t(8;21;8)(p23;q22;q22): a new variant form of t(8;21) translocation in acute myeloblastic leukemia with maturation.

The complex variants of t(8;21) involving chromosomes 8 and 21 as well as a variable chromosome account for 1.1-5% of acute myeloid leukemia (AML) patients. This paper reports a case of AML-M2 with t(8;21;8) translocation for the first time. The patient was a female, aged 47 years. Her myelogram was compatible with AML-M2. Chromosome study using R-banding technique revealed a karyotype 46, XX, t(8;8)(p23;q22). Dual-color FISH assay with two probes P1 164(green signal) and YAC 225B8 (red signal) both of which closely located on the 8q showed that one yellow signal consisting of a green signal and a red signal and one red signal appeared on the long and the short arm of the same der(8) chromosome, respectively, further confirming this translocation occurred between both homologous chromosomes 8. RT-PCR analysis detected the AML1/ETO fusion transcript in our patient, thus indicating that this chromosomal aberration was, in fact, a complex three-way rearrangement t(8;21;8)(p23;q22;q22). In conclusion, combining conventional karyotype, FISH or RT-PCR analyses is a rational strategy for identification of the complex variants of t(8;21) translocation.

Changes and significance of circulating hepatocellular carcinoma cells in recurrent hepatocellular carcinoma patients after combined treatment.

OBJECTIVE: To study the changes and significance of circulating hepatocellular carcinoma cells in recurrent hepatocellular carcinoma (HCC) patients after combined treatment of transarterial chemo-embolization(TACE) and percutaneous ethanol injection(PEI). METHODS: We detected 19 blood samples from the recurrent HCC patients by nested RT-PCR to find out AFP mRNA before and after the treatment. RESULTS: There were 7 patients with AFP mRNA positive before treatment (36.8%), and none of patients with AFP mRNA positive after treatment. CONCLUSIONS: Combined treatment of TACE and PEI can effectively eliminate circulating hepatocellular carcinoma cells and thereby prevent metastasis and recurrence of HCC.

[The distribution and homology of mitochondrial plasmid-like DNAs in different cucumber varieties].

Four kinds of mitochondrial plasmid-like DNA, designated pC1, pC2, pC3 and pC4, have been found in cucumber (Jinyan No. 4). Their distribution in 14 Cucumber varieties was analyzed. Plasmid-like DNAs were detected in Jinchun No. 2, Jinchun No. 5, Jinxinmici, Jinlu No. 4 and Jinyan No. 4, and the rest 9 varieties contained no plasmid-like DNAs, suggesting that their distribution is irregular. There was homology among the same plasmid-like DNA family in different varieties. pC4 showed homology to the nuclear DNA of Jinyan No. 4, in addition to the nuclear genomes of other 7 varieties either with plasmid-like DNAs or without. The homologous sequences of pC4 in the cucumber nuclear DNA were repetitive. Sequences homologous to pC4 were also found in the nuclear genomes of towel gourd and pumpkin (other cucurbitaceous plant). Therefore, we propose that mitochondrial plasmid-like DNA occurred before cucumber diverged from cucurbit and had integrated into the nuclear DNA. The varieties without plasmid-like DNAs might lost them during evolution.

Field monitoring of toxic organic pollution in the sediments of Pearl River estuary and its tributaries.

Field monitoring of the toxic organic compounds (PCBs, PAHs, organochlorine pesticides) in the top sediments of Pearl River Estuary and its up-streams were made. It was found that the highest concentrations of these toxic organic compounds occurred in the sediment sampled at Macau inner harbor (ZB013), which is a sink of suspended fine particles transported from the upstream waterways. Because of the affinity of the hydrophobic organic compounds (PAHs, PCBs) for the solid phase, these fine particle depositions led to accumulation of these compounds in the sediment of Macau. The atmospheric dry deposition may be another source of the toxic organic pollution in the sediment.

[Analysis of toxic organic compounds in MGP-Wastewater and investigation of Wastewater's general characteristics].

