Bombesin plasma levels in patients with benign and malignant breast disease.

Some reports suggest a role for bombesin-like peptides in the pathology of breast tumors. Bombesin and related gastrin-releasing peptides have been shown to influence the inositol phospholipid signalling pathway and stimulate growth of some cells, including some human breast cancer cell lines. We measured the plasma concentration of bombesin in 23 breast cancer patients, 32 patients suffering from benign breast disease and in 21 healthy controls. The bombesin concentration in plasma taken from the thoracodorsal vein, in the vicinity of the tumor in breast cancer patients was higher than that in the peripheral circulation (mean +/- SEM, 91.3 +/- 54.3 vs. 40.9 +/- 27.4 pg/ml; p < 0.05). Bombesin concentrations in the cubital vein in breast cancer patients and in those with benign breast disease (61.7 +/- 49.3 pg/ml) was significantly higher than that in the control group (23.7 +/- 5.06 pg/ml; p < 0.05). Our findings suggest storage or synthesis of bombesin-like peptides within the affected breast and may confirm the role of these peptides in the growth of breast tumors.

[Results of treatment for acromegaly with long-acting bromocriptine (Parlodel LAR)].

Long-acting bromocriptine (Parlodel LAR) was used for treatment of 25 patients with acromegaly during the period of 3 to 24 months. Even after the first intramuscular injection of 50 milligrams of the drug a decrease in growth hormone (GH) concentration by at least 50% of the initial values was observed in 28% of patients and an improvement in a sense of well-being in 44%. After 6 months of administration of 100 mg of Parlodel LAR at intervals of 28 days a decrease in GH level by at least 50% was observed in a larger percentage of patients (36.8%), and in 10.5% of them there was a fall of GH concentration to below 10 microU/ml. Side effects, like nausea, vomiting and orthostatic hypotony, appeared within several hours after the injection of Parlodel and lasted in most cases up to 24 hours. After consecutive injections of the drug the side effects were of lesser intensity or completely disappeared. The results obtained allow to conclude that Parlodel LAR is an effective drug in some cases of acromegaly. In most patients the therapeutic effect can be seen after the first injection, but in some cases it appears only after several months of treatment.

[Vitex agnus castus extract in the treatment of luteal phase defects due to latent hyperprolactinemia. Results of a randomized placebo-controlled double-blind study].

The efficacy of a Vitex agnus castus preparation (Strotan capsules) was investigated in a randomized double blind study vs. placebo. This clinical study involved 52 women with luteal phase defects due to latent hyperprolactinaemia. The daily dose was one capsule (20 mg) Vitex agnus castus preparation and placebo, respectively. Aim of the study was to prove whether the elevated pituitary prolactin reserve can be reduced and deficits in luteal phase length and luteal phase progesterone synthesis be normalized. Blood for hormonal analysis was taken at days 5-8 and day 20 of the menstrual cycle before and after three month of therapy. Latent hyperprolactinaemia was analysed by monitoring the prolactin release 15 and 30 min after i.v. injection of 200 micrograms TRH. 37 complete case reports (placebo: n = 20, verum: n = 17) after 3 month of therapy were statistically evaluated. The prolactin release was reduced after 3 months, shortened luteal phases were normalised and deficits in the luteal progesterone synthesis were eliminated. These changes were significant and occurred only in the verum group. All other hormonal parameters did not change with the exception of 17 beta-estradiol which rouse up in the luteal phase in patients receiving verum. Side effects were not seen, two women treated with the Vitex agnus castus preparation got pregnant. The tested preparation is thought to be an efficient medication in the treatment of luteal phase defects due to latent hyperprolactinaemia.

Granulocyte pyruvate kinase in psoriasis vulgaris and psoriasis arthropathica.

The activity of granulocyte pyruvate kinase in 46 patients with psoriasis vulgaris and eight patients with psoriasis arthropathica was investigated. It was found that in both groups of patients the activity of pyruvate kinase was significantly higher than in healthy individuals. This suggests that the rate of glycolysis in granulocytes of patients with psoriasis is markedly increased. The activity of granulocyte pyruvate kinase in 11 psoriatic patients with actively spreading lesions was compared with the activity of this enzyme when the patients were in remission. The activity of the enzyme was significantly increased during the active stage of the disease and returned to normal during remission.

An approach to the elucidation of the quaternary structure role in the activity of pyruvate kinase studies on the immobilized enzyme.

1. Studies on pyruvate kinase immobilized via -S-S- linkages showed that single subunits of this tetrameric enzyme are inactive. 2. Pyruvate kinase was coupled with different preparations of CNBr-activated Sepharose via one and more than one bond, and after removal of non-covalently bound subunits by denaturant treatment the subunit derivatives were obtained composed of different pyruvate kinase species. 3. Heat inactivation studies of immobilized enzyme derivatives suggested that not only tetramers of pyruvate kinase are active. 4. This idea is further supported by experiments with 2 M urea dissociation of immobilized tetrameric enzyme carried out in the presence of Mg2+ ions.
