Effect of protein hydrolysis on the dialysability of amino acids and minerals in infant formulas.

The objectives of this study were to evaluate the dialysed quantities of amino acids and minerals (calcium, phosphorus, magnesium, iron and zinc) in two types of infant formulas, one with a basis of native cow milk proteins and the other highly hydrolysed, to evaluate the possible application of Miller's in vitro method in amino acid and mineral availability studies. The percentage of dialysis differed between proteins. The hydrolysis treatment applied to the proteins significantly increased the dialysis percentage of almost all the amino acids. The dialysability of all the minerals was statistically greater in the formula made with a basis of hydrolysed proteins. Miller's method was seen to be of use for showing the effect which the hydrolysis treatment has on the availability of amino acids. Despite its limitations of having to be carried out in vitro, the study suggests that the nutritional value in relation with amino acids, minerals and trace elements differs between the both types of formulas.

Influence of dietary protein type and iron source on the absorption of amino acids and minerals.

The apparent digestibility coefficient (ADC) of amino acids and the balance of minerals (calcium, phosphorus, magnesium and iron) has been determined in rats fed four diets differing in the protein type (casein or soy protein) and iron source (ferrous sulphate or lactate) in order to study the possible interactions of these nutrients. The availability of amino acids, especially essential amino acids, was greater in the diet made with animal protein (casein). The iron source also affected the absorption of most amino acids in all the diets assayed with ferrous sulphate being greater. The balance of iron, magnesium and phosphorus was higher in the diets containing animal protein. The retention of calcium and magnesium was significantly greater when ferrous sulphate was used as iron source. These results demonstrate the important interaction between amino acids and minerals and between the minerals themselves, which must be carefully studied when selecting different types of protein or mineral sources in human or animal nutrition.

Influence of exercise on plasma and muscle free amino acids in trained rainbow trout.

Variations in the concentration of free amino acids in the muscle and plasma of trout submitted to 5 minutes of intense exercise have been studied. The responses of untrained fish and those trained performing the same type of exercise twice daily for 28 days are compared. Total amino acid concentrations in muscle tend to diminish after intense exercise. Significant decreases are observed in muscle content of alanine, beta-alanine, isoleucine and ornithine. Plasma amino acids tend to increase after exercise with significant differences in glutamate, GABA, methionine and NH4+. The small variations due to intense exercise suggest that the amino acids are mobilised. Training led to a decrease in total amino acid concentration in plasma but not in muscle, where levels of aspartate and ornithine increased. This suggests a metabolic adaptation to exercise, with amino acid level retention in the muscle.

Perfusion of trout liver in situ. Description and validation of the technique.

The aim of this study was to develop a method for perfusing isolated trout livers that would make it possible to study hepatic metabolism in the whole organ. Rainbow trout (Oncorhynchus mykiss Walbaum) subjected to different fasting periods of 24, 48 or 96 h were used in all the experiments. A non-recirculating system was applied at a flow rate of 0.2 mL/min. The perfusion medium was a specific saline solution for salmonids oxygenated by a multi-bulb glass oxygenator. Viability assays included the measurement of oxygen consumption, lactate dehydrogenase activity and liver metabolic capacity. In addition, a histological study was carried out. Our results showed that the metabolic capacity of the liver survived throughout the perfusion process and that the functioning of this organ changed depending on the length of the fasting period to which the animal had been submitted. The method described here was shown to be suitable for studying the intermediate metabolism of fish.

Identification by spoligotyping of a caprine genotype in Mycobacterium bovis strains causing human tuberculosis.

We have used spoligotyping to characterize 18 Mycobacterium bovis strains isolated from cattle and 23 M. bovis strains isolated from goats. The spoligotypes revealed that caprine strains form a separate and well-differentiated group that we refer to hereafter in this abstract as the caprine genotype. To evaluate the importance of this genotype as a cause of tuberculosis in other animal species, including humans, we applied the spoligotyping method to 112 strains, including to all isolates identified as M. bovis by a Mycobacterial National Reference Laboratory (Majadahonda, Madrid) from 1994 to 1996. Eighty-three of these strains were identified in human isolates. In addition to being identified in three goat isolates and two sheep isolates, the caprine genotype was also found in three isolates causing human tuberculosis. Evidence to support the argument that there is a zoonotic risk of caprine tuberculosis was presented by the identification of the caprine genotype in an isolate from a veterinary worker with a recent history of contact with tuberculous goats.

[Treatment of solitary toxic thyroid nodule with 131I: results in 43 patients].

