[The effect of a low-protein diet on the epithelium of nephron segments in sheep].

Changes in the epithelium thickness of proximal and distal convoluted tubule of cortex, thin descending limb of Henle loop from inner stripe of outer medulla, thick ascending limb from outer stripe of outer medulla and of collecting duct from inner medulla were investigated in young growing sheep fed a low protein diet (LP). Sheep on LP-diet were given a daily ration with 5.73 g of nitrogen and 6.13 MJ of digestible energy (DE) while the daily intake of control group was 19.51 g of N and 12.29 MJ of DE. Both groups of animals were fed these diets at least 6 weeks before collection of kidneys. Morphometric analysis with digitizing tablet showed that intake of LP-diet resulted in significant reduction of epithelium thickness in both distal convoluted tubule and thick ascending limb while the epithelium of collecting duct was found to be thicker. No changes of the epithelium dimensions were determined in proximal tubule and in thin descending limb of Henle loop. Presented results point to the morphological expression of the adaptation of sheep kidneys to a low dietary protein intake which is associated with the increased renal reabsorption of urea.

[The effect of hypodynamia on striated muscles in Japanese quail].

The effect of hypodynamia during 9, 30, 60 and 90 days on the striated muscle m. gastrocnemius in Japanese quails was studied. We have found different developed dystrophic changes of mitochondria, myofibrils and sarcotubular system. Changes of the studied tissue caused by hypodynamia in the first two groups (9 and 30 days hypodynamy) are the reaction to stress and developing situation. Gradually Japanese quails have come up to the influence of the hypodynamia (60 days hypodynamia). In the last experimental group deposit changes were observed (90 days hypodynamia). In comparison with control group we have found perturbations considered as non-specific, short-term and reversible.

[The effect of weightlessness on embryonic development of striated muscles in Japanese quail].

In the conditions of gravitationless state, the development of skeletal muscles and myocardium was identical in experimental, synchronous and laboratory control. There were not larger differences between the investigated groups of animals. Myofibrils were in the stage of myofibrillogenesis. They were mostly disarrayed, but they already formed sarcomeres, without marked boundaries between I and A zones. Sarcoplasm contained large amounts of ribosomes and polyribosomes. Mitochondria were intensively splitting, and they contained small numbers of mitochondrial particles. Osmiophilic bodies and granules were detected frequently. Sarcoplasmic reticulum and Golgi apparatus were well developed. Our findings are in agreement with the results published in papers dealing with the embryonal development of striated muscles.

Mutagenicity studies on paracetamol in human volunteers. III. Cytokinesis block micronucleus method.

As paracetamol (PC) affected the frequency of chromosome aberrations and unscheduled DNA synthesis in human volunteers, the genotoxic effects of PC were studied in a group of 12 healthy volunteers (9 females; 3 males, aged 37.8 +/- 8.7 years) using the cytokinesis block micronucleus method in human peripheral lymphocytes. As a positive control a group of elderly people was used, 20 females and 10 males, aged 79.9 +/- 9.5 years. PC was administered orally 3 times in a dose of 1000 mg during 8 h. Blood samples were taken at intervals of 0, 24, 72 and 168 h after the first dose of PC. Cytochalasin B was added to the cultures 44 h after the beginning of the 72-h cultivation at a concentration of 3.0 micrograms/ml. The frequency of cells with micronuclei in the group of volunteers was not significantly increased after PC administration. Using the cytokinesis block micronucleus method, the frequency of micronuclei was stable and the interindividual variability was low. The application of the micronucleus technique in genetic monitoring, e.g., for the occupational exposure to mutagens, is questioned.

Mutagenic activity of paracetamol. A study conducted on volunteers.

The mutagenic activity of paracetamol (PC) was studied on a group of healthy persons (3 men, 8 women) after the simultaneous administration of 3 X 1000 mg of PC and an identical dose of PC + 1000 mg of ascorbic acid within a period of 8 hours. Blood sample tests were made at intervals of 0, 24, 72 and 168 hours. A cytogenetic analysis showed that in 24 hours PC had increased the aberrant cell frequency to 2.77 +/- 0.37 per cent as distinct from 1.68 +/- 0.30 per cent (p less than 0.05) prior to the drug. In 72 hours, the rate of buccal mucosa micronuclei had risen to 0.38 +/- 0.07% against 0.19 +/- 0.06% prior to PC (p less than 0.01). Unscheduled DNA synthesis in peripheral lymphocytes had dropped to T/C = 2.06 +/- 0.54 (p less than 0.01) against 3.16 +/- 0.84 before PC. The lipid peroxidation level in the plasma remained unchanged. Ascorbic acid administered simultaneously with PC had no effect on the changes observed. A further follow-up of other PC side effects is recommended.

[The micronucleus test in human peripheral lymphocytes].

A new methodology is reported determining the cytogenetic damage in stimulated human peripheral lymphocytes. The methodology is based on the investigation of the frequency of cells with micronuclei utilizing cytochalasin B. The spontaneous frequency of cells with micronuclei in the group of healthy volunteers (9 females and 3 males) of the mean age of 37.8 years was 18.42 micronuclei/1000 cells. Furthermore, the action of the analgesic agent paracetamol on the frequency of cells with micronuclei was examined; after the dose of 3 X 1000 mg of paracetamol administered within 8 h no increase in the occurrence of micronuclei within 1 week was found. The advantage of the use of the test for the determination of micronuclei with cytochalasin B in the testing of new drugs are discussed.

[Mutagenic activity of paracetamol. A study in volunteers].

