Influence of the soil bioavailability of radionuclides on the transfer of uranium and thorium to mushrooms.

The soil-mushroom transfer of thorium and uranium was analyzed in two ecologically similar but geographically separated Spanish ecosystems by means of the transfer factor, TF. Uranium TF values were in the range 0.043-0.49, and thorium TF values in the range 0.030-0.62. These values were similar to those of (90)Sr, (239+240)Pu, and (241)Am found previously in the same ecosystems. Given the low availability of uranium and thorium, the available transfer factors, ATF, were also determined. These were higher than the TF values by one order of magnitude for (234, 238)U, and by 2-3 orders of magnitude for (228, 230, 232)Th. The ATF value of thorium was similar to that of (137)Cs, and that of uranium similar to that of (40)K. Hebeloma cylindrosporum presented the highest uranium and thorium transfer factors, confirming this species as a good bioindicator of a soil's radioactive content.

Soil-fungi transfer coefficients: Importance of the location of mycelium in soil and of the differential availability of radionuclides in soil fractions.

Soil-fungus transfer coefficients are usually defined as the ratio between the content of the fruiting bodies and that of the soil. Since, however, the methodology of how to determine the soil content is not firmly established, there exist a variety of definitions in the literature. We analyzed the 137Cs, 90Sr, 40K, and 226Ra content of mushroom and soil samples from two pine-wood ecosystems in Spain. The location of the mycelium in the soil profiles of these ecosystems was determined by means of the ergosterol concentration. The results showed the mycelium to generally be localized in the surface layer of soil (0-5 cm). We also carried out a speciation procedure for this layer of soil to determine the different degrees of association of the radionuclides in the soil. The results led us to propose some variations to the traditional definition used in quantifying radionuclide transfer. With these modifications, we were able to analyze Cs-K competition in several species of mycorrhizal and saprophytic fungi.

Anthropometric measurements in both sides of the body in the assessment of nutritional status in prepubertal children.

OBJECTIVE: To analyze the impact of choosing the left or the right side of the body on the anthropometric measurements and derived nutritional indices, in prepubertal children. DESIGN: Cross-sectional pilot nutrition survey. SETTING: General prepubertal school-age population. SUBJECTS: One-hundred and sixty-four children (97 boys and 67 girls) aged 7-9 y. INTERVENTIONS: None. RESULTS: The agreement between anthropometric measurements in both sides of the body showed that in males and in females, suprailiac skinfold thickness and arm circumference were significantly higher in the left than in the right side of the body. The agreement between body composition assessed by anthropometric measurements in both sides of the body showed that only in males was arm muscle area significantly higher in the left than in the right side, and arm fat percentage was higher in the right than in the left side of the body. Total body fat percentage calculated from skinfold thickness did not show statistically significant differences when skinfolds were obtained in the both sides of the body, either in boys and in girls. CONCLUSIONS: Our results show that differences between the sides of the body were lower than the technical error of measurement of the anthropometric measurements obtained and seem not to be biologically significant in this age group. It is necessary to standardize the method of anthropometric assessment of the nutritional status in terms of body side.

Both morphological and molecular characters support speciation of South American siskins by sexual selection.

South American siskin radiation was studied by both mitochondrial cytochrome b (mt cyt b) DNA sequencing and homologous phenotypic characters; the latter were coded separately according to sex. Mixed phenetic and molecular (total evidence) dendrograms were constructed and the corresponding analyses suggest that speciation started in the South American siskin group with a north to south separation (Carduelis notata/C. barbata) along the Andean spine. A second split may have taken place around the Peruvian Andean mountains, corresponding to the present distribution pattern of C. olivacea. The most recent speciation events seem to have occurred in three sister species pairs: (i) C. xanthogastra/C. atrata, (ii) C. magellanica/C. yarrellii, (iii) C. cucullata/C. crassirostris. Accumulation of consistent characters in both morphological and molecular data at the basal nodes of the dendrograms indicate that speciation events occurred within a short period of time. Our data also suggest that speciation probably occurred by sexual selection through female mating choice in this radiation. Additionally, studies of variable amino acid residues in the mt cyt b molecule show that the three variable amino acids found are placed in the mitochondrial transmembrane region, which is also part of the hypervariable region in mammals. Each of the three amino acid changes occur in each of the three postulated evolutionary groups.

Phylogeography of crossbills, bullfinches, grosbeaks, and rosefinches.