BACKGROUND: Evaluation of the therapy with high-dose 131I in solitary toxic thyroid adenoma, with particular attention to the effects on thyroid function and on the nodular size. METHODS: A retrospective study of 43 patients with solitary thyroid nodule treated with radioactive iodine (mean dose 26.6 mCi, range 10-35) and followed up for 49.7 +/- 36.6 months (range 6-186) with periodical clinical, laboratory and echographic evaluations. RESULTS: Thirty-two patients (74.4%) had a normal thyroid function during follow-up. Five (11.6%) remained with hyperthyroidism and in 6 (13.9%) hypothyroidism developed 6-30 months after the administration of radioactive iodine. Three of these 6 had subclinical hypothyroidism, with mild increases in serum thyrotropin (TSH). Neither the development of hypothyroidism nor its persistence were significantly correlated with the initial thyroxin (T4) or triiodothyronine (T3) levels, the nodular size, the 131I dose, the incomplete inhibition of the extranodular thyroid parenchyma or the previous therapy with antithyroid drugs. The nodule diminished in size in 15 cases (38.4%), it disappeared in 9 (23%), it remained unchanged in 12 (30.7%) and it increased in 3 (7.7%). CONCLUSIONS: The treatment of the solitary toxic thyroid nodule with relatively high 131I doses is a safe and effective procedure, with a prevalence of residual hypothyroidism which is lower than previously reported. The disappearance of the nodule was only achieved in a minority of cases.

The CHARGE association and athyreosis.

We report on a male infant with congenital hypothyroidism owing to athyreosis occurring with the CHARGE association (bilateral papillary coloboma, congenital heart disease, dysmorphic ears, sensorineural deafness, psychomotor retardation, cryptorchidism, facial palsy, and vesicoureteral reflux). The coexistence of these two disorders has not been described previously.

Postnatal development of the interstitial cells (palisade cells) of the pars intermedia in the cat pituitary gland. An immunocytochemical and ultrastructural study.

The postnatal development of the interstitial agranulated cells (so-called palisade cells) of the pars intermedia in the cat was investigated immunocytochemically and at the ultrastructural level. Since the first postnatal days, a strong vimentin immunoreactivity and a weaker S-100 protein immunoreactivity were detected in the marginal cells lining the pituitary cleft and in the interstitial bipolar cells located within the pars intermedia. No glial fibrillary acidic protein cells have been found in the pars intermedia of any of the animals studied. This immunocytochemical pattern was maintained throughout the postnatal development. Ultrastructurally these cells showed a vast number of cytoplasmic filaments and well-developed junctional complexes. Secretory granules were never seen. In older animals they lined microcavities and microchannels where they project microvilli and present pinocytotic vesicles on their apical surface. No transitional forms between these cells and granulated secretory cells were found. There is a large number of axons and synaptic endings in contact with the granulated secretory cells. From our findings we guess that palisade cells are not a glial derivative, but they may share a common origin with secretory granulated cells.

[Latex allergy].

OBJECTIVE: To review the etiology, diagnosis, prevention and treatment of latex allergy in the hospital setting, and to establish a list of potentially hazardous drugs. METHOD: A literature search was performed. A guide about latex-containing drugs (any component) was developed by contacting manufacturers. RESULTS: The most relevant factor for latex sensitization is exposure extent. Regarding diagnosis the medical history and skin testing are crucial; regarding secondary prevention, identifying the various at-risk groups. Immunotherapy is currently considered a valid option for the management of this condition. CONCLUSIONS: The development of a guide listing latex-containing drugs is essential for the primary prevention of allergic reactions to this substance in hospital.

[In vitro availability of minerals in infant foods with different protein source].

As the result of the digestion process, it is produced at gastrointestinal level interactions between proteins-minerals and minerals-minerals that might modify the bioavailability of the nutrients initially designed for an adequate nutrition in infant formulas. The aim of the present study is to compare the in vitro availability of some minerals and trace elements (calcium, phosphorus, magnesium, iron and zinc) in infant formulas of initiation elaborated with different protein sources: formulas based on cow milk protein (whey-casein) versus vegetal protein (soy-based infant formulas). Also, for evaluating the effects of the different mineral supplementation in the availability of minerals, it was used infant formulas from two different manufacturers. Milk-protein based infant formulas showed for both manufacturers higher dialysis percentage (%) of phosphorus and zinc than the soy-protein based formulas. The availability of iron in the soy formula of the manufacturer A lowered significantly (P < 0.05) respect to the whey-casein based formula (9.6 +/- 2.3 versus 4.6 +/- 0.8), but not respect to the whey-casein formula of manufacturer B (9.6 +/- 1.1 versus 9.0 +/- 0.7), which might be due to the lowest proportion of phytic acid in this last commercial formula. Dialysability of all the minerals analysed from soy-protein based formulas showed significant differences depending on the manufacturer. The purification processes of the soy protein have a high repercussion in the mineral availability of soy-based infant formulas. It could be more interesting to use soy proteins more purified, with low level of phytic acid, in the elaboration of soy infants formulas, than the supplementation them with high amounts of minerals.

[Characteristics of fats of some foods of the meats group and their relations with health].

The fatty acids and cholesterol contents of ten fresh protein foods were determined, 6 meats (pork, rabbit, lamb, turkey, chicken and beef) and 4 fishes (tuna, sea bream, hake and sardine); the fat quality by different chemical index of the foods (saturated fat/polyunsaturated fat ratio, index of atherogenicity and index of thrombogenicity) and the effect of their intakes on the health were also determined. The study shows that the fat content was lower in meat pork than lamb, rabbit, tuna, sea bream and sardine; and it was similar to chicken beef and hake. Pork, chicken and turkey meat had the lowest content of myristic acid. Fishes showed lower index of thrombogenicity than the other analyzed food and pork, chicken and turkey showed the lowest atherogenicity index.