Mutagenic activity of paracetamol (PC) was studied in a group of healthy persons (3 men, 8 women) after a simultaneous administration of 3 x 1000 mg of PC and of identical dose of PC + 1000 mg of ascorbic acid within an 8 hr. period. Blood sample tests were made at intervals 0, 24, 72 and 168 hrs. Cytogenetic analysis showed that in 24 hrs. the PC increases the aberrant cell frequency to the value of 2.77 +/- 0.37 percentage vs. 1.68 +/- 0.30% (p less than 0.05) prior to the administration of the drug. In 72 hrs., the occurrence of micronucleoli in the buccal mucosa increased to 0.38 +/- 0.07% vs. 0.19 +/- 0.06% prior to PC administration (p less than 0.01). Unscheduled DNA synthesis in peripheral lymphocytes decreased to T/C = 2.06 +/- 0.54 (p less than 0.01) compared to the value of T/C = 3.16 +/- 0.84 before PC administration. The lipid peroxidation level in the plasma was unchanged. The ascorbic acid administered simultaneously with the PC had no effect on the changes observed. The authors recommend a further follow-up of other side PC effects.

[The micronuclei detection test in stimulated human peripheral lymphocytes using cytochalasin B].

The authors introduced a technique for assessment of cells with micronuclei in stimulated human peripheral lymphocytes using cytochalasin B (CB technique). The spontaneous level of cells with micronuclei was assessed in an ageing population group. The group comprised fifteen subjects (12 women and 3 men), mean age 79.7 +/- 6.7 years. The spontaneous level of cells with micronuclei was 33.7 per 1000 evaluated cells, i.e. 3.37%. The authors consider the possibility of introducing the test for assessment of micronuclei in human peripheral lymphocytes using cytochalasin B as an alternative test in the cytogenetic analysis of chromosomal aberrations in human peripheral lymphocytes.

Mutagenicity studies on paracetamol in human volunteers. II. Unscheduled DNA synthesis and micronucleus test.

The possible genotoxic effect of paracetamol (PC) was studied in a group of 11 healthy volunteers. PC was administered in the form of tablets 3 x 1000 mg in the course of 8 h. Blood samples and buccal mucosa cells were taken 0, 24, 72 and 168 h after the first administration of the drug. Each blood sample was used for the termination of the unscheduled DNA synthesis (UDS) in peripheral lymphocytes and ascorbemia in plasma. Buccal mucosa cells were analysed for micronuclei. After PC administration the level of UDS induced by MNNG was decreased to T/C = 4.11 +/- 0.56 after 24 h vs. T/C = 5.02 +/- 0.47 (p less than 0.01) at 0 h. The frequency of micronucleated cells in the buccal mucosa was increased after 72 h to 0.38 +/- 0.07% vs. 0.19 +/- 0.06% (p less than 0.01) before PC administration. If PC was administered simultaneously with ascorbic acid (AA), also in a dose of 3 X 1000 mg, a decreased level of UDS was observed after 24, 72 and 168 h and the increased number of micronuclei was qualitatively the same as the PC alone: 0.38 +/- 0.09% after 72 h vs. 0.20 +/- 0.05% at 0 h AA did not decrease the genotoxic effect of PC, but prolonged the influence of PC on UDS.

[Study of the submicroscopic structure of striated muscles in Japanese quail under conditions of stress].

An electronmicroscopic study of striated muscles was made in Japanese quail stressed by hypokinesis. During 15-day hypokinesis influencing m. gastrocnemius, small pale vacuoles were found out on some mitochondria. The process of alterations went on, and in the 15-day hypokinetic animals, alterations in myofibrils were observed besides the alterations in mitochondria. They manifested themselves by indistinct lines of actin and myosin fibers and by an uneven course of Z-lines. The alterations of m. gastrocnemius mitochondria in Japanese quail during hypokinesis were probably caused by energy insufficiency and subsequent hydration of the organella. Nearly all components of the muscular tissue were impaired by the 30-day hypokinesis.

[The effect of a protein-free diet on the submicroscopic structure of striated muscles in Japanese quail].

The effect of protein-free diet on the striated muscle m. gastrocnemius of Japanese quail was studied in three test groups and the data were compared with a control group. Each test group included three birds. The first group was fed protein-free diet for three days, the second group for six days and the third for nine days. Egg albumin was absent in the protein-free diet. The remaining components (maize starch, germ oil, cellulose, mineral and vitamin premix) were represented by the same percentage in both diets. In the second group of birds given protein-free diet for six days, doubled Z-lines were found. No such alterations were found in the remaining groups in comparison with the controls. Since doubled Z-lines occurred only in the group given the protein-free diet for six days, further study is needed to provide evidence indicating that the doubling of Z-line is due to this factor.

[Abnormalities of the sperm neck of poultry].

Defects in the structure of the middle piece of poultry spermatozoon were studied by the electron-microscopic method. The defects included loose jointing of the head and falgellum owing to the shallow invagination fossa of the nucleus, and structural and mainly shape alterations of the proximal centriole. These defects existed in spermatozoa with a normal shape of nucleus as well as in those with rounded or otherwise irregularly shaped nuclei.

[Electron microscopic studies of striated muscles of the Japanese quail (Coturnix coturnix japonica) obtained from eggs centrifuged during incubation].

Reported in this paper are results obtained from investigations into the effect of hypergravitation on embryogenesis of striped muscles in Japanese quail. Specimens of striped muscles were excised from the birds after hatching and used in the study. The eggs had been centrifuged on zero day as well as on the first and 15th days of incubation. The muscle fibrils were found to proliferate in all groups, that is the control group, the group with centrifugation on zero day, and that with centrifugation on the first and 15th days. The process thus is characterised as physiological, known to occur prenatally and postnatally in developing individuals.
