A platelet abnormality in the Chediak-Higashi syndrome of man.

Platelets from two probands homozygous for the Chediak-Higashi syndrome have approximately 10% of the normal number of serotonin-containing dense bodies as visualized electron microscopically in air-dried whole mounts. Since transport of serotonin across the platelet plasma membrane proceeds at a normal rate, and the few dense bodies present appear to store normal amounts of serotonin, the absence of dense bodies may account for the low platelet serotonin values found in these patients.

Serotonin storage in platelets: estimation of storage-packet size.

Storage-body diameter and volume, and the number of molecules of serotonin contained in a storage body, were estimated for blood platelets. In the human, 5.23 x 10(5) molecules of serotonin are contained in a storage body 198 nanometers in diameter, while in the cat, 31.2 x 10(5) molecules of this amine are contained in a storage body 298 nanometers in diameter.

Recovery in non-vascular locked-in syndrome during treatment with Sinemet.

A case of successful pharmacological therapy of non-vascular locked-in syndrome (LIS) is described. A 22-year-old male experienced aqueductal stenosis following an episode of viral meningitis 11 months prior to admission. A ventriculo-peritoneal shunt was inserted and subsequently revised three times. Following the last procedure, the patient remained mute, quadriplegic and aphagic. There was no change in the patient's status for 9 days. Bromocriptine (2.5 mg daily) was begun and increased to 45 mg daily without any change in PECS scores. It was then tapered and Sinemet (75/300 daily) was begun and increased to 75/750 daily. The PECS scores in all areas rose steadily during the subsequent 6 weeks. Sinemet may be of value in the rehabilitation therapy of non-vascular LIS; serial patient evaluation by a PECS system may facilitate assessment of the response.
