The effects of minoxidil, 1% pyrithione zinc and a combination of both on hair density: a randomized controlled trial.

BACKGROUND: Recent studies of antidandruff shampoos or tonics containing antifungal or antibacterial agents produced effects suggestive of a potential hair growth benefit. OBJECTIVES: The purpose of this 6-month, 200-patient, randomized, investigator-blinded, parallel-group clinical study was to assess the hair growth benefits of a 1% pyrithione zinc shampoo. The efficacy of a 1% pyrithione zinc shampoo (used daily), was compared with that of a 5% minoxidil topical solution (applied twice daily), a placebo shampoo and a combination of the 1% pyrithione zinc shampoo and the 5% minoxidil topical solution. METHODS: Two hundred healthy men between the ages of 18 and 49 years (inclusive) exhibiting Hamilton-Norwood type III vertex or type IV baldness were enrolled. Total hair counts, the primary efficacy measure, were obtained using fibre-optic microscopy and a computer-assisted, manual hair count method. Secondary measures of efficacy included assessments of hair diameter, as well as patient and investigator global assessments of improvement in hair growth. These were based on photographs of the scalp using both midline and vertex views. RESULTS: Hair count results showed a significant (P < 0.05) net increase in total visible hair counts for the 1% pyrithione zinc shampoo, the 5% minoxidil topical solution, and the combination treatment groups relative to the placebo shampoo after 9 weeks of treatment. The relative increase in hair count for the 1% pyrithione zinc shampoo was slightly less than half that for the minoxidil topical solution and was essentially maintained throughout the 26-week treatment period. No advantage was seen in using both the 5% minoxidil topical solution and the 1% pyrithione zinc shampoo. A small increase in hair diameter was observed for the minoxidil-containing treatment groups at week 17. Assessments of global improvements by the patients and investigator generally showed the benefit of 5% minoxidil. The benefit of the 1% pyrithione zinc shampoo used alone tended (P < 0.1) to be apparent only to the investigator. CONCLUSIONS: Hair count results show a modest and sustained improvement in hair growth with daily use of a 1% pyrithione zinc shampoo over a 26-week treatment period.

[Structure and function of mouse p16INK4a locus].

Inactivation of p16INK4a is highly related with tumors. We obtained a 14.5 kb mouse genomic DNA fragment through screening the 129/SvJ mouse genomic library. Bioinformatics study suggests that the fragment contain three exons, which encode a 17,941 dolton polypeptide consisted of 168aa. The polypeptide has 7 potential phosphorylation sites, which indicates that the function of p16INK4a itself may be under the control of phosphorylation. Many interspersed elements and simple repeats were found in the noncoding region. These elements and repeats may cause retroposition and recombination and thus be responsible for some of the p16INK4a locus deletion. We also compared our sequence of exon 1 alpha with those published elsewhere and found that polymorphism may exist.

[Construction of a Flp "exchange cassette" contained vector and gene targeting in mouse ES cell].

Using the HPRT genomic DNA fragment and synthesized oligonucliotides, pSP-HPRT-F-Neo-F3 was designed and constructed as a replacement gene targeting vector by usual molecular cloning techniques. Structure of pSP-HPRT-F-Neo-F3 was identified by restrictive digestion analysis and partly sequencing. Then linearized pSP-HPRT-F-Neo-F3 DNA was electroporated into ES cells, and transfected cells were screened by being cultured in medium containing 200 micrograms/mL G418 and 2 micrograms/mL 6-GT. Twenty-four double drug resistant clones were picked up and analyzed, among them, two clones were proved to have taken place the required recombination by PCR and southern blotting analysis.

[The study of a HPRT locus-specific transgenic method based on FLP recombinase mediated cassette exchange].

Exchange plasmid pF-HPRT-F3 and Flp expression plasmid pCMV-Flp were constructed and then introduced using electroporation system into F18 ES cell line, which have an exchange cassette F-Neo-F3 at HPRT locus. After HAT selection, HAT resistant clones were obtained. Then G418 sensitivity test and Southern blotting were carried out to screen RMCE recombinants. The results indicated that RMCE had taken place in three of 12 HAT resistant clones. The frequency is 25%. The result demonstrates that it is realizable to introduce transgene to HPRT locus by using Flp recombinase mediated cassette exchange reaction.

[Construction of gene targeting vector for duplicating p35Nck5a gene and its application in the gene targeting of ES cells].

