Enhanced activity of lysosomal beta-galactosidase after silicone implantation: an experimental study in rats.

Biologic consequences of silicone implantation may include changes in host connective tissue metabolism. Lysosomal beta-galactosidase (beta-GAL) activity, which is a sensitive marker of fibrotic diseases and may be a useful marker of collagen turnover, was examined in the serum of rats with implanted silicones. No significant difference in spectrofluorometrically determined enzyme activity was demonstrated in rats subjected to dorsal submuscular pocket dissection without implantation and corresponding nonoperative controls. Rats with implanted solid silicone elastomer or free polydimethylsiloxane gel (both components obtained from mammary implant) revealed enhanced activity of serum beta-GAL. Higher enzyme activity was observed in animals with implanted silicone gel with a peak level of 2.73 +/- 0.08 pmol/30 min/ml 16 weeks after implantation. Increased collagen deposition and capsular thickness was demonstrated around implanted gel material as compared with that around elastomer shell. Animals with implanted absorbable and nonabsorbable materials, polyglactin and Teflon (polytetrafluoroethylene), respectively, after initial increase of beta-GAL activity demonstrated enzyme activity within the normal range. Findings indicate that there is enhanced lysosomal beta-GAL activity after silicone implantation in rats. Clinical relevance and its possible significance as a predictor or indicator of local or systemic fibrosis after silicone implantation seems worthy of further investigation.

Activity of specific IgG, IgM and IgE antibodies in human trichinellosis.

The activity of IgG, IgM and IgE antibodies to somatic antigen of Trichinella spiralis in the sera of patients with trichinellosis at various intervals after infection was examined by means of ELISA. Mathematical analysis of the dose-response curves was used. Elevated level of IgG and IgM antibodies of relatively high avidity and of rather low IgE avidity was documented. Amount or avidity of IgG antibodies was found to be most useful for the diagnosis of trichinellosis (85% positive results in patients' sera). The isotype of IgM avidity constitutes a better diagnostic value than the amount of it (60% and 35% of positive results, respectively).

Characterization of antibody response to Pseudomonas aeruginosa in patients with wound infections.

ELISA was used to measure the amount and avidity of IgG antibodies to exotoxin A (ExA) and 7 Fisher's immunotypes of lipopolysaccharide (LPS) in the sera of 13 patients with mild or moderate Pseudomonas aeruginosa infections. Changes in the specificity of tested sera during the course of infection were demonstrated. A statistically significant increase was seen in the amount and avidity of the antibodies to ExA in a majority of the sera, and an increase was seen in amount of antibodies to LPS immunotype 4 in the sera of patients with moderate infections.

[A simplified method of rapid hemagglutination test for determining the levels of tetanus antibodies].

A new, simplified methodology of preparation of lyophilized chrome-coated turkey blood cells (formalin-treated ) for rapid identification of tetanus antibodies in indirect hemagglutination test is described. Blood cells diagnostic preparations obtained in this way were easier and three times faster to prepare than tanned cell preparations . They maintained unchanged capability for specific agglutination during at least one year of storage when kept at 4 degrees C. proposed methodology enable to start a production of laboratory kits necessary for controlled prophylaxis of tetanus.

Amount, avidity, and specificity of antibodies to Pseudomonas aeruginosa in normal human sera.

Seventy-two normal human sera from healthy blood donors were tested by an enzyme-linked immunosorbent assay in order to determine the amounts and avidities of immunoglobulins M and G antibodies to lipopolysaccharides of seven Fisher's immunotypes of Pseudomonas aeruginosa and to exotoxin A. The patterns of specificity for seven immunotypes in all individual sera were determined. These data show a predominance of antibodies directed to Fisher's immunotypes 7 and 4 in the human population tested and may reflect frequency of occurrence of immunotypes outside the hospital environment.

The cell-surface components of Bacteroides thetaiotaomicron as coating antigens in ELISA test.

Cell-surface antigens were extracted out of three Bacteroides thetaiotaomicron strains of different origin. Lipopolysaccharides, their fractions, L1 preparations and capsular antigen were obtained. All substances were tested as coating antigens in ELISA test against antibacterial rabbit immune sera. The highest absorbances were observed with lipopolysaccharides (LPS), their polysaccharide fractions (PS) and capsular material (CPS). Lipopolysaccharides after nuclease treatment (N-LPS), free from nucleic acids, were more active than crude phenol-water extracts (PW-LPS).

Characterization of the specificity of lipopolysaccharide antigens from seven Fisher's immunotypes of Pseudomonas aeruginosa, using ELISA technique.

