Tumour necrosis factor alpha and interleukin 2 in normal and infected human seminal fluid.

The presence of cytokines such as the tumour necrosis factor alpha (TNF alpha) and interleukin 2 (IL2) in human spermatozoa is still to be defined. The aim of this study was to measure the concentration of both soluble factors in seminal fluid. Data from normal semen samples (n = 24) confirmed the presence of IL2 (258 +/- 84 fmol/ml corresponding to 953 +/- 369 fmol/total volume of ejaculate) and TNF alpha (62.2 +/- 16.4 fmol/ml corresponding to 231.3 +/- 86 fmol/total volume of ejaculate). A significant positive correlation (r = 0.59; P < 0.01) was observed between the TNF alpha and the IL2 concentrations. The concentrations of these cytokines were not related to sperm parameters. In contrast, IL2 concentrations (196.9 +/- 60.4 fmol/ml; 686.2 +/- 236.7 fmol/total volume of ejaculate) evaluated in 16 seminal fluids with identified bacterial agents were lower than in the control group, whereas TNF alpha concentrations (68.6 +/- 12.3 fmol/ml; 241.3 +/- 78.9 fmol/total volume of ejaculate) were not significantly different from the controls. Further studies are needed to determine the potential role of these cytokines in the physiology of semen and their usefulness as indicators of reproductive pathology.

[Interleukin-2 and human seminal fluid].

Involvement of the cytokines in male infertility were considered from in vitro studies, however the nature and the role of the mediator susceptible of arising in vivo is not completely elucidated. The purpose of this study is to measure the concentrations of interleukin-2 (IL-2) in normal and pathologic seminal fluid and to analyse their relationship with semen parameters. Our results demonstrate the presence of IL-2 in seminal fluid. Comparatively to normal seminal fluid, significant lower IL-2 concentrations were observed in infected semen samples or with sperm antibodies. No correlation was found between the cytokines values and the sperm parameters.

Immunocytochemical localization of angiotensinogen in rat liver and kidney.

The renin substrate, angiotensinogen, was localized by immunocytochemistry in liver and kidney of normal rats by the use of an antiserum directed against pure rat angiotensinogen. This substrate was also examined in rats after bilateral nephrectomy, which is known to increase plasma angiotensinogen, and in rats treated with colchicine, which inhibits serum protein secretion. In normal rat liver, light microscopy showed the presence of immunoreactive material in a very few cells. The number of stained hepatocytes rose in rats treated with colchicine or after bilateral nephrectomy. Immuno-staining increased further when rats were both nephrectomized and colchicine treated. In the kidney, angiotensinogen was specifically located as granular formations in nephrocytes of the proximal tubule but never in the granular cells of the juxtaglomerular apparatus. The localization of these granular formations under the brush border suggests that angiotensinogen is reabsorbed from the glomerular ultrafiltrate rather than synthesized in the kidney.

[Histofluorescence study of epiphyseal monoamines in the garden dormouse (Eliomys quercinus L.) during various experimental conditions in hibernation period (author's transl)].

During the hibernation period, the epiphyseal catecholamine charge is well detected in the garden dormouse; it appears more important in darkened animals at 22 degrees C and much less in animals under continuous lighting. Serotoninergic elements have only been shown in animals living under complete darkness. An important catecholaminergic and indolaminergic charge appears in sexually-hypoactive-animals; during ovarian activation after illumination a diminution (of this charge) is observed. In this species, an inverse relation exists between the importance of epiphyseal monoaminergic metabolism and the hypothalamic gonadostimulating catecholamine discharge.

[Influence of the photoperiod on the ultrastructure of the pineal gland before and during the seasonal genital cycle in the female garden dormouse (Eliomys quercinus L.) (author's transl)].

The pinealocyte of the female garden dormouse presents ultrastructural characteristics correlated with the sexual cycle. In winter, during the period of sexual quiescence, the pinealocyte is very rich in liposomes, myelinic bodies and lysosomes; the same holds during the summer quiescence state. These organelles and a great number of intracellular vacuoles are very well developed in dormice artificially maintained in anoestrous during the normal sexual activity period by light deprivation. In spring liposomes are strongly reduced in animals that are awake and sexually active, whereas the number of "synaptic ribbons" and of densecored vesicles increases. This ultrastructural aspect can be obtained during winter quiescence in animals living at 22 degrees C, under continuous light, and showing early sexual activation. If illumination is prolonged over the seasonal sexual phase dilated cisterns of granular endoplasmic reticulum, filled with proteinaceous material, appear in the cell. It seems that light no more activates, but inhibits gonadostimulator mechanisms. The pinealocyte's ultrastructural elements which are characteristic of a definite sexual state generally develop before this state is fully established. Actually, the functional significance of these organelles is not known.

[Early teratogen effect of neutral desoxyribonuclase on young chick embryo].

Neutral DNA-ase acts as a general teratogen when given to young chicken blastoderms. The rate of the differents abnormalities varies according to the dose given and the embryos' stage. The frequency of the different defects reaches a maximum value with increasing doses it falls generally down with more important doses. DMSO added to the enzyme solution changes the susceptibility of the embryos.