A full-scale investigation and analysis of MGP(Manufacture Gas from Petroleum)-Wastewater was carried out in this study. It's the first time that this paper publicizes the wastewater's general data, pretreatment method of GC/MS analysis and compositions of toxic organic compounds. The concentrtion of wastewater's CODCg, BOD5, phenolic, N-NH4+ and extractive-organic is: 400 mg.L(-1)-600 mg.L-1, 60 mg.L(-1)-80 mg.L-1, 20 mg.L(-1)-30 mg.L-1, 60 mg.L(-1)-80 mg.L-1 and 170 mg.L(-1)-200 mg.L-1 respectively. There are 79 kinds of aromatic compounds were detected by GC/MS, the total concentration of influent and effluent is 140.79 mg.L-1 and 129.11 mg.L-1 respectively. The data approved that MGP-wastewater is toxic and difficult for bio-treatment.

[Vertical distribution characteristics of organochlorinated pesticides in sediment core from Macao estuary, Pearl River delta].

This work presents the analysis result of concentration for organochlorinated pesticides in a sedimentary core sample. Pesticides were quantified by GC/ECD with the internal standard material. The core dated by 210Pb methods. The vertical distribution of pesticides concentration with the depth was discussed. The results showed the concentration of BHCs and DDTs in the core ranged from 0.48 ng/g to 26.28 ng/g and 1.92-39.13 ng/g respectively. The distributing characteristics of organochlorinated pesticides in the core varied with the water fluxes of Pearl River. Organochlorinated pesticides may come from the residue of agricultural soils. Concentration of DDTs was above the effects range-low value for marine and estuarine sediments.

[The function of 5'flanking region of mouse neuronal CDC2-like kinase regulatory subunit p35].

OBJECTIVE: To clarify the possible mechanisms which control p35(nck5a) neural-specific expression. METHODS: Thirteen luciferase expression vectors which contain different parts of 5'flanking sequence were constructed. Transient expression assay, DNase 1 hypersensitive site(DHSS) assay and primer extension assay were made to reveal the possible mechanisms of p35(nck5a) expression control. RESULTS: It was found that 5'flanking sequence of p35(nck5a) contained 5455bp and there were a few possible promoters in the fragment. The result of transient expression assay in primary cerebrum cortex cell of rat (Neuron) and established non-neuron cell such as HeLa showed the loss of neural-specific expression of all the luciferase expression vectors. The result of DNase 1 hypersensitive site assay revealed a DHSS located at the -400bp upstream of p35(nck5a) in brain but not in liver. The primer extension assay found the putative transcription initiation site near the position of DHSS. CONCLUSION: The results above suggest that there is no cis-element to control the neural-specific expression in the 5'flanking sequence, and the control on the chromosome level may be critical to the neural-specific expression of p35(nck5a).

Effect of Salvia miltiorrhiza Bunge injection on anticardiolipin antibody production induced by beta2 glycoprotein.

AIM: To explore the therapeutic effect and the mechanism of Chinese herbs on antiphospholipid syndrome (APS) by observing the effect of Salvia miltiorrhiza Bunge injectio (SmBI) on anticardiolipin antibody (aCL) induced by beta2 glycoprotein I (beta2-GP I). METHODS: Sixty female mice randomly fell into 6 groups: group A, B, C, D was injected through abdominal cavity with different dosage of SmBI daily; after 14 d, group A, B, C, E was immunized with 150 microg of purified human beta2-GP I in complete Freund's adjuvant subcutaneously; group F as control. The titre of aCL were detected by enzyme linked immunosorbent assay; subsets of T cell were grouped by streptavidin-biotin complex technique; and the activity of IL-2 was measured by MTT chromatometry. RESULTS: (1) Compared with group E, the absorbance (A) of aCL in group A, B, and C was decreased (P < 0.05 or P < 0.01). By linear correlation, the dosage is negatively correlated with the A values of aCL in 1, 2, and 3 weeks (P < 0.01). (2) Compared with group E, TH/TS ratio was reduced in group A, B, and C (P < 0.05 or P < 0.01); there is no significant differences between group D and F (P>0.05). By linear correlation, the dosage is negatively correlated with TH/TS ratio (P < 0.01). (3) Compared with E, the activity of IL-2 in group B and C decreased significantly (P < 0.01). By linear correlation, there is negative correlation between dosage and IL-2 activity (P < 0.01). There is no significant difference between D and F (P > 0.05). (4) There is positive correlation between TH/TS ratio and IL-2 activity in different dilutions (P<0.01). CONCLUSION: The mechanism of suppressive effect of SmBI on aCL induced by beta2-GP I may be realized by resuming the elevated TH/TS ratio and IL-2 activity. The state that SmBI have no effect on normal mice indicates that SmBI has selective immunoregulative functive.

