Occupational exposure to benzene, toluene and xylene and the T lymphocyte functions.

In 72 workers having occupational contact with benzene, toluene or xylene during 31 to 122 months, the T lymphocyte functions were evaluated. Workers after a service of 55-122 months showed a decreased T lymphocyte count without, however, alterations in their functions. No clinical signs of diminished immunological reactivity were noted in the subjects studied. The E rosette test is of importance in the evaluation of early effects on the lymphocyte system of the toxic action of benzene and its homologues.

[Cytochemical and immunological tests in workers exposed to organic paint and varnish solvents. IV. Nonspecific esterase activity in lymphocytes].

By the histochemical semi-quantitative method, the activity of nonspecific EN esterase in lymphocytes of those occupationally exposed to benzene, toluene and xylene was determined. The airborne concentrations at the workstations were: benzene--0-370, toluene--0-580, xylene--0-560 mg/m3. A reduced number of lymphocytes containing EN in lysosomes and increased number of cells exhibiting EN activity in lysosomes and cytoplasm were found in workers employed over 54 months. That would imply a damage to lysosomes with subsequent displacement of lysosomal enzymes to cytoplasm. In view of the positive correlation between exposure duration and number of lysosomes with damaged EN--positive lysosomes, the authors suggest the histochemical method of semi-quantitative determination of EN activity may be applied to evaluate toxic effects of benzene and its homologues upon lymphocytes.

The neutrophil and lymphocyte nonspecific esterase in patients with malignancies.

In 52 patients with malignancies such as lung cancer, carcinoma of the stomach, and cancer of the colon (25, 18, and 9 cases, respectively) activity and intracellular localization of alpha-naphthyl acetate esterase has been evaluated by means of semiquantitative cytochemical method. Patients with advanced metastatic process occurring during clinical evolution of those malignancies showed decreased activity of above enzyme within the peripheral blood neutrophils and decreased numbers of enzyme-positive lymphocytes having intact enzyme-positive lysosomal granules. The authors discuss the significance of that observation in relation to the enzymatic alterations of cells during antitumor response of immune system.

N-acetyl-beta-D-glucosaminidase in lymphocytes of workers exposed to benzene and its homologues.

In 108 workers having occupational contact with benzene and its homologues by 1 to 122 months, the activity of N-acetyl-beta-D-glucosaminidase (NAG) within the peripheral blood lymphocytes was estimated by means of cytochemical method. The benzene, the toluene and the xylene concentration at work stands was equal to 0-370, 0-580 and 0-560 mg/cu . m, respectively. In 41 workers having the service time equal to 55-122 months the NAG deficiency within the lymphocyte lysosomes was stated. Above phenomenon is probably due to disintegration of lysosomes caused by lipophilic properties of benzene.