Lipopolysaccharides of seven Fisher's immunotypes of P. aeruginosa, extracted by water-phenol method, were fractionated on Sepharose 2B column. On the basis of molecular weight sugar and KDO content, eluents were separated into 4 fractions. An analysis of the antigenic specificity of the chromatographic fractions of seven immunotypes of LPS was carried out, using sera of mice vaccinated with several crude LPS preparations or whole-cell suspensions each of P. aeruginosa immunotypes, by ELISA. The antigenic specificity of fraction 1 and 2 of several immunotypes (with the exception of LPS from immunotype 2) in reaction with mice antisera for crude LPS was shown. Not quite full specificity of fractions 1-3 of all LPS preparations during analysis of these fractions reactivity with antisera to whole P. aeruginosa cells were observed, but specific reactions predominated in all test systems except LPS 2.

Enzyme-linked immunosorbent assay (ELISA) for the detection and differentiation of Clostridium botulinum toxins type A and B.

Affinity chromatography has been used for a two-step purification of commercial horse botulinum antitoxic globulins type A and B. The first step performed using CH-Sepharose 4B conjugated to toxin type A (or B), permitted the removal of non-botulinal antibodies from antitoxic globulin type A (or B). The anti-botulinal antibodies obtained from the first step were cross-absorbed in the second affinity chromatography using CH-Sepharose 4B conjugated to toxin B (for the purification of antibodies to type A) or to toxin A (for antibodies to type B). The antibodies obtained were used to coat polystyrene wells in an ELISA for the detection of botulinum toxin type A and type B. The first purification step increases the sensitivity of such an ELISA whereas the second step improved the specificity of the test. Only slight cross-reactions were observed between the type A and type B detection systems. The sensitivity achieved with ELISA was 100 and 300 DLM (dosis lethalis minima) for type A and B respectively.

Host serum proteins in Taenia saginata metacestode fluid.

Taenia saginata cyst fluid was examined for host proteins; IgG1 and IgG2 as well as haemolytic complement activity were detected. Polyacrylamide gel electrophoresis revealed differences in proteinograms among the samples taken from 1-, 4-, and 10-month old cysts. Fluid from older cysts had fewer protein components and showed a weaker antigenic reaction with sera of bovines infected with T. saginata than that of younger cysts. The roles of antibody and complement in initiating degeneration of the parasite are discussed.

Determination of soluble antigens of Clostridium botulinum A by chemiluminescent--immunosorbent assay (CLISA).

The chemiluminescent--immunosorbent assay (CLISA) was adopted for Cl. botulinum A soluble toxic antigens determination. Luminol (ABEI) labelled botulinum antitoxin globulins showed a strongly positive specific immunochemiluminescent reactions with the native botulinum toxin preparations coupled (adsorbed) on polystyrene balls. The sensitivity of the reaction reached 20 DLM/ml (5,000 light impulses per 40 sec) in comparison with 2 X 10(6) DLM (57,000 impulses), control preparations (1,500 impulses) and the background (150-300 impulses). The results give a perspective further investigations with the CLISA for rapid indication of Cl. botulinum toxic antigens.

[Non-invasive methods in the diagnosis of latent heart rhythm disorders].

In the investigations carried out the value of the noninvasive methods (routine-, three-minute-, 24-hour-, and load-ECG) for the diagnostics of latent ventricular disturbances of cardiac rhythm was controlled. 114 registrations of ventricular arrhythmias according to the Lown-classes with increasingly prolonged periods (1, 6, 12, and 24 hours) show that for the establishment of a ventricular extrasystolia of the classes IV and V a registrations lasting at least 24 hours is necessary. 1- and 3-minute-ECG are not suited for this. The load-ECG has a limited value in the diagnostics of the ventricular arrhythmias. In 155 persons by means of the transoesophageal diagnostic atrial stimulation additionally the sinus node recovery time and the sinuatrial conduction time were determined. In 33 patients of the group with sinus node syndrome the 24-hour-ECG was registered. The sensitivity for the sinuatrial conduction time is 75% and for the sinus node recovery time is 82%. Altogether the two investigation methods, the non-invasive electrophysiology and the long-term ECG, allowed to make a correct diagnosis in 94% of the patients with sinus node syndrome.

Circulating fibronectin in burned patients.

Plasma and serum fibronectin levels were determined in a serial study of 24 non-septic burned patients by means of the passive haemagglutination test and electroimmunoassay. The results showed a consistent pattern of observed changes: a decrease of circulating fibronectin in the initial (first 72 hours post-burn), and during post-operatyive period (usually by the end of the first week and during the second week post-injury). Fibronectin level decreases were more remarkable than total serum protein concentration changes following injury, as reflected by decrease of fibronectin/protein ratio.
