T cell epitopes of human myelin oligodendrocyte glycoprotein identified in HLA-DR4 (DRB1*0401) transgenic mice are encephalitogenic and are presented by human B cells.

Myelin oligodendrocyte glycoprotein (MOG) is an Ag present in the myelin sheath of the CNS thought to be targeted by the autoimmune T cell response in multiple sclerosis (MS). In this study, we have for the first time characterized the T cell epitopes of human MOG restricted by HLA-DR4 (DRB1*0401), an MHC class II allele associated with MS in a subpopulation of patients. Using MHC binding algorithms, we have predicted MOG peptide binding to HLA-DR4 (DRB1*0401) and subsequently defined the in vivo T cell reactivity to overlapping MOG peptides by testing HLA-DR4 (DRB1*0401) transgenic mice immunized with recombinant human (rh)MOG. The data indicated that MOG peptide 97-108 (core 99-107, FFRDHSYQE) was the immunodominant HLA-DR4-restricted T cell epitope in vivo. This peptide has a high in vitro binding affinity for HLA-DR4 (DRB1*0401) and upon immunization induced severe experimental autoimmune encephalomyelitis in the HLA-DR4 transgenic mice. Interestingly, the same peptide was presented by human B cells expressing HLA-DR4 (DRB1*0401), suggesting a role for the identified MOG epitopes in the pathogenesis of human MS.

[Cystic pancreas adenoma. On the clinical relevance of histologic typing].

Although pancreatic cystadenomas are rare neoplasms, they are found today in rising frequencies due to improvement of diagnostic tools. There exist two types of cystadenoma: microcystic serous cystadenoma and mucinous cystadenoma. Usually, histological distinction from cystadenocarcinoma is readily made, but it may be difficult in the case of the mucinous variant. This subtype is supposed to be potentially malignant, whereas microcystic serous cystadenoma is always benign. In order to elucidate the characteristics of both variants, 5 own cases are reported in this article.

Comprehension and production of the /-ez/ plural allomorph.

Children's knowledge of morphological elements has traditionally been tested using Berko-type production tasks. Failure to respond correctly in this task may reflect production constraints independent of underlying linguistic knowledge. Accordingly, a detailed comparison of the comprehension of one morphological unit, the /-ez/ plural allomorph, was investigated. Two groups of subjects were tested; (1) those who produced this allomorph correctly and (2) those who failed to produce it. For the subjects as a group, the results indicated that children who could produce the /-ez/ allomorph in nonsense word contexts performed significantly better than children who were unable to produce this allomorph. An analysis of individual performance indicated that almost all of the subjects who produced the /-ez/ allomorph correctly reliably comprehended it. A little more than half of the subjects who failed to produce it evidenced reliable comprehension. The conclusion was drawn that comprehension learning precedes production learning and that production-type tasks may consistently underestimate linguistic knowledge.

Study of the fine structure of adeno-associated virus DNA with bacterial restriction endonucleases.

A physical map of the adeno-associated virus type 2 genome has been constructed on the basis of the five fragments produced by the restriction endonucleases HindII + III from Hemophilus influenzae. There are three endo R-HindII cleavage sites and one endo R-HindIII site. Evidence has been obtained to support the existence of two nucleotide sequence permutations in adeno-associated virus DNA, the start points of which have been estimated to be separated by 1% of the genome. The three cleavage fragments produced by endo R-Eco RI have been ordered and oriented with respect to the endo R-HindII + III cleavage map.
