Determination of selective biochemical variables in pregnant and lactating mice after stobadine administration.

The aim of this study was to investigate the effects of the oral administration of stobadine (STB), a neuro- and cardioprotective drug with high antioxidant properties, on selective biochemical variables in pregnant and lactating mice. STB was administered orally at a dose of 50 mg/kg from day 15 of gestation to day 21 of lactation. Creatinine and urea were determined in serum, while acidity, proteins, glucose, ketones, bilirubin, urobilinogen, blood and creatinine were determined in urine from females on days 0, 15 and 18 of gestation and on days 7, 14, 21, and 28 postpartum (pp). In the biochemical variables investigated, no significant differences in STB-treated animals compared with controls were recorded on any of the days studied. Histopathological examination of kidney tissue did not reveal any adverse effect of STB administration.

Mammary gland carcinoma-related increase of type I iodothyronine 5'-deiodinase activity in Sprague-Dawley rats.

Type I, iodothyronine 5'-deiodinase (5'-DI) catalyses deiodination of the prohormone thyroxine (T4) to the metabolically active 3,5,3'-triiodo-L-thyronine (T3). The present study was undertaken to investigate the activity of 5'-DI in rat mammary gland tumours representing various combinations of histologically defined papillary, cribriform or comedo patterns of ductal carcinomas. Female Sprague-Dawley rats were given two doses 50 mg x kg(-1) 1-methyl-1-nitrosourea (MNU) in abdominal parts on the 52nd day and 113th day of age. We have found that in comparison with non-lactating mammary gland, the activity of 5'-DI in all mammary gland tumours studied was significantly (p < 0.0001) increased and that the 5'-DI activity, expressed as pmol of 125I- released per min and per mg of protein, in malignant mammary gland tumours was found to be at least two order higher than that of intact mammary non-lactating gland. From our data, we suggest that thyroid hormone in mammary gland tumours might play a significant role to support high energetic expenditure of neoplastic tissues.

Reduction of 1-methyl-1-nitrosourea-induced mammary gland carcinoma by in vivo application of immunostimulatory CpG motifs in Sprague-Dawley rats.

In the present work the role of 13-cis retinoic acid and CpG oligodeoxynucleotides (CpG-ODN) in a 1-methyl-1-nitrosourea (MNU)-induced mammary gland carcinoma animal model was investigated. Treatment with both components, applied either alone or in combination, induced a significant decrease of the tumour burden and the volume of tumours only in rats that received CpG-ODN (p = 0.046, compared to the MNU control group). The data indicate that the Th-1 biased immunostimulatory capacities of CpG motifs may play a significant role in induction of protective immune responses against mammary gland tumours in Sprague-Dawley rats.

[Detection of silicon in a human spleen using x-ray microanalysis].

There is a lack of information on x-ray microanalysis (XMA) of the human spleen. We have examined the spleen of a patient with autoimmune thrombocytopenic purpura. The specimens were fixed in glutaraldehyde and coated by copper. The XMA was performed by a Quantum detector and a Delta-Class analyzer. In addition to spectral lines of elements normally present in human tissues, we have demonstrated silicon. The presence of silicon in human spleen may be a consequence of the organ's cleansing function.

Treatment of rat gliomas with recombinant retrovirus harboring Herpes simplex virus thymidine kinase suicide gene.

The retrovirus vector containing Herpes simplex virus type 1 thymidine kinase (HSVtk) gene was constructed. The vector was transfected into the packaging cell line PG13. It was shown that individual transfected cells differ in the production of recombinant retrovirus and in their susceptibility to be killed by ganciclovir. Recombinant retrovirus with a gibbon envelope was able to transduce the HSVtk gene into rat glioma cells. In vivo studies confirmed the ability of intraperitoneal ganciclovir administration to influence subcutaneous and intracerebral tumors developed after injection of C6 rat glioma cells with subsequent injection of HSVtk retrovirus producing cells.

[Extrauterine choriocarcinoma--a rare form of gestational trophoblastic disease].

Choriocarcinoma represents the most serious form of trophoblast gestation disease. In the majority of cases the carcinomatous tissues fill out the uterine cavity, or they grow in a form of nodes deep in the uterine wall. The primary extrauterine localization of this tumour is very rare. The authors describe two cases of choriocarcinomas with tubal or ovarian localization. (Fig. 4, Ref. 19.)

Determination of selective biochemical parameters in pregnant and lactating rats after stobadine administration.

The effect of oral administration of stobadine, a cardioprotective drug, was studied on the basis of determination of selective biochemical parameters in pregnant and lactating rats. Stobadine was administered orally at a dose of 50 mg/kg from day 15 of gestation until day 21 of lactation. We determined creatinine and urea in serum, acidity, protein, glucose, ketones, bilirubin, urobilinogen, blood, and creatinine in urine from females on days 15 and 20 of gestation and 7, 14, and 21 of lactation. In the biochemical parameters investigated no significant differences between control and experimental animals were recorded on any of the days studied. Histopathological examination of kidney tissue did not reveal kidney damage after stobadine administration.

