Building blocks, cohort genetic algorithms, and hyperplane-defined functions.

Building blocks are a ubiquitous feature at all levels of human understanding, froin perception through science and innovation. Genetic algorithms are designed to exploit this prevalence. A new, more robust class of genetic algorithms, cohort genetic algorithms (cGA's), provides substantial advantages in exploring search spaces for building blocks while exploiting building blocks already found. To test these capabilities, a new, general class of test functions, the hyperplane-defined functions (hdf's), has been designed. Hdf's offer the means of tracing the origin of each advance in performance; at the same time hdf's are resistant to reverse engineering, so that algorithms cannot be designed to take advantage of the characteristics of particular examples.

[Phase-II-study with cis-diamminedichloroplatinum (II) in the treatment of advanced malignant lymphomas].

29 patients with advanced malignant lymphoma were included in a phase-II trial conducted by Cancer and Acute Leukemia Group B. The patients received cis-diamminedichloroplatinum (DDP) in a dose of 70 mg/m2 every three weeks. In this still ongoing study, 23 patients are already evaluable. Out of 23 cases partial remission was observed in 6. The main toxicity was profuse vomiting. Myelosuppression and nephrotoxicity were both manageable. Based on these preliminary data, the incorporation of DDP in combination chemotherapy for malignant lymphoma appears to be warranted.

Comparative study of cytosine arabinoside therapy alone and combined with thioguanine, mercaptopurine, or daunorubicin in acute myelocytic leukemia.

Three hundred twenty-six patients with acute myelocytic leukemia were randomly and prospectively assigned to four therapeutic regimens: cytosine arabinoside either alone or in combination with daunorubicin, 6-mercaptopurine, or 6-thioguanine. The results in 231 qualified previously untreated patients were analyzed. The combination treatments produced a significantly greater frequency of complete or partial remission than single drug therapy. Treatment with cytosine arabinoside and thioguanine led to 48% age-adjusted complete and partial responses. The median sur survival from diagnosis of all 66 evaluable patients treated with these two drugs was 18 weeks, while the median survival for those who responded to this combination was 15 months.
