Potential histamine H2-receptor antagonists. Synthesis, conformational studies and activity of novel 3-oxo-1,2,5-thiadiazole 1,1-dioxide derivatives.

New histamine H2-receptor antagonists bearing a novel "urea equivalent", the 3-oxo-1,2,5-thiadiazole 1,1-dioxide ring, have been synthesized in a transamination reaction. Open chain derivatives have also been obtained. Theoretical conformational analysis of the compounds has been carried out using a molecular modelling program. Semiempirical CNDO/2 calculations have also been performed. The antisecretory and cytoprotective activities of the compounds have been evaluated.

Animals

[Study of lipids (cholesterol, triglycerides and phospholipids), plasma lipoproteins (HDL-cholesterol) and apoproteins (apo A and apo B in patients with acute myocardial infarction].

Several lipid parameters were evaluated in 88 patients with acute myocardial infarction (AMI). A reduction in cholesterol and phospholipid level was observed, with minimal values between the days 10-20, followed by a subsequent increase. Triglycerides increased after 12-14 hours, with a maximal value after 20-30 days. Cholesterol bound to high-density lipoproteins (HDL-cholesterol) decreased after 24 hours, with a minimal value after 20-30 days. Apoprotein A decreased from the initial measurement until days 20-30. The initial lipid profile (indicating the risk of coronary artery disease) was constituted by high cholesterol (particularly in males below 60 years), high triglyceride and low HDL-cholesterol levels. The latter finding was more marked and characteristic in the overall group of evaluated patients. All values were compared with a control group of 24 healthy persons. Relevant data for prognosis included the following: males above 60 years had a greater reduction in cholesterol and apoprotein A during AMI; the males who died had lower apoprotein A levels than the survivors; females with higher haptoglobin levels also had higher apoprotein B levels. All these findings were statistically significant (p less than 0.01).

[Activities of twelve enzymes in the blood plasma of rainbow trout and tench subjected to various forms of thermal disturbance].

The levels of ornithine carbamyl transferase, acid phosphatase, alkaline phosphatase, leucine amino peptidase, creatine kinase, amylase, glutamic oxalacetic transaminase, glutamic pyruvic transaminase, lactate deshydrogenase, hydroxybutyrate deshydrogenase, glutamate deshydrogenase and malate deshydrogenase were determined in the plasma of Rainbow Trout and Tench submitted to water temperature increases. In the Rainbow Trout a thermal shock from 12 to 21 degrees, increases activities of some enzymes while temperature increase up to pre-mortem stage causes very important changes in enzymatic levels. In the Tench a thermal shock from 12 to 28 degrees causes more changes of enzymatic activities than a shock from 12 to 25 degrees. In Tench acclimated to 25 degrees, various enzyme levels are increased in comparison with 12 degrees control animals. A high potassium level in water causes complex changes in enzyme levels. The most sensitive enzymes to thermal disturbance are GOT and GPt transaminases which increase whatever the aggression form, amylase when thermal disturbance is moderate, alkaline phosphatase and malate deshydrogenase in case of strong thermal stress. The study of these enzymes is recommended for watching the state of fishes living in artificially heated waters.

[Immediate effects of thermal shocks on the plasma level of various components in the Rainbow trout: compounds indicating stress and protein fraction].

Rainbow trout were subjected to thermal shocks (9 degrees water temperature increase, 1 hr stay at 21 degrees and return to initial temperature) at the rate of 2 shocks a day during 1 day or 3 successive days. The observed changes only show a moderate reaction. cAMP does not vary; lactate slightly increases at 17 hr after the end of the shocks. Glucose seems to be the most reliable stress indicator; it increases at 2 hr and remains again above control value at 17 hr after a 3 day shock time. Fibrinogen increases after a 3 day shock time. Lastly, a strong decrease in low density lipoprotein level is seen at 17 hr following both shock duration times and from 2 hr in case of a 3 day shock time.

Animals

[Conditions for greater survival of neonatally thymectomized Swiss mice (author's transl)].

A total of 385 albino Swiss mice, of both sexes, between 24 and 30 hours of age are neonatally thymectomized, by means of a suction pump method. One batch is operated under ether anesthesia, another with sodium pentobarbital, and a third one with Cryoanesthesia. The last batch is subdivided into 2. In one of these the mother is anesthetized with sodium pentobarbital intraperitoneally, during the postoperative recovery of the litter, and in the other the mother is left without treatment. The mothers of the first two batches of thymectomized animals are anesthetized with the same products as their offspring. The percentage of survival of thymectomized breeding mice in greatest in the batch operated under Cryoanesthesia and whose mothers received the above mentioned treatment. Statistically the differences are very significant and they might be accounted to the fact that the mothers are anesthetized while their offspring normalize their physiological constants with better recovery in feeding, hydration, shelter and rest.

Anesthesia, General

The perchlorosoluble proteins of the serum of the rainbow trout (Salmo gairdnerii Richardson): albumin like and hemoglobin binding fraction.

1. The main perchlorosoluble fraction of rainbow trout serum has some physico-chemical characters kindred to those of human serum albumin (low molecular weight, solubility with ammonium sulfate, electrophoretic mobility, no glycoproteinic staining). 2. However, on account of obvious differences (heterogeneity and existence of various phenotypes, lack of bromophenol blue or bilirubin binding, low concentration, solubility in perchloric acid), the term "para-albumin" seems more suitable to name this compound. 3. The perchlorosoluble fraction binds hemoglobin causing an increase of peroxidasic activity. But, unlike to human haptoglobin, hemoglobin binding is partial, reversible and labile.

Animals

Antibody-like activity to polyacrylic acid in normal rabbit sera.

Most unimmunized rabbit sera agglutinate acrylic particles coated with polyacrylic acid. The factor responsible for the agglutinating activity turned out to be a 2-mercaptoethanol-resistent immunoglobulin, present in low concentration. It was tentatively classified as "natural antibody". This is assumed to have arisen upon previous contact with naturally occurring cross-reacting polyanions.

Acrylates

Acute renal insufficiency in the rabbit by glycerol.

The appearance of an acute renal insufficiency in the rabbit, after glycerol injection (10, 13 or 15 ml/kg of a 50% solution) is investigated. After a 24 hours of intoxication, especially in the ten following days, cylinders, erythrocytes and renal cells appear in the urine sediment. Proteinuria appears after 24 hours and practically disappears after 72 h. Glucosuria persists from 24 hours to 6 days. Haemoglobinuria is intense after 24 and 48 hours and persists slightly about 6 days. Na, K and Cl elimination in urine diminishes clearly in all animals. Plasma K increases in non-surviving animals and does not change in those surviving. Plasma Na does not change in the dying ones, and decreases in those surviving. In non-surviving animals, pH, pCO2 and CO3H minus decrease sharply. In the surviving ones pCO2 decreases clearly after 24 hours, increasing afterwards slowly to normal values. pH increases, slightly during the first 48 hours, and then neatly during approximately 6 days. Standard CO2H minus does not change during the first 48 hours, increasing afterwards during 6 to 7 days. Histologically, the chief lesion is a vacuolar degeneration of the proximal tubule. The possible mechanisms of such alterations are discussed.
