Transfer of plasmids from Escherichia coli and Pseudomonas aeruginosa to methylotrophic bacteria and their detection in the new hosts.

Several plasmids of incompatibility group P were transferred from Escherichia coli and Pseudomonas aeruginosa strains to Methylophilus methylotrophus and two other methylotrophs to test their recipient ability. The presence of plasmids in transconjugants was confirmed by electrophoretic analysis. Optimal conditions for detection of plasmid DNA in the strains tested based on alkaline lysis of cells at elevated temperature were established. Special behaviour of plasmids carrying the Mu phage in methylotrophic hosts is described.

Mini-Mu transposition of bacterial genes on the transmissible plasmid.

Using the pRM30 plasmid, an Aps deletion derivative of broad host range plasmid RP4 with integrated new miniMu 5 (11 kb), we followed the transfer of Escherichia coli chromosomal genes to the recipient strain. The miniMu 5-mediated transposition of chromosomal genes occurs onto the plasmid with integrated miniMu 5 rather than onto the "recipient" plasmid pNH602. The plasmid DNA in recipient cells was detected by electrophoresis. One of the acquired hybrid plasmids pTB2 was analyzed genetically and by restriction endodeoxyribonuclease digestion. A structure consisting of miniMu-chromosomal segment-miniMu as a product of Mu-mediated transposition was detected.

[The final picture of wound healing in CO2 laser scalpel surgery].

The terminal reparative phase was studied in wounds caused by carbon dioxide laser on rat skin, auricle, striated muscle, liver and spleen. Comparative histology was especially concerned with resorptive granulomas around carbonized tissue remnants. Presence of carbonized residua was connected with a more extensive fibrosis and seemed to lengthen the healing process.

Isolation and characterization of mutants of the RP4 plasmid coding for increased resistance to ampicillin.

E. coli strain J53(RP4) was mutagenized with ethyl methanesulfonate and N-methyl-N'-nitro-N-nitrosoguanidine. Clones showing a two-to threefold increase in resistance to ampicillin were produced. This increase was not due to an increased number of RP4 copies per chromosome. The level of penicillinase activity was twice higher in comparison with the parental strain. No detectable changes were found in the region coding for the resistance to ampicillin on the plasmid by restriction analysis.