Mitochondrial cytochrome b (cyt b) from 24 Carduelini species including crossbills, bullfinches, grosbeaks, rosefinches, and other related, but not conclusively classified species, was sequenced. These sequences were also compared with all the available sequences from the genera Carduelis, Serinus, and Passer. Phylogenetic analyses consistently gave the same groups of finches and the calculated divergence times suggest that speciation of the studied species occurred between 14 and 3 million years ago (Miocene-Pliocene), appearing before the Passer, Carduelis, and Serinus genera. Pleistocene glaciations may have been important in sub-speciation. Crossbills are integrated within the genus Carduelis, and within redpolls; the common crossbill shows subspeciation with Loxia japonica in the Pleistocene epoch. Pinicola enucleator groups together with bullfinches and is probably the ancestor of the group. Hawfinch is only distantly related to the studied groups, and might either represent an isolated genus or be related to the New World genus Hesperiphona. The grosbeak genera Eophona and Mycerobas are clearly sister groups, and species belonging to the former might have given rise to Mycerobas species. The isolated (in classification) Uragus sibiricus and Haematospiza sipahi are included within the genus Carpodacus (rosefinches); Carpodacus nipalensis is outside the genus Carpodacus in the molecular analyses and might be an isolated species or related to the genus Montifringilla.

The Old World sparrows (genus Passer) phylogeography and their relative abundance of nuclear mtDNA pseudogenes.

The phylogenetic relationships of genus Passer (Old World sparrows) have been studied with species covering their complete world living range. Mitochondrial (mt) cyt b genes and pseudogenes have been analyzed, the latter being strikingly abundant in genus Passer compared with other studied songbirds. The significance of these Passer pseudogenes is presently unclear. The mechanisms by which mt cyt b genes become pseudogenes after nuclear translocation are discussed together with their mode of evolution, i.e., transition/transversion mitochondrial ratio is decreased in the nucleus, as is the constraint for variability at the three codon positions. However, the skewed base composition according to codon position (in 1st position the percentage is very similar for the four bases, in 2nd position there are fewer percentage of A and G and more percentage of T, and in 3rd codon position fewer percentage of G and T and is very rich in A and C) is maintained in the translocated nuclear pseudogenes. Different nuclear internal mechanisms and/or selective pressures must exist for explaining this nuclear/mitochondrial differential DNA base evolutive variability. Also, the phylogenetic usefulness of pseudogenes for defining relationships between closely related lineages is stressed. The analyses suggest that the primitive genus Passer species comes from Africa, the Cape sparrow being the oldest: P. hispaniolensis italiae is more likely conspecific to P. domesticus than to P. hispaniolensis. Also, Passer species are not included within weavers or Estrildinae or Emberizinae, as previously suggested. European and American Emberizinae sparrows are closely related to each other and seem to be the earliest species that radiated among the studied songbirds (all in the Miocene Epoch).

Seasonal variations in radionuclide transfer in a Mediterranean grazing-land ecosystem.

We made quarterly determinations of the transfer coefficients and effective transfer coefficients for the radionuclides 137Cs, 90Sr, 40K, 226Ra, 228Ra and 228Th over a full annual cycle, in a Mediterranean grazing-land ecosystem. The input and output fluxes of the radionuclides between the different compartments of this ecosystem were quantified for the following processes: root uptake; variation in root and aerial biomass; pasture production; translocation; leaf fall; efflux due to grazing action; resuspension and subsequent aerial deposition of radionuclides. We observed there to be a marked seasonal variation for this type of ecosystem in both the transfer coefficients and the radionuclide fluxes, which impedes the soil-plant transfer being characterized on the basis of values that are constant with time.

Radiocaesium and radiostrontium uptake by fruit bodies of Pleurotus eryngii via mycelium, soil and aerial absorption.

There has been an extraordinary increase in interest concerning the transfer of radioactive contamination to the fruit bodies of fungi since the Chernobyl nuclear power plant accident. These investigations, however, have focused exclusively on field studies aimed fundamentally at quantifying the behaviour of the radiocaesium component of the contamination. The results have shown great variability. As a contribution towards this body of knowledge, we have made a comparative study of the temporal evolution of the transfer of 85Sr and 134Cs via three routes of radioactive contamination--from the mycelium, from the surface layer of the soil, and directly onto the caps of the fruit bodies--for the saprophyte species, Pleurotus eryngii, under controlled laboratory conditions. The results indicate that the last of the above three uptake routes is the most efficient, and that the temporal evolution of the transfer is closely related to the radionuclide and the radioactive contamination route being considered.

Recovery of chronic parkinsonian monkeys by autotransplants of carotid body cell aggregates into putamen.