To duplicating the regulatory subunit p35Nck5a gene of mouse neuronal cdc2-like kinase in embryonic stem (ES) cells, about 12.2 kb of pGDTV vector for p35Nck5a gene duplication was constructed. The linearized pGDTV vectors were transfected into ES cells by electroporation. 245 drug-resistant cell clones were obtained in both G418 and GANC medium and the frequency of surviving cells was 6.22 x 10(-5). The ES cell clones were identified to have duplicated the p35Nck5a gene by use of both PCR and genomic Southern blotting, and the frequency of homologous recombination is 5.08 x 10(-7). The use of negative selection gene (HSV-tk) results in 7-fold increase at selection efficiency. This study lays the foundations of preparing mouse models of Alzheimer's disease.

Desflurane and isoflurane produce similar alterations in systemic and pulmonary hemodynamics and arterial oxygenation in patients undergoing one-lung ventilation during thoracotomy.

UNLABELLED: We tested the hypothesis that desflurane (DES) and isoflurane (ISO) produce similar effects on systemic and pulmonary hemodynamics and arterial oxygenation before, during, and after one-lung ventilation (OLV) in patients undergoing thoracotomy. After obtaining informed consent, anesthesia was induced with sodium thiopental or thiamylal, fentanyl, and vecuronium in 61 ASA physical status II-IV patients. Patients were randomly assigned to receive either DES (n = 30) or ISO (n = 31) in 100% O2 in separate groups. Hemodynamic data (radial and pulmonary artery [PA] catheters) were recorded, and blood gas values were obtained before and after induction; at selected intervals before, during, and after OLV; and before emergence. DES significantly (P < 0.05) increased heart rate (HR) and decreased mean arterial pressure (MAP) and cardiac output (CO). PA pressures and pulmonary vascular resistance (PVR) increased; systemic vascular resistance (SVR) was unchanged. Increases in HR and CO and decreases in MAP and SVR occurred during OLV and DES. Reductions in PaO2 (411 +/- 88 to 271 +/- 131 mm Hg 5 min after beginning OLV; mean +/- SD) and content (CaO2) and increases in shunt fraction (Qs/Qt; 0.25 +/- 0.12 to 0.40 +/- 0.19 at 5 min after beginning OLV) were also observed. ISO increased HR and PA pressures but did not alter MAP, CO, and PVR, in contrast to the findings with DES. Reductions in MAP and SVR and increases in CO and PA pressures were observed during OLV in the presence of ISO. Similar to the findings during DES, decreases in PaO2 and CaO2 and increases in Qs/Qt occurred during OLV and ISO. We conclude that DES and ISO produce very similar alterations in systemic and pulmonary hemodynamics and arterial oxygenation in patients undergoing OLV during thoracotomy. IMPLICATIONS: Desflurane and isoflurane produce similar cardiovascular and pulmonary effects before, during, and after one-lung ventilation in patients undergoing lung surgery.

Perspectives on genetics in china.

The long civilization of China traces its origin to the development of agriculture. Early records and archaeological findings demonstrate the success of plant and animal breeding in contributing to human livelihood and the national economy. Teaching and research in modern genetics began in China in the 1920s with individuals who received advanced training in the West. Over the following 30 years, notable contributions to genetics, both basic and applied, were made by Chinese scientists in universities and research institutions. Unfortunately, these activities were brought to a virtual standstill first by the official advocacy of Lysenkoism, followed by ten years of Cultural Revolution. Schools were closed and scientific research was interrupted. Not until 1978 did the universities begin to enroll new students and scientific research, including genetics, again receive endorsement. Since then, Chinese scientists have resumed contacts with colleagues in other countries and publication of their research findings in national and international journals. The material summarized in this article covers a long time span and a broad spectrum of scientific subjects and events. Representative examples of Chinese contributions to various areas of genetics are cited to depict the history, current status, and prospects for the development of genetics in China. After many ups and downs and despite limitations in material resources, the study of genetics in China seems to be on a sound footing, with a relatively complete system in place for training workers in this discipline.

Neurovirulence determinants of genetically engineered Theiler viruses.

Theiler murine encephalomyelitis viruses (TMEVs) are picornaviruses that cause enteric and neurological disease in mice. The GDVII strain and other members of the GDVII subgroup are highly virulent and cause an acute, fatal polioencephalomyelitis following intracerebral inoculation, whereas the DA stain and other members of the TO subgroup cause a persistent, demyelinating infection. We previously produced a full-length, infectious DA cDNA clone. We now describe the generation of a full-length, infectious GDVII cDNA clone and the subsequent production of intratypic chimeric cDNAs and intratypic recombinant viruses. Inoculation of the recombinant viruses into mice demonstrated that a major determinant of TMEV neurovirulence is within the GDVII 1B (capsid protein VP2)-2C coding region, most likely in the GDVII 1B (VP2)-2A coding region. Genomic sequences 5' to this region of GDVII RNA also contribute to expression of the full neurovirulence phenotype. These data demonstrate the multigenic nature of TMEV neurovirulence, as has been reported for other viruses.

