Pentobarbital in tobacco.

The spectrometric analysis of extracts from tobacco and tobacco smoke revealed the presence of pentobarbital in the analyzed substances. Tobacco samples and tobacco smoke were extracted with chloroform, determinations were performed with the Perkin-Elmer Autosystem XL system, on a Turbo Mass spectrometer. Subject to analysis were 4 cigarette brands manufactured in Poland and raw, unprocessed tobacco. The presence of pentobarbital in the analyzed samples was confirmed by the analysis of the mass spectrum of the substance, as well as by comparison of retention time with standard of pentobarbital. The determined pentobarbital concentrations in tobacco amounted to 3-6 microg/cigarette, and in tobacco smoke they were approximately 45% lower. In case of tobacco extracts it can with high probability be excluded that pentobarbital is synthesized during chromatographical analysis. The presence of pentobarbital in tobacco is thus beyond question.

IL-15 in the culture supernatants of PMN and PBMC and the serum of patients with Lyme disease.

PURPOSE: The aim of this study was to estimate the production of interleukin-15 (IL-15) by neutrophils (PMNs) and peripheral blood mononuclear cells (PBMC) confronted with the serum levels of IL-15 in patients with Lyme disease. MATERIAL AND METHODS: PMN and PBMC were isolated from heparinized whole blood of patients. The cells were incubated for 18 hs at 37 degrees C in a humidified incubator with 5% CO2. After 18 hs incubation, supernatant was removed and assessed for IL-15 using ELISA kits. RESULTS: The results obtained showed significant increase in the ability of patient's PMNs and peripheral blood mononuclear cells (PBMC) to release IL-15. Although PBMC produced higher concentrations of IL-15 than PMN, the quantitative dominance of PMN in the peripheral blood suggest a significant role for these cells in the defense reactions controlled by this cytokine. Similar changes in the secretion of IL-15 by PMN and PBMC in patient group may be caused by the same regulatory mechanisms which influence the functional abnormalities of the cells examined. CONCLUSIONS: A change in the ability of PMN and PBMC to release IL-15 may have various implications for the cellular and humoral response to the Borrelia burgdorferi (B.b.) infection in patients with Lyme disease.

Priming effects of GM-CSF, IFN-gamma and TNF-alpha on human neutrophil inflammatory cytokine production.

Identifying and evaluating the priming agents for cytokine release by neutrophils might be helpful in controlling the innate immune response of the host. In the present study we examined the role of granulocyte/macrophage colony-stimulating factor (GM-CSF), interferon-gamma (IFN-gamma) and tumour necrosis factor-alpha (TNF-alpha) as priming agents for interleukin (IL)-1beta, IL-6 and TNF-alpha production by stimulated neutrophils from control subjects and malignant melanoma patients. When the cells from controls and patients were preincubated with primer agents, opsonized zymosan-stimulated inflammatory cytokine production was enhanced. The major neutrophil-priming factor for IL-6 secretion by polymorphonuclear leukocytes (PMNs) in the control and patient groups was TNF-alpha. However, GM-CSF and IFN-gamma are also significant primers. GM-CSF priming was critical for the release of TNF-alpha from PMNs in control and melanoma patients. The ability of GM-CSF, IFN-gamma and TNF-alpha to serve as effective priming agents for inflammatory mediator production by PMNs revealed a new role for these cytokines in the innate immune response of the melanoma-bearing host.

TNF-alpha, IL-6 and their soluble receptor serum levels and secretion by neutrophils in cancer patients.

Simultaneous evaluation of cytokines and their soluble receptor production and the serum levels can be helpful in understanding the local and systemic immune response of a tumor-bearing host. In the present study we examined serum levels of TNF-alpha, IL-6 and their soluble receptors: sTNFRp55, sTNFRp75 and sIL-6R confronted with their production by the polymorphonuclear neutrophils (PMN) from cancer patients. Examinations were carried out in patients with adenocarcinoma breast cancer and squamous cell carcinoma of the oral cavity and related to the clinical course and to different phases of therapy. Secretion of IL-6, sTNFRp55 and sTNFRp75 by PMN appeared to be dependent on tumor type, clinical progression of disease as well as on therapy, suggesting a significant role of these cells at different phases of the immune response to cancer associated with these mediators. Changes in values of TNF-alpha, IL-6 and their soluble receptors in sera of both cancer groups, dependent on tumor type, clinical progression and cancer therapy, could have a diagnostic and prognostic role in cancer disease.

[N-nitrosodimethylamine--toxicologic significance].

In this paper we have presented data on an environmental exposure to N-nitrosodimethylamine (NDMA) and factors which favour endogenous biosynthesis of this compound. The factors influencing metabolism and toxicity as well as health effect of exposure have been reported.

[Treatment of corneal complications after cataract surgery with soft contact lenses].

