Antimicrobial effects of some dicarbonyl and tricarbonyl sugar hydrazone derivatives.

Several dicarbonyl and tricarbonyl sugars were prepared by the use of fungal enzymes and the antimicrobial effects of their N,N-diphenylhydrazine derivatives were tested. G+ bacteria were more sensitive than G- bacteria especially in the group of disubstituted compounds. Peracetyled derivatives were not active. No inhibition of yeast growth was found.

Degradation of 2-chlorobenzoic and 2,5-dichlorobenzoic acids in pure culture by Pseudomonas stutzeri.

A strain of Pseudomonas stutzeri KS25 utilizing 2-chlorobenzoic and 2,5-dichlorobenzoic acids as the sole carbon and energy source was isolated from polychlorophenol-contaminated soil and sewage, using the method of enrichment cultures. This strain was also able to grow on 2-fluoro-, 2-iodo-, 2-bromo- and 2,5-dihydroxybenzoate, but did not utilize 3-, 4-chloro-, 2,4- and 2,6-dichlorobenzoates as the sole carbon and energy source, however, it cometabolized 3-chloro-, 2,4- and 2,6-dichlorobenzoates, but not 4-chlorobenzoate. The yield of released chlorine during utilization of 2-chloro- and 2,5-dichlorobenzoates amounted to 100% of the theoretical. The concentration of 2-chloro- and 2,5-dichlorobenzoates, not substantially inhibiting the isolated microorganism, was within the range 0.25-0.5 and 2.5-3.0 g/L, respectively.

The psychopathology of suicide in the north western Peloponnesus.

A postmortem inquiry of 105 legal documents concerning suicide deaths obtained from the Office of the District Attorney covering the province of Achaia was studied for the 15-year period 1970-1984. The suicides were classified into two groups; group A consisting of 63 suicides (with a history of psychiatric illness) and group B consisting of 42 suicides (with no history of psychiatric disturbance). The findings suggest that people with a history of psychiatric illness committing suicide are disorganized in their personal and social life, with frequent previous suicidal attempts. Whereas, the nonpsychiatrically diagnosed group seemed to commit suicide under outside pressures which dislocated their lives progressively or abruptly. The differences between the two suicide groups are discussed and compared with similar studies regarding suicidal behavior.

Illness behaviour in mothers with young children.

113 families with young children were studied over a 12-month period, when all contacts with the general practitioner and hospital emergency services were recorded. Socioeconomic data was obtained at interview, together with responses to a vignette instrument describing common childhood problems. Health diaries were completed by 70% of mothers, providing further information about illness and mothers anxiety about it. Analyses showed that significant disease in any child was the strongest predictor of frequency of new consultations, with measures of mothers anxiety, number of children in the family, mothers education level and her response to the vignettes also contributing significantly. Socioeconomic deprivation was associated with increased utilisation, even when the effects of prevalence of chronic diseases, size of family and mothers' anxiety were taken into account. From the spontaneous comments made during the vignette responses further information was obtained about how mothers reach the decision to consult the family doctor about children's illness. Thus in a Primary Care setting some systematic differences have been demonstrated between high and low consulting families, which may partly explain the wide variation in spontaneous consultation rates observed.

Child consultation patterns in general practice comparing "high" and "low" consulting families.

All children's consultations with their general practitioner over a 12 month period in a small urban practice were analysed. Overall consultation rates ranged from 2.2 per child a year for 8 to 11 year olds, to 6.8 for those under 2. Families were grouped according to their average rate of new consultation for children, standardised for age. Families with higher consulting rates scored higher on an index of economic disadvantage, with mothers who scored higher on a test of "tendency to consult" and who were less educated than those in lower consulting families. The presence of any doctor-defined "significant disease" in any child was highly correlated with the family's consultation rate.

Accumulation of low molecular weight DNA and changes in chromatin structure in HeLa cells treated with human fibroblast interferon.

The addition of human fibroblast interferon (IFN-beta) (100 units/ml) at the S/G2 boundary of the cell cycle of synchronously grown HeLa cells is characterized by the accumulation of newly synthesized low molecular weight DNA and changes in chromatin assembly. In addition, there is a 3-fold stimulation in the incorporation of tracer amounts of [3H]thymidine, but not [3H] deoxyguanosine, into DNA and a 2-fold increase in the incorporation of [3H]dTTP into the DNA of isolated nuclei. Fluorescence-activated cell sorting by laser flow cytometry revealed that IFN-beta-treated cells were delayed in entering and passing through the S phase. The inhibition of proliferation of HeLa cells treated with IFN-beta is characterized by a 3-fold accumulation of newly synthesized DNA of Mr less than 56 X 10(6) compared to untreated cells as determined by alkaline sucrose gradient centrifugation. The newly synthesized DNA in IFN-beta-treated cells was replicative and not repair DNA. The observation that IFN-beta inhibits the processing of newly synthesized low molecular weight DNA into normal DNA might be explained by the intracellular accumulation of S-adenosylhomocysteine in IFN-beta-treated HeLa cells (de Ferra, F., and Baglioni, C. (1983) J. Biol. Chem. 258, 2118-2121) which could change the soluble ribonucleotide and deoxyribonucleotide pool and ultimately affect DNA processing. Interferon may also affect processing of DNA by interfering with normal chromatin assembly. Evidence for the effect of IFN-beta on chromatin assembly is provided; we have observed a more condensed structure in IFN-beta treated cells by circular dichroism spectroscopy. Simultaneous with the affect on chromatin assembly, there is a 70% decrease in poly(ADP-ribosylation) of either histone and/or non-histone proteins. The loss of coordination between the pool size for DNA synthesis, decreased postsynthetic modifications of chromatin, and normal chromatin formation may explain the inability of the cell to differentiate and to continue cell division.

