[Ocular lesions induced by saccharin and its pollutants in the rat fetus].

The authors analyze and compare the teratogenic effects produced on the embryo eyes of rats by Maumee saccharin, by non purified Remsem saccharin and several pollutants. The study has been made on 2708 embryos of rats whose mothers have received per os several pollutants at several rates; 16 diets have been studied. The histological lesions are described: the cataract, the retinal coloboma, the major microphthalmos, the anophthalmos, the presence of aberrant nervous fibers, the anarchic globes. The authors describe the retinal eversion phenomenon in the retinal coloboma as well as its repercussion on the ocular malformations (retinal dysplasia and colobomatous cyst of the orbit). The presence of aberrant nervous fibers inside the retinal coloboma could explain the cases of double papilla, which were observed in clinic. The major microphthalmia are frequent. The serious anophthalmia are explained by an early perturbation of the embryogenesis. The anarchic globes could be interpreted as a form of congenital cystic eye. Under the different pollutants of the saccharin, four of them are more toxic: it is the ortho-sulfobenzoic acid, the para-sulfobenzoic acid, the para-sulfamolybenzoic acid, the para-toluenesulfonamide which communicate the Remsen saccharin its teratogenic effect. On the other hand, the ortho-toluene sulfonamide is without any teratogenic effect. The authors conclude on the necessity of a large purity of the commercial saccharin.

[Cigarettes, contraceptive drugs and lipidogenesis in the female].

In women between 18 and 45 years of age consuming either tobacco or an oral contraceptive triglycerides are significantly higher and HDL cholesterol significantly lower than in controls. In women consuming both tobacco and an oral contraceptive, a cumulative effect is demonstrated on these parameters.

[Lipoproteinemia throughout puberty].

In prepubertal children, total cholesterol and HDL cholesterol concentrations are identical in both sexes, while total cholesterol/HDL cholesterol and LDL + VLDL cholesterol/ HDL cholesterol ratios are slightly higher in girls. During puberty, total cholesterol concentrations decrease slightly more in boys than in girls; HDL cholesterol concentrations remain unchanged in girls but decrease significantly in boys. When puberty is completed, in comparison to prepubertal values, total cholesterol/HDL cholesterol and total cholesterol/LDL + VLDL cholesterol ratios are significantly increased in boys, while they are unchanged in girls. During puberty, rises in triglyceride concentrations are significant in boys only.

[Physical activity in the management of hyperlipoproteinemia].

Increasing patients' physical activity significantly enhanced the efficiency of nutritional management in three forms of hyperlipoproteinemia: hypercholesterolemia, hypertriglyceridemia, and hypercholesterolemia with hypertriglyceridemia. Although total cholesterol concentrations are only moderately decreased by physical exertion, HDL cholesterol concentrations are increased, resulting in a significant fall in total cholesterol/HDL cholesterol ratios. In addition, triglyceride concentrations decrease significantly.

Are acetylcholine-induced increases in 42K efflux mediated by intracellular cyclic GMP in turtle cardiac pace-maker tissue?

1. 42K efflux has been measured from small strips of turtle sinus venosus tissue in order: (a) to characterize further the pharmacology of the acetylcholine response and (b) to test whether cyclic guanosine 3':5'-monophosphoric acid (cyclic GMP) is the intracellular mediator of this response. 2. The 42K wash-out curves show that the fractional escape rate (FER) of 42K efflux is nearly constant after 60-80 min, indicating that after this time period 42K FER is controlled by barrier-limited diffusion from a single intracellular compartment. 3. The threshold of the dose-response relationship for the acetylcholine-induced increase in 42K FER is about 10(-8) M and the Km is 2.75 x 10(-7) in non-eserinized preparations. 4. This acetylcholine response is completely blocked by atropine; but nicotinic blockers produce no detectable reduction of it. 5. Exogenous application of lipid-soluble analogues of cyclic GMP (dibutyryl or 8-bromo-cyclic GMP applied at 2-3 mM for 30 min) failed to mimic the acetylcholine-induced augmentation of 42K FER. 6. Experiments in which sodium nitroprusside (5 x 10(-4) M for 30 min) was applied in order to stimulate the guanylate cyclase and hence produce a large, maintained increase in intracellular cyclic GMP failed to show a significant increase in 42K FER. 7. When acetylcholine (10(-6)M) was applied in the presence of O[Ca2+]0 (in an attempt to inhibit the guanylate cyclase) there was no significant reduction in the acetylcholine-induced increases in 42K FER. 8. Hence, these three indirect tests indicate that the muscarinic acetylcholine-induced increase in 42K FER in cardiac pace-maker tissue is unlikely to be mediated entirely by changes in the levels of intracellular cyclic GMP.

[Linoleic acid in nutritional balance: experimental studies].

In the male rat a diet rich in beef fat facilitates the occurrence of hyperinsulinemia after a glucose load whereas fats rich in linoleic acid produce no such effect. The combination of saturated fat and saccharose facilitates the occurrence of hypertriglyceridemia in the male rat, no such effect is produced by the combination of fats rich in linoleic acid and saccharose. Linoleic acid prevents natrium chloride from provoking hypertriglyceridemia in male and in female rats subjected to a diet enriched in saccharose and fat. Estrogen-induced hypertriglyceridemia in castrated animals is strongly inhibited if the diet is rich in linoleic acid. Physical effort can prevent saccharose combined with saturated fats from inducing hypertriglyceridemia.

[Lipidogen effect of sorbitol with saturated fat in the diet of the male rat (author's transl)].

In the male rat, sorbitol, beef fat nor M.C.T. in the diet provoke hypertriglyceridemia but the association sorbitol-beef fat and sorbitol-M.C.T. significantly rise the triglyceridemia and the cholesterolemia. In the castrated male rat estrogens provoke a considerable rise of the triglyceridemia and the cholesterolemia with the diet enriched in M.C.T. but not modify the hypertriglyceridemia and the hypercholesterolemia provoked with the diet enriched with the association sorbitol-T.C.M.

Neo natal androgenisation of the female rat provoke at 3 months : 1) an accentuation of the insulin secretion at the glucose loading with the diet rich in saturated fat an a reduction with the diet rich in sucrose or rapesced oil; 2) Hypercholesterolemia with all the diets and 3) hypertriglyceridemia with diet rich in saturated fat or sucrose. Neo natal oestrogenisation of the male rata provoke at 3 months : 1) a reduction of the insulin secretion at the glucose loading with all the diets 2) hypercholesterolemia with the diet rich in saturated fats 3) hypertriglyceridemia with diets rich in M.C.T. or sucrose.