Infectious cDNA clones of the DA strain of Theiler's murine encephalomyelitis virus.

The DA strain and other members of the TO subgroup of Theiler's murine encephalomyelitis viruses cause a persistent demyelinating infection, whereas the GDVII strain and other GDVII subgroup strains cause an acute lethal polioencephalomyelitis. We generated an infectious DA cDNA clone inserted into a transcription vector. Virus derived from transfection of transcripts produced a demyelinating disease indistinguishable from that of wild-type virus. The infectious clone provides a critical reagent for the production of interstrain recombinant viruses to help identify genetic loci responsible for the biological activities of the strains.

[Comparative study on protein components of ouabain resistant mutant and wild cells of Chinese hamster V79 cell line by two-dimensional gel electrophoresis].

In the sense of gene expression, the primary and direct change in mutant cells should be involved in the protein component. Two-dimensional polyacrylamide gel electrophoresis has been used to compare the protein components of ouabain resistant mutant cells (ouaR) and sensitive wild type cells (ouaS) of chinese hamster lung cell line V79. The procedure is based on O'Farrell's method and we have made some modifications. In order to make the study reliable, we took twenty polypeptide spots as a reference marker. These marker spots could always be reproduced in a series of parallel experiments with both mutant and wild cells. In addition, these markers covered different small areas, representing different isoelectric point (pI) and molecular weight (MW) on the electrophoretogram. With reference to these marker spots, some polypeptide spots showed variations in their position, size and density. It is notable that the changes appeared in the small area 4 (pI/MW:5.0/28-30kd) and the small area (pI/MW:6.3/39kd) is highly reproducible and significant, so that these changes might be specific to ouabain resistant mutant of V79 cells. Since the level of genetic polymorphism of mammalian cells on two-dimensional gel electrophoresis is quite low, these specific changes might reflect the differences in gene structure and expression. This primary study should enhance the usefulness of two-dimensional gel electrophoresis for mutagenesis research with cultured mammalian somatic cells.

Molecular cloning and sequence determination of DA strain of Theiler's murine encephalomyelitis viruses.

Theiler's murine encephalomyelitis viruses (TMEV) belong to the Picornaviridae, and are divided into two subgroups. TO subgroup strains produce a persistent demyelinating central nervous system infection in mice, while GDVII subgroup strains cause acute polioencephalomyelitis. We generated three overlapping clones of the genome of DA strain, a member of TO subgroup. Sequence analysis revealed that the genome is 8093 nucleotides long with a poly(A) tail. The 5' noncoding region stretches from nucleotides 1 to 1065 and lacks a poly(C) tract. The open reading frame stretches from 1066 to 7968 and encodes 2301 amino acids. DA strain sequence is more closely related to members of the Cardiovirus genus than to members of other Picornavirus genera. Comparison with sequence of BeAn strain, another TO subgroup strain, showed that the P1 area has the greatest number of differences, while the noncoding regions are more well-conserved. The three overlapping clones will be important in recombinant infectious cDNA studies between strains of both subgroups.

Trypsin-sensitive neutralization site on VP1 of Theiler's murine encephalomyelitis viruses.

We generated Theiler's murine encephalomyelitis virus mutants resistant to several neutralizing monoclonal antibodies (MAbs) having their epitopes near a trypsin cleavage site of VP1. Neutralization and Western blot (immunoblot) studies suggest that two of the MAbs have identical epitopes that partly overlap the epitope of a third MAb. Sequencing of RNA of the mutants localized the epitopes to a site near the carboxyl end of VP1. The limited diversity of nucleotide changes seen in the mutants and the immunodominance of the site suggest that the carboxyl end of VP1 may have an important function.

Measurement of acetol in serum.

A method for the derivatization of acetol (1-hydroxyacetone) with 2,4-dinitrophenylhydrazine (DNPH) and for the measurement of the acetol dinitrophenylhydrazone derivative (acetol-DNPH) by high-performance liquid chromatography is presented. The chromatographic separation described here resulted in baseline resolution of the acetol-DNPH peak. Peak integration was proportional to serum acetol concentration over a 5- to 500-nmol/ml range. No other method for the determination of acetol in serum currently exists. Serum from rats in diabetic ketoacidosis was found to contain 11.2 +/- 1.1 nmol acetol/ml serum (N = 3). Serum from a 21-day-fasted human contained 16 nmol/ml acetol. Serum from rats maintained on drinking water containing 1% acetone (v:v) for 6 days contained 152 +/- 31 nmol/ml acetol (N = 5). The presence of acetol in serum under conditions where acetoacetate and acetone are chronically elevated suggests that acetoacetate may be converted to glucose through the conversion of acetone to acetol and L-1,2-propanediol.