[Mechanism of multidrug resistance caused by retinoic acid].

OBJECTIVE: To investigate the regulatory mechanism of multidrug resistance(MDR) caused by all-trans retinoic acid (ATRA). METHODS: ATRA and IL-4 were used to treat human hepatoblastoma cell line (HepG2) cells. The proliferative activity, synthesis of alpha fetal protein (AFP), and cell cycle distribution of tumor cells were observed to evaluate the degree of cell differentiation. Flow cytometry and in situ hybridization were used to determine the expressing levels of p53, bcl-2, P-glycoprotein (P-gp) and c-jun and c-myc mRNA. MTT assay was used to evaluate the sensitivity of the tumor cells to chemotherapeutic agents. RESULTS: Both ATRA and IL-4 could induce the differentiation of HepG2 cells. ATRA treatment of the tumor cells led to drug resistance in chemotherapy (resistant factors: 1.6-3.1), and IL-4 increased the sensitivity of the tumor cells to antineoplasic drugs (reversal index: 4-17). The level of P-gp expression in ATRA-treated cells was increased from 54.2% +/- 8.6% up to 98.5% +/- 1.4% (P < 0.01), but IL-4 markedly inhibited expression of P-gp down to 25.4% +/- 7.3% (P < 0.01). Both ATRA and IL-4 treatment could down-regulate c-jun and c-myc mRAN expressions. The level of p53 and bcl-2 expression could be up- or down-regulated by IL-4 treatment but they were unaffected by ATRA treatment. CONCLUSION: Degree of cell differentiation and level of c-jun and c-myc mRNA expression might not be related to change in drug sensitivity by inducing differentiation of HepG2 cells with ATRA and IL-4. Increased expression of P-gp caused by ATRA might be one of the factors up-regulating MDR. p53 (or bcl-2) might be involved in regulating the sensitivity of antineoplastic drugs by inducing differentiation.

[Gene marking of human colon cancer LoVo cells with enhanced green fluorescent protein].

OBJECTIVE: The aim of this study is to obtain a human colon cancer cell line marked with enhanced green fluorescent protein (EGFP) gene. METHODS: The G1FP retroviral vector, carrying EGFP gene, was constructed. When combined with liposome-mediated transfection with cross infection, EGFP virus producer cells were generated. The human colon cancer LoVo cells were transduced with EGFP retrovirus. The integration and expression of EGFP gene were analyzed with polymerase chain reaction (PCR) and flow cytometry. RESULTS: The ecotropic EGFP virus was obtained by cocultivation of packaging cells for two weeks, while the amphotropic virus producer cells Am12/G1FP, expressing high-level EGFP, were generated through cross infection with a titer of 1.1 x 10(5) infectious particles/ml. The genetically modified LoVo/GFP cells gave off strikingly bright green fluorescence, in which the integration of EGFP provirus was confirmed by PCR analysis. CONCLUSION: The LoVo/GFP cells, which express high-level green fluorescence, may provide an useful tool for the study of micrometastasis of colon cancer in vivo.

[Preliminary study on CTL enhancement induced by IL-4 gene modified HL-60 cells].

