Isolation of Mycoplasma mycoides, mycoides (LC variant), from two naturally aborted caprine fetuses.

Described in this study are 2 cases of caprine abortion associated with the LC type of Mycoplasma mycoides , subsp. mycoides . This mycoplasma and Mycoplasma mycoides , subsp. capri had been previously reported in adult goats in this herd. The abortion took place in the latter part of gestation. Samples from heart blood, lung and pleural exudate were collected for the isolation of mycoplasmas and other bacterials in both fetuses. Two strains of Mycoplasma mycoides , subsp. mycoides (LC variant) were isolated. The only gross lesion of the internal organs in the aborted fetuses was congestion of the lungs. Microscopic lesions were encountered in the lungs, and these were characterized by patchy to diffuse pneumonia. Exfoliated cells, many alveolar macrophages, scattered neutrophils and lymphocytes were seen in the lumen of the terminal airways and alveolar spaces. This report appears to be the first isolation of Mycoplasma mycoides , subsp. mycoides (LC variant) from aborted caprine fetuses.

High mortality in goats associated with the isolation of a strain of Mycoplasma mycoides subsp. mycoides (Large Colony type).

A goat pleuropneumonia outbreak occurring in a herd of 800 animals in the province of Ciudad Real, Spain, is described in this paper. Severe respiratory signs and high mortality were the most significant clinical observations. The adult goats presented mainly respiratory symptoms and/or mastitis, whereas the young animals died showing arthritis and/or keratoconjunctivitis. The most significant lesions were found in the thoracic cavity. A focal extensive fibrinonecrotic pleuropneumonia was macroscopically seen, and the histopathological analysis confirmed a fibrinopurulent and necrotic pleuropneumonia with areas of acute pyogenic bronchopneumonia and fibrinous pericarditis associated with a multifocal purulent mastitis and/or a fibrinopurulent arthritis in some goats. Microbiologically, the mycoplasmas isolated grew rapidly (18-24 h), and, after 48 h, there were colonies of 1-1.5 mm diameter. These isolations were biochemically characterized as Mycoplasma mycoides spp. and showed serological characteristics corresponding to Mycoplasma mycoides subsp. mycoides Large Colony. One of the isolations, the strain 2/93, experimentally showed its pathogenicity causing acute interstitial pneumonia and arthritis when it was inoculated in 1-week-old kids.

Polyarthritis in kids associated with Mycoplasma putrefaciens.

Fifteen two- to three-week-old kids, fed artificially with goats' milk from a dairy, were found to have polyarthritis. The most affected joints were the carpals, either unilaterally or bilaterally, and in some cases the tarsal joints. Mycoplasma putrefaciens was isolated from the joints which showed an acute fibrinopurulent arthritis. No clinical mastitis was detected in the dairy.

Sjögren-like pluriglandular exocrine insufficiency after drug-induced toxic epidermal necrolysis.

We present the case of a patient that progressively developed xerophthalmia, xerostomia, cutaneous xerosis and exocrine pancreatic insufficiency 3 months after metamizole-induced toxic epidermal necrolysis. Though the association of Sjögren's syndrome and exocrine pancreatic impairment is well established, the Sjögren-like syndrome after drug-induced toxic epidermal necrolysis in association with such a wide exocrine glandular insufficiency has not been previously described, to our knowledge.

Selective intestinal decontamination increases serum and ascitic fluid C3 levels in cirrhosis.

Selective intestinal decontamination for 7 days with norfloxacin was performed in 14 cirrhotic patients with ascites and low ascitic fluid total protein. Variations in serum and ascitic fluid of C3 and C4 and ascitic fluid total protein after therapy were compared with those of a control group of 14 untreated patients with similar characteristics. After oral norfloxacin administration, we saw a significant increase of C3 in serum (p less than 0.05) and ascitic fluid (p = 0.01). A significant increase was also observed in ascitic fluid total protein (p less than 0.05) but not in serum and ascitic fluid C4. There were no changes in serum C3, ascitic fluid C3, ascitic fluid C4 or in ascitic fluid total protein in group 2. These data demonstrate that selective intestinal decontamination increases serum and ascitic fluid C3 levels and, therefore, might be useful in preventing spontaneous infections in cirrhotic patients at high risk of infection.

Chromatin structure of transposon Tn903 cloned into a yeast plasmid.

Transposon Tn903 contains the APH gene for kanamycin resistance, which is active in yeast [A. Jiménez and J. Davies (1980) Nature (London) 287, 869-871] and is flanked by two inverted repeats (IR) 1057 bp long. When plasmid pAJ50, carrying Tn903 and the 2-microns circle origin of replication, is cloned into Saccharomyces cerevisiae, nucleosomes are assembled in vivo on the prokaryotic DNA of the transposon. Indirect end labeling revealed that three nucleosomes are preferentially positioned on symmetrical sequences from both IRs. DNase I digestion also confirmed that the chromatin structure is symmetrical in both IRs. This suggests that sequence determinants are decisive for chromatin structure in these regions. We have calculated the rotational and translational fits [H. R. Drew and C. R. Calladine (1987) J. Mol. Biol. 195, 143-173] for the Tn903 sequence and the results indicate that the nucleosome positioning on the IRs is sequence-directed. Nucleosome deposition on the APH gene also occurs, but no clear positioning exists. Some sequence preference for positioning nucleosomes on the promoter can be predicted, especially from the translational fit. Experimental data indicate, however, that nucleosomes are absent from the promoter. Therefore, chromatin can be organized on prokaryotic DNA in a manner that resembles the typical eukaryotic chromatin structure.