Specific lysis of Plasmodium yoelii infected mouse erythrocytes with antibody and complement.

An assay for complement-dependent antibody-mediated cytolysis of Plasmodium yoelii infected mouse erythrocytes was designed. By means of a rat hyperimmune serum to P. yoelii the presence of parasite associated antigens was demonstrated on erythrocytes from P. yoelii infected mice. Tests with such erythrocytes fractionated on Percoll gradients showed that the target antigens for the cytotoxic antibodies were confined to the surface of only infected erythrocytes containing late stages of the parasite. The cytotoxic antibodies were of IgG class.

Serum calcium and magnesium levels in chronic schizophrenics.

Serum calcium and magnesium levels were studied during neuroleptic treatment in a group of 29 chronic schizophrenic inpatients who had previously remained drug free for at least 4 weeks. The drug-free values of both electrolytes were not significantly different between patients and controls. In three patients who developed catatonic stupor during the drug-free period, calcium was significantly increased at the onset of the catatonic manifestations. Both electrolytes were significantly decreased in the whole group during treatment; the magnitude of the decrease was independent of the neuroleptic used. No correlation was found between the observed increase in prolactin and calcium changes. Patients who remained free of extra-pyramidal symptoms had a higher mean drug-free magnesium to calcium ratio compared to those who developed such symptoms. Our study provides evidence for an interconnection between serum electrolyte changes and neuroleptic drug action.

Cytotoxicity of tumour-bearer serum to D23 hepatoma cells derived from solid tumours, ascites or in vitro cultures.

The complement-dependent cytotoxicity of antibodies in tumour-bearer serum (TBS) from rats carrying the chemically induced D23 hepatoma was investigated. Target cells were D23 cells from solid tumours (D23sol), from ascites tumours (D23asc), or from in vitro growing cell cultures (D23cc). The D23asc and D23cc cells were not lysed when used as target cells in the assay, although they evoke cytotoxic antibodies when growing in vivo. The D23 ascites cells became susceptible to complement-dependent lysis after trypsin treatment. This was, however, not due to unmasking of target antigens, since untreated D23 ascites cells absorbed cytotoxic antibodies as efficiently as trypsinized cells. No increase in susceptibility to complement-dependent lysis was observed after trypsin treatment of D23cc cells. Absorption of cytotoxic antibodies with D23cc cells showed no or very low antigen expression on the surface of these cells. They did, however, contain the antigen(s), since 3 M KCl extracts of the D23cc cells could inhibit the complement-dependent cytotoxicity of D23sol TBS against D23sol cells. From these data it was concluded that the susceptibility of hepatoma cells for antibody-mediated complement lysis is not only correlated with antigen expression, as was the case with the in-vitro-cultivated cells, but is also dependent on increased lytic susceptibility after trypsin treatment of the cells.

Tumour-associated antigens reacting with cytotoxic antibodies in serum of hepatoma-bearing rats.

3M-KCl extracts of the hepatoma D23 contain antigens that inhibit the complement-dependent cytotoxicity for D23 hepatoma cells of serum from D23 tumour-bearing rats (D23 TBS). Inhibition was not due to a general anticomplementary activity of the extracts. Although a minor part (25%) of the protein of D23-KCl extract was insoluble in PBS, this part contained most of the inhibitory activity. Fractionation of the PBS-soluble material of the extract on Concanavalin A-Sepharose showed that the inhibitory activity did not bind to the lectin. Analysis of D23-KCl extracts on a Sepharose CL-4B column showed that the antigens involved in the cytotoxicity were heterogeneously distributed in the high-mol. wt region (greater than 200,000). Precipitation with 10% trichloroacetic acid (TCA) of D23 KCl extracts revealed that most of the antigenicity was insoluble in TCA. Heating of D23 KCl extracts at 100 degrees C did not affect the antigenicity. Enzyme treatment of D23 extra nuclear membranes (D23 ENP) revealed that the inhibitory activity was not sensitive to proteolytic digestion, while treatment with phospholipase A2, C or D abrogated partly the inhibitory activity. The lipid nature of the antigenicity was indicated by its solubility in organic solvents as chloroform or n-butanol.

Determination of the immunoglobulin class of complement-dependent cytotoxic antibodies in serum of D23 hepatoma-bearing rats.

The immunoglobulin class of the complement-dependent cytotoxic antibodies in serum from D23 hepatoma-bearing rats (D23 TBS) for D23 hepatoma cells was analysed. When studied by affinity chromatography with concanavalin A, protamine, or staphylococcal protein A conjugated to Sepharose, the cytotoxic activity bound to the former two but not protein A. The binding fractions were further characterized by column chromatography on Sepharose CL-4B. The cytotoxic activity was recovered exclusively in the high molecular weight fractions corresponding to human IgM. Monitoring with IgG- or IgM-specific rabbit antibodies indicated that these high molecular weight cytotoxic fractions contained both IgG and IgM. However, fractionation of D23 TBS at low pH suggested that cytotoxicity was due to IgM antibodies rather than to immune-complexed IgG antibodies. This was supported by the findings that rabbit antirat IgM antibodies inhibited the cytotoxicity of TBS completely when added at high dilutions.