OBJECTIVE: To explore the mechanism of enhancing killing activity of tumor cell-specific cytotoxic T lymphocyte (CTL) by IL-4 gene modified tumor cells (IL-4mTC). METHODS: IL-4 gene was introduced into HL-60 cells through retroviral vector PL-IL-4-SN. Wild and IL-4 mTC were used to induce CTL responses, and cell surface molecules were assayed by flow cytometry. RESULTS: The killing activity of tumor cell-specific CTL in IL-4 mTC group was increased from 11.8% to 77.2%, about sevenfold higher than that induced by wild HL-60 cells (P < 0.01). High level expression of MHC class II antigens as well as B7-1 and ICAM-1 molecules was observed in IL-4 mTC. The expression of MHC class I antigen was not affected by IL-4 gene modification. The expression of cell surface molecules induced by IL-4 mTC was completely abrogated by anti-IL-4 McAb. A significant increase of IL-2 secretions was detected during IL-4 mTC inducing CTL responses. IL-2 secretion and CTL response were inhibited by anti-IL-4 or anti-surface molecule McAbs. CONCLUSION: IL-4 gene modification might enhance the tumor cell-specific CTL killing activities by inducing cell surface molecules expression and IL-2 secretion.

[Discovery of natural variant strain of HIV1 in env gene C2-V3 encoded PND of membrane protein].

OBJECTIVE: To sequence and analyze the env gene C2-V3 region of proviral genome from a HIV1 isolate(WWBH7) which was obtained from Huadong Area in China. METHODS: The env gene C2-V3 DNA fragment was amplified by nested-primer PCR with genome DNA of eripheral blood mononuclear cells from a confirmed HIV1 infected individual as a template. The amplified DNA fragment was inserted into pGEM-T vector. The recombinant plasmid was confirmed by restriction enzyme analysis. The inserted DNA fragment was sequenced by ABI737 autosequencer and analyzed by PROSIS software. RESULTS: The HIV1 strain was the derivatives of HIV1 B subtype. But there was mutation of 192 bp fragment repeated insertion at env C2-V3 region of the HIV1 strain compared with standard HIV1 B subtype such as SF2 strain. The mutation brought about a double V3 region in gene encoded PND (principal neutralizing domains). The DNA sequence was registered in GenBank (AF220245). CONCLUSIONS: This was a natural mutated variant strain of HIV1 whose genome showed a 192 bp repeated insertion at C2-V3 region of env gene encoded PND of membrane protein.

[Investigation of the bioadhesion of carbopol and hydroxypropyl methylcellulose to rat gastrointestinal mucosa in vivo and in vitro].

AIM: To investigate bioadhesion of carbopol and hydroxypropyl methylcellulose of different viscosity grade to rat gastrointestinal mucosa. METHODS: The maximum adhesion force has been adopted as an index of adhesive evaluation in vitro, gastro-emptying rate and migrating length in intestine have been adopted as an index of adhesive evaluation in vivo. RESULTS: In vitro adhesion studies indicated that the adhesive force of low-vicosity materials was stronger than that of high-vicosity materials, while the transferation rate of high-vicosity materials is significantly higher compared to that of low-vicosity materials in vivo. CONCLUSION: Adhesive force of materials gives no correlation with rate of the materials in rat; Cb934 have the optium bioadhesion among the studied materials and can be used as the preferred adjuvant in oral bioadhesion preparation.

Primary LAMP-2 deficiency causes X-linked vacuolar cardiomyopathy and myopathy (Danon disease).

"Lysosomal glycogen storage disease with normal acid maltase" which was originally described by Danon et al., is characterized clinically by cardiomyopathy, myopathy and variable mental retardation. The pathological hallmark of the disease is intracytoplasmic vacuoles containing autophagic material and glycogen in skeletal and cardiac muscle cells. Sarcolemmal proteins and basal lamina are associated with the vacuolar membranes. Here we report ten unrelated patients, including one of the patients from the original case report, who have primary deficiencies of LAMP-2, a principal lysosomal membrane protein. From these results and the finding that LAMP-2-deficient mice manifest a similar vacuolar cardioskeletal myopathy, we conclude that primary LAMP-2 deficiency is the cause of Danon disease. To our knowledge this is the first example of human cardiopathy-myopathy that is caused by mutations in a lysosomal structural protein rather than an enzymatic protein.