[Clinical characteristics of chronic autoimmune hepatitis. Study of 51 cases].

The clinical and histological features of the 51 patients affected with chronic autoimmuno hepatitis are analyzed. 65% of them were over 50 years old. 41% were without symptoms. The incidence of systemic symptoms was of 25.5%. 84.3% had antinuclear antibodies superior of 1/40 and 53% had positive LMA. The incidence of HCA and CH was similar. 26.7% of the patients had complications.

[Variation of C3 in ascitic fluid from cirrhotic patients subjected to intestinal sterilization or selective intestinal decontamination].

Patients with liver cirrhosis with ascites and low levels of C3 and Total Proteins in ascitic fluid show a greater predisposition to the development of spontaneous bacterial peritonitis. The variation of C3 and Total Proteins levels in ascitic fluid in two groups of patients with liver cirrhosis was studied. Group I (n = 14) underwent intestinal sterilization (n = 7) or selective intestinal decontamination (= 7). Group II was a control. A statistically significant increase in C3 (p less than 0.01) and Total Proteins (p less than 0.015) levels in ascitic fluid in Group I patients was found. There were no changes in the control group. The incidence of side effects was higher in those patients who underwent intestinal sterilization. Our results suggest that selective intestinal decontamination may be useful as a prophylactic measure against spontaneous bacterial peritonitis in those patients with liver cirrhosis at high risk of infection by increasing the bactericidal capacity of ascitic fluid.

Osteochondroma induced by reflection of the perichondrial ring in young rat radii.

In order to investigate the effects of reflection of the perichondrial ring in osteochondroma formation, the perichondrial rings of the epiphyseal growth plates in 42 young rat radii were turned to the metaphyseal periosteum by means of blunt dissection. Seven days after surgery a small nest of chondrocytes appeared on the metaphyseal-diaphyseal bone surface at the level of the tip of the reflected perichondrial ring. From the 9th to the 15th days the histological pattern of the osteochondroma was established. The osteochondroma was not connected with the hypertrophic cartilage of the growth plate. During the third and fourth weeks the osteochondroma began to regress with the disappearance of the cartilage nest. During the development of the lesion the bone grew normally and the growth plate migrated distally while the lesion remained at its initial site. The growth plate zone devoid of perichondrial ring was covered by fibrous connective tissue and no removal of the perichondrial ring occurred. These results suggest that the origin of this osteochondroma is the perichondrial ring cells whose polarity has been surgically changed.

[Comparative immunopathologic study of oral aphthae, non-involved mucosa from aphthae patients, healthy mucosa and trauma-induced ulcers].

The presence of IgG, IgA, IgM, C1q, C3c, fibrinogen and properdin has been studied using direct immunofluorescence technique. The same study has been performed in clinically normal mucosa of patients with aphthous ulcers, normal mucosa of healthy volunteers and traumatically induced ulcers. We have also studied the possible presence of reactive serous immunoglobulins with normal mucosa by indirect immunofluorescence. Our findings do not support previous studies which claimed the presence of vasculitis induced by the presence of immunoreactants in the vessel walls.

[Problems of neonatal screening for hypothyroidism in the very low birth-weight neonate].

21,013 newborns were screened on the first year of a neonatal screening program on congenital hypothyroidism in the Basque Community. The evaluation of the results showed some points of interest. One of them, would be the high analytical retesting rate needed in the screening of very low-birth weight children. The rate decreased with increasing birth-weight. Forty per cent of the children with a birth-weight lower than 1,000 gr needed retesting. The rate decreased to 7.7% at birth-weights between 1,001 and 1,500 gr, to 2% at birth weights between 1,501 and 2,500 gr and only to 0,42% at birth-weights higher than 2,501 gr. The consequences of these results are discussed.

Anti-golgi complex autoantibodies in a patient with Sjögren syndrome and lymphoma.

During routine immunofluorescence studies of the serum of a patient with Sjögren's syndrome and lymphoma we detected antibodies giving a cytoplasmic pattern which did not correspond to previously described patterns found for autoantibodies. Using different cells and tissues as substrates for indirect immunofluorescence, including rat liver, rat small bowel, rat testicle, human thyroid, guinea-pig plasma cells and cultured human fibroblasts, the cytoplasmic structure to which these autoantibodies are directed seems to be the golgi complex, a conclusion supported by histochemical studies. Furthermore, these antibodies were absorbed by isolated golgi vesicles. The autoantibodies are of IgG and IgA classes, and the antigen(s) with which they react is(are) resistant to treatment with DNAse and RNAse. None of the sera from 50 normal individuals, seven patients with Sjögren's syndrome (five of them primary and two associated with rheumatoid arthritis; none of them with lymphoma), 25 patients with mixed connective tissue disease, 10 patients with systemic lupus erythematosus and five patients with progressive systemic sclerosis, had antibodies directed against this cytoplasmic specificity, as determined by indirect immunofluorescence. This is the first time that autoantibodies directed to the golgi complex are reported. The significance of this finding awaits further descriptions in patients with a clinical picture similar to the one reported here.

Effects of carbenicillin on blood coagulation: a study in patients with chronic renal failure.

A study of coagulation has been performed on 8 chronic renal failure patients receiving carbenicillin therapy. All showed a prolongation of the bleeding, recalcification, partially-activated thromboplastin, prothrombin and thrombin times. These findings suggest the presence of an anticoagulant with an heparin-like mode of action. In vitro tests suggest that carbenicillin may be this factor. We have also shown that the drug produces a disturbance in the normal polymerization process. The implications of these findings for the treatment of (CRF) patients with carbenicillin are discussed.