PURPOSE: To evaluate results of therapeutic soft contact lenses in treatment of corneal complication after cataract surgery. MATERIAL NAD METHODS: We examined 25 patients (25 eyes) with cataract operated on in the years 1990-1994, who developed corneal complications. All patients were treated with use of soft contact lenses and the effect on healing a cornea was evaluated. We used Bausch & Lomb and Cooper Vision lenses. We qualified for treatment such cases as: bullous keratopathy, marginal infiltration erosion, central ulceration, complicated with perforation and protracted ulceration with no result after pharmacological therapy. Contact lenses were applied for period from 10 days to 6 months. During follow up examinations we evaluated time of healing a cornea, its anatomical condition and function. RESULTS: The best therapeutic result was achieved in 12 cases such as erosions, central ulceration, marginal infiltration, in 7 cases temporary improvement appeared, in 6 no result was observed. Two last groups applied patients with bullous keratopathy. CONCLUSION: Soft contact lenses are valuable methods of treating some corneal disorders, in other cases they may be used during a preparation period before corneal transplantation. The main function of lenses is to protect diseased tissue and help it return to its anatomical and physiological functional state. At the same time they are meant to be the best therapeutic repository for medications.

[Late effects after anterior intraocular lens implantation].

MATERIAL AND METHODS: During ten years, 890 operations of cataract with anterior chamber intraocular lens implantation were performed. TWO GROUPS OF EYES WERE OBSERVED: Eyes after planned intracapsular cataract surgery and eyes in which intraoperative complications occurred making impossible to apply posterior IOL. PURPOSE: To evaluate late complications and functional status of the eye. RESULTS: Our results indicate that anterior chamber intraocular lens implantation yields in many cases good visual acuity and, although not free of complications, is a safe procedure.

[Evaluation of the usefulness of extrelut(R) columns obtained from MERCK for isolation of N-nitrosodimethylamine (NDMA) in the water phase].

The aim of the study was to evaluate the method of N-nitrosodimethylamine (NDMA) isolation from the water phase using Extrelut columns (Merck). Results were compared with data obtained from isolation of NDMA by classic distillation method. The estimation was carried out on fortificated water and blood samples. Extraction Extrelut columns are found very useful in isolation of NDMA both in water and blood samples. Their advantage over the method of water vapour distillation is also indicated.

[Effect of n-nitrosodimethylamine (NDMA) on activity of selected enzymes in blood serum of the rat].

Changes in the activity of GOT, GPT, AP and GGTP in blood of rats intoxicated by low doses of N-Nitrosodimethylamine (NDMA) are presented. A significant increase in the activity of GOT and GPT was found after single doses (20 micrograms/kg b.w.) of NDMA as well as a significant increase in the activity of GOT, GPT, AP and GGTP after a prolonged intoxication per os by doses of 20 micrograms/dm3 given in drinking water.

[Intraocular implants in the posterior chamber after intraoperative posterior capsule tear].

PURPOSE: To present the results of cataract surgery with IOLs implantation complicated by posterior capsule tear. MATERIAL AND METHODS: 38 eyes of 38 patients with cataract, operated on between August 1993 and May 1994 were examined. In all cases posterior capsule tear occurred during surgery but the IOLs were supported by the residual capsule or, when the tear was large, by the residue of the anterior capsule with haptics being placed in the ciliary sulcus. RESULTS: Useful visual acuity was achieved in 78% of the eyes. The main complications observed in post-operative period were increase of intraocular pressure in 11 eyes, dislocation of IOL in 4 and dyscoria in 24 eyes. CONCLUSION: The applied surgical technique seems to be very useful in cases of posterior capsule tear but in each case the surgeon must decide what is the best for the eye: anterior or posterior chamber IOL.

[Results of surgery after intraocular disc lens implantation].

PURPOSE: To evaluate the results of Medi Contur IOLs implantation, which are a slightly modified Galand disc lens. MATERIAL AND METHODS: In the years 1994-1995, 31 eyes were operated on using modified method with horizontal, linear anterior capsule incision and intercapsular disc lens implantation. The follow-up was 6 months. RESULTS: Mean visual acuity was 0.7-0.8. There were no complications different from those occurring in other types of IOLs. The residual anterior capsule has no negative influence on visual acuity. The shape, size and localization of the IOL in physiological place, between anterior and posterior capsule, cause the good position and stability of the lens. The risk of development of the posterior capsule folds and opacity was decreased on account of a uniform capsular tension and contact of the IOL with the posterior capsule. CONCLUSION: The presented method of cataract extraction and IOL implantation, with all manipulations in intercapsular space is safer in comparison with others methods and residual anterior capsule decreases the risk of lens dislocation.

[Nitrosamines].

The paper reviews data on the property and mechanism of toxicologic effects of nitrosamines. Particular attention is turned to metabolism and carcinogenesis of these compounds in view of their concentration in human environment.

[Intraocular pressure after cataract extraction with implantation of an intraocular lens].

The subject of our investigations was the behaviour of the IOP after extracapsular and intracapsular operations with implantation of artificial lenses as well into the anterior as into posterior chamber. The investigations were performed on the material of 395 eyes operated in the years 1985-1988. It was established that after these operations glaucoma is a very rare complication.

[Retinal detachment in pseudophakia].

Retinal detachment in an aphakic eye is a grave therapeutical problem. There exists some fears that cataract extraction with implantation of an artificial lens may increase the number of these complications. Three hundred and seventy cases of cataract extraction with implantation of an artificial lens performed in our department were analysed from the point of view of the incidence of retinal detachment. The cataract was extracted extra- and intracapsularly and the artificial lens was implanted in the anterior as well as in the posterior chamber. During the period of observation--4 months to 3 years--it was established that the number of the retinal detachment in pseudophakia was not larger than in aphakic eyes without implant.