The use of lectins identified with specific antibodies in lectin histochemistry of NZB/W F1 mouse kidney.

The affinity of Helix pomatia, peanut, Pisum sativum, soy bean, and wheat germ agglutinins to various nephron parts of NZB/W F1 mice was different and is assumed to be age dependent. The affinity of Pisum sativum agglutinin to basal membranes of small renal vessels increased with the age of NZB/W F1 mice. The wheat germ agglutinin bound to structures with alkaline phosphatase activity.

[Structural basis of appendiceal function in rabbits].

Enzymo-, immuno- and lactin-histochemical methods were used to study the structure of the rabbit appendix wall. The value of some structural components in the function of this part of the intestine is discussed. Some findings were documented electronmicroscopically. In addition to its resorptive function, the rabbit appendix is equipped with a potent defense mechanism against adverse environmental effects of the appendix content. Individual structures of this defensive barrier are closely characterized with regard to cellular equipment and possibilities of its morphological identification.

Effect of the cardioprotective agent stobadine on reproduction in rats.

The cardioprotective drug stobadine in the form of dipalmitate salt DP 1031 was evaluated for its effects on perinatal and postnatal development in the rat. Doses of 5, 15 and 50 mg/kg/day representing approximately 1, 3, and 10 times the anticipated maximum daily human therapeutic dose were administered to rats orally in aqueous suspension. Treatment of pregnant rats with stobadine continuously from day 15 of gestation through parturition and lactation had no adverse effects on reproductive parameters of dams or on survival and development of F1 offspring at any dose used. There were only signs of slight maternal toxicity at 50 mg/kg/day, which consisted of sedated behaviour, reduced liver weight and reversible histopathological changes in kidney tissue. Reproductive toxicity studies with cis-(-)-2,3,4,4a,5,9b-hexahydro-2,8-dimethyl-1H-pyrido-[4,3-b]indole dipalmitate in rats.

cis-(-)-2,3,4,4a,5,9b-Hexahydro-2,8-dimethyl-1H-pyrido-[4,3-b]indole dipalmitate (stobadin dipalmitate; in the following briefly called STB) a new prospective cardioprotective drug, was evaluated for effects on fertility, general reproductive performance, prenatal and peri-postnatal development in the rat. Doses of 5, 15 and 50 mg/kg/d were administered orally in aqueous suspension to rats in all studies. Daily treatment of male rats for 70 days before mating and female rats for 14 days before mating and during gestation and lactation had no adverse effects on fertility, survival rate and weight gains of parental animals or on prenatal and postnatal development of pups. There was only evidence of slight adult toxicity late in the experiment, significant increase of anomalous foetuses in both the 15 and 50 mg/kg/d doses and decreased body weight of the young at 50 mg/kg/d on day 21 post partum. Daily oral treatment of pregnant rats with STB throughout organogenesis (day 4 to 16) had no overt effects on dams or on embryo-foetal development, except of increased incidence of some skeletal variations in all treated groups. In the peri-postnatal toxicity study treatment of pregnant dams with STB continuously from day 15 of gestation through parturition and lactation had no adverse effects on reproductive parameters of dams or on survival and development of F1 offspring at any dose used. There were only signs of slight maternal toxicity at 50 mg/kg/d, which consisted of sedated behaviour, reduced liver weight and reversible histopathological changes in kidney tissue. The results of these studies did not reveal serious developmental hazard potentials of STB administered to rats in doses up to 50 mg/kg/d.

[The estrogenic effect of the fungicide Novozír MN 80 on the uterus in neonatal rats].

The estrogenic effect of the ethylenebisdithiocarbamate fungicide Novozír MN 80 (Duslo, Sal') was studied by means of the screening method determining the reproductive risk of chemical substances, recommended by the Environmental Protection Agency. Compared to intact controls, a significant increase in relative uterine weight was recorded in the group of animals which received 1/25 of the maximal tolerable concentration of Novozír MN 80. Compared to negative controls (Tween 80), the relative uterine weight was increased significantly after administration of 1/25 and 1/50 of the maximal tolerable concentration of Novozír MN 80. On histologic examination the cuboidal epithelium of the mucous membrane was found to be changed into columnar epithelium, with uneven distribution of enlarged nuclei in the vacuolized cytoplasm. At 1/25 maximal tolerable concentration thickening of the uterine wall was observed.

[Structural basis of the spleen in rats].

The basic function of the spleen in the rat, similarly as in man, is to cleanse the blood of damaged old particles of the body itself, but also of foreign particles. To fulfill this function, the spleen is equipped with the white and red pulp with a specific structure of blood circulation. In an open system of circulation, blood from the arterial terminals opens into the cords of the red pulp, where it is adequately processed. From the cords the blood gets through the walls of sinuses, acting as the last filtration barrier, into the lumen of sinuses and then into the venous circulation. Unlike the human spleen, this organ in the rat has a marked marginal sinus and channel systems bridging the marginal zone. The channels mediate the circulation of lymphocytes between the red white pulp.
