fibers were the most common "abnormalities" present. The number of fibers of these types were quantified, along with other more rare deviations from normal morphology. Several of the abnormalities were more common in the peroneus brevis than in the vastus lateralis.

Oral contraceptives and increased formation of soluble fibrin.

Soluble fibrin was measured weekly for two months in 12 normal women and in 12 women on combined oestrogen-progestogen therapy (Ortho-Novin, Norinyl, Enavid, Ovral, Ovulen, Demulen). Plasma soluble fibrin concentration in women on oral contraceptives showed an increase of 97.2% (P <0.001) above that of normal women. In three cases, where each woman on the oral contraceptive served as her own control, stopping medication led to a return to normal of the plasma content of soluble fibrin.

"Sex ratio" in Drosophila pseudoobscura: spermiogenic failure.

Contrary to earlier reports, testes of "sex ratio" Drosophila pseudoobscura males have only half as many sperm per bundle as testes of normal males do. This fact, determined from electron micrographs, indicates that the functional pole hypothesis is not applicable to the action of "sex ratio."

A neuropsychological perspective on dual diagnosis.

The purpose of this study was to investigate whether patients experiencing both a major psychiatric disorder and a concurrent substance abuse problem (dual diagnosis) manifest greater neuropsychological dysfunctions than patients experiencing a major psychiatric disorder alone. Differences in diagnostic variability and fluidity between dual diagnosis and non-dual diagnosis patients and the occurrence of polysubstance abuse among the dual diagnosis patients were also investigated. The hypotheses were tested by retrospectively reviewing the psychiatric records of 50 dual diagnosis patients and comparing them to 36 chronically hospitalized non-dual diagnosis psychiatric patients. Results revealed that dual diagnosis patients manifested significantly greater neuropsychological impairments than non-dual diagnosis patients and that dual diagnosis patients showed a greater range of diagnoses and diagnostic fluidity than non-dual diagnosis patients. Results also revealed that over one-half of the dual diagnosis patients abused a variety of different substances. An etiological role of neuropsychological dysfunction in the development of the dual diagnosis syndrome in at least a subgroup of dual diagnosis patients is suggested. More specifically, it is suggested that the neuropsychological dysfunctional pattern that is described may represent an organic substrate of a nonspecific vulnerability to developing both (atypical) psychoses and (poly) substance abuse.

Assessment of early childhood caries and dietary habits in a population of migrant Hispanic children in Stockton, California.

OBJECTIVE: This study estimated the prevalence of early childhood caries (ECC) and related behavioral risk factors in a population of low-income, Mexican-American children in Stockton, California. METHODS: We collected data for 220 children ages six years or less using a parent-completed questionnaire and clinical dental examinations during the Su Salud Health and Education Fair in July 1995. We employed five case definitions of ECC: buccal or lingual caries on one or two primary maxillary incisors; caries on any surface of one or two primary maxillary incisors; and five decayed, missing (due to caries), or filled primary teeth. RESULTS: The prevalence of ECC ranged from 12.3 percent to 30.5 percent, depending upon the case definition. More than 17 percent of children age two years had one primary maxillary incisor affected by caries on the buccal or lingual surface; 13.2 percent had two affected. Mean age at weaning from breast--or bottle-feeding and patterns of bottle use during sleep did not differ significantly between children with ECC and those without. There were no clear patterns of cariogenic food frequency and disease status. CONCLUSIONS: Our findings question whether feeding patterns with human breast milk, formula, or bovine milk are sufficient etiologic factors for this condition.

Fibrous dysplasia associated with an idiopathic bone cavity.

A patient with fibrous dysplasia of the maxilla had an associated idiopathic bone cavity (traumatic bone cyst). The idiopathic bone cavity was localized over the roots of the right maxillary second molar and mimicked periapical pathosis. The patient was inappropriately treated with root canal therapy.