We have studied the effect of unilateral autografts of carotid body cell aggregates into the putamen of MPTP-treated monkeys with chronic parkinsonism. Two to four weeks after transplantation, the monkeys initiated a progressive recovery of mobility with reduction of tremor and bradykinesia and restoration of fine motor abilities on the contralateral side. Apomorphine injections induced rotations toward the side of the transplant. Functional recovery was accompanied by the survival of tyrosine hydroxylase-positive (TH-positive) grafted glomus cells. A high density of TH-immunoreactive fibers was seen reinnervating broad regions of the ipsilateral putamen and caudate nucleus. The nongrafted, contralateral striatum remained deafferented. Intrastriatal autografting of carotid body tissue is a feasible technique with beneficial effects on parkinsonian monkeys; thus, this therapeutic approach could also be applied to treat patients with Parkinson's disease.

Omega-3 fatty acids in adipose tissue and risk of myocardial infarction: the EURAMIC study.

Omega-3 fatty acids have potential antiatherogenic, antithrombotic, and antiarrhythmic properties, but their role in coronary heart disease remains controversial. To evaluate the association of omega-3 fatty acids in adipose tissue with the risk of myocardial infarction in men, a case-control study was conducted in eight European countries and Israel. Cases (n=639) included patients with a first myocardial infarction admitted to coronary care units within 24 hours from the onset of symptoms. Controls (n=700) were selected to represent the populations originating the cases. Adipose tissue levels of fatty acids were determined by capillary gas chromatography. The mean (+/-SD) proportion of alpha-linolenic acid was 0.77% (+/-0.19) of fatty acids in cases and 0.80% (+/-0.19) of fatty acids in controls (P=0.01). The relative risk for the highest quintile of alpha-linolenic acid compared with the lowest was 0.42 (95% confidence interval [CI] 0.22 to 0.81, P-trend=0.02). After adjusting for classical risk factors, the relative risk for the highest quintile was 0.68 (95% CI 0.31 to 1.49, P-trend=0.38). The mean proportion of docosahexaenoic acid was 0.24% (+/-0.13) of fatty acids in cases and 0.25% (+/-0.13) of fatty acids in controls (P=0. 14), with no evidence of association with risk of myocardial infarction. In this large case-control study we could not detect a protective effect of docosahexaenoic acid on the risk of myocardial infarction. The protective effect of alpha-linolenic acid was attenuated after adjusting for classical risk factors (mainly smoking), but it deserves further research.

Multiple neuroanatomical tracing in primates.

The present report deals with a multiple tract-tracing procedure in non-human primates enabling the simultaneous visualization of retrogradely transported Fluoro-Gold (FG) and cholera toxin B subunit (CTB) in combination with anterogradely transported biotinylated dextran amine (BDA). Two issues have played key roles on the achievement of this reliable procedure: first, the recent development of a commercial antiserum against FG that allows us to convert the original fluorescent signal of this dye in a permanent precipitate via standard peroxidase-anti-peroxidase methods; second, the introduction of the novel peroxidase substrate Vector(R) VIP (V-VIP), resulting in a purple precipitate. The combination of these neuroanatomical tracers in one and the same histological section opens a possibility for the permanent visualization of the convergence of inputs from a particular brain area onto identified, two different subsets of projection cells of another area. Furthermore, this combination of three tracers emerges as a powerful technical tool for obtaining broad amounts of complementary data regarding the monkey brain connectivity, thus significantly reducing the number of animals needed to complete a particular study.

Subthalamotomy in parkinsonian monkeys. Behavioural and biochemical analysis.

Nineteen Macaca fascicularis monkeys were divided into four different groups: Group A (n = 3), control; Group B (n = 3), monkeys treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP); Group C (n = 8), animals treated with MPTP in which the subthalamic nucleus (STN) was unilaterally lesioned by kainic acid injection; in Group D (n = 5), the STN was lesioned prior to MPTP administration. Subthalamotomy resulted in a bilateral improvement of tremor, spontaneous activity, bradykinesia (evaluated by a manual motor test) and freezing in Group C. All these monkeys developed hemichorea contralateral to the lesion. The improvement was maintained and the hemichorea continued until death. The monkeys in group D showed severe hemiballism which persisted throughout MPTP administration and developed parkinsonian signs mainly on the side ipsilateral to the lesion. Analysis of the in situ hybridization of the mRNA coding for glutamic acid decarboxylase (GAD) of MPTP monkeys showed a significant increase in the mean density of silver grains over every labelled neuron in the globus pallidum lateralis (56.8% over control) as well as the globus pallidus medialis (GPM) (45.7% over control) and the substantia nigra reticulata (SNR) (35.8% over control). No significant change was observed in the thalamic nucleus reticularis. Subthalamotomy (Groups C and D) produced a significant reduction in mRNA GAD expression on the side of the lesion in the GPM and the SNR (34% and 42.3%, respectively) with respect to the ipsilateral (non-lesioned) side and also when compared with parkinsonian monkeys. These results confirm and expand, at the cellular level, the paramount role of STN hyperactivity in the pathophysiology of parkinsonism. The therapeutic consequences of these findings for surgical treatment of Parkinson's disease are discussed.

Functional interaction between dopamine D1 and D2 receptors in 'MPTP' monkeys.

We have studied the motor response induced by independent administration of 4 different doses of a dopamine D2 [(+)-PHNO] and a dopamine D1 (CY 208-243) receptor agonist in 5 MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) monkeys. Both drugs had similar antiparkinsonian effects and both elicited choreic dyskinesias. Simultaneous administration of (+)-PHNO [(+)-4-propyl-9-hydroxynaphthoxazine] and CY 208-243 [(-)4,6,6a,7,8,12b-hexahydro-7-methylindolo[4,3a-b]phenan thyxidine] did not result in modification of the dose-response curve induced by each dopamine receptor agonist given alone. Pretreatment with the dopamine D1 receptor antagonist SCH 23390 (0.8 mg/kg) and the dopamine D2 receptor antagonist sulpiride (60 mg/kg) reduced the magnitude and the duration of the motor response induced by (+)-PHNO and CY 208-243, respectively, but did not modify the intensity and characteristics of choreic dyskinesias. These results demonstrate that the motor effects and the dyskinesias cannot be dissociated by selective dopamine D1 and D2 receptor stimulation. It appears that stimulation of dopamine D1 and D2 receptors by endogenous dopamine is required to obtain the full motor response induced by selective dopamine receptor agonists as demonstrated by the reduction of the motor improvement found after pretreatment with SCH 23390 and sulpiride.

The AMPA receptor antagonist NBQX does not alter the motor response induced by selective dopamine agonists in MPTP-treated monkeys.

We studied the motor response induced by the administration of the AMPA receptor antagonist, NBQX (2,3-dihydroxy-6-nitro-7-sulfamoylbenzo[f]quinoxaline(1H,4H)dione sodium salt), in two parkinsonian monkeys and its interaction with the selective D-2 dopamine receptor agonist, (+)-PHNO [(+)-4-propyl-9-hydroxynaphthoxazine], and the partial dopamine D-1 receptor agonist, CY 208-243 [(-)-4,6,6a,7,8,12b-hexahydro-7-methyl-indolo[4,3a-b]phen anthyxidine]. NBQX treatment did not reverse the parkinsonism or change the responses previously induced by dopamine agonists. These results do not support an antiparkinsonian effect of the AMPA receptor antagonist, NBQX. In addition they indicate that there is no functional interaction between AMPA and dopamine receptors in monkeys with a substantia nigra lesion.

[Warning systems: a priority in epidemiological surveillance].

One of the functions of public health is to reduce the impact of emergency situations; hence the need to establish an early warning and response system. Community reactions to emerging and reemerging diseases, exposure to environmental risks, and potential terrorist acts are the factors that have that have mainly contributed to the instauration of warning systems based on mathematical models. Information is obtained from diverse sources (for example, laboratory data, sentinel physicians, or visits to particular websites). Once the data are gathered, and the situation provoking the alert is detected, control measures to reduce risks and minimize damage are taken at the earliest.

[Adrenal transplantation and Parkinson's disease].

Although substantial therapeutic advances have been developed during the last decade, L-dopa remains as the most successful treatment for Parkinson's disease. However, about 80% of the patients show motor and psychiatric complications after several years of levodopa treatment. Thus, new therapeutic approaches have been undertaken during the last decade in order obtain a better control of the motor symptoms. Adrenal medullar grafts into caudate nucleus and/or putamen have been widely studied as a new therapeutic strategy. They have been undertaken in more than 300 parkinsonian patients and the general finding is that adrenal medullar grafts induce a significant improvement of motor symptoms which remain 1 year after the surgery. The mechanisms by which they can elicit a motor improvement are not well understood since chromaffin cells do not survive in the host brain after transplantation as demonstrated by PET scan studies. The present work summarizes the results found by different authors as well as the mechanisms involved in the motor improvement observed in these patients.
