Synthetic antigens. Horse "natural" antibodies against interpolymer of styrene and maleic acid (PSM).

Properties of horse natural anti-PSM antibodies are described. The antibodies were of IgG class. Electrostatic forces were mainly involved in reaction of PSM with horse antibodies. The reaction was inhibited by low molecular compounds resembling structural unit of PSM. Studies of difference spectra and ORD and CD spectra showed no major conformational changes in horse antibodies after reaction with PSM.

Anti-TNP antibodies from bovine serum and colostrum.

Specifically purified anti-TNP antibodies of subclass IgGI and IgG2 were isolated using immunoadsorbent prepared from AH-Sepharose and TNP-BSA. Isoelectric focusing in poliacrylamide gel showed differences between antibodies obtained from colostrum and serum. These differences were more observable when H chains of IgG1 subclasses were compared. No differences were observed in L chains. The antibodies obtained were the non-precipitating ones. Spectral measurements in the presence of SO3- ions, showed that the antibodies studied were of low affinity (below 10(-6) M). The difference spectra showed, that binding of a hapten (epsilon-TNP aminocapronate acid) to the investigated antibodies caused the shift of the hapten's absorption bands indicating that the binding occured in the hydrophobic pocket of the antibody binding site. The strongest effect was observed in the case of antibodies of IgG1 subclass.

Properties of TFX-A product obtained from calf thymus.

Properties of TFX--a product obtained from calf thymus by Pharmaceutical Comp. in Jelenia Góra--are described. The preparate is a mixture of polypeptides and peptides of various molecular weights with various biological activities. Attempts of isolation of active fractions, devoid of balast proteins are described.

Synthetic antigens. Rabbit antibodies against interpolymer of styrene and maleic acid (PSM).

Rabbit antibodies against interpolymer of styrene and maleic acid (PSM) are described. Rabbits immunized with PSM (mol. weight 292,000) produced precipitating and non-precipitating antibodies of IgG class. Interaction of rabbit antibodies with PSM was mainly of electrostatic character. It was found that various low molecular weight compounds resembling structural unit of the polymer inhibited reaction of PSM with antibodies. The reaction was also inhibited by picric acid and epsilon-TNP-aminocaproic acid. It suggested a multifunctional character of antibodies studied. The apparent affinity constant was 2 x 10(5) M-1 for interaction of PSM with the rabbit antibodies. Spectral studies suggested the presence of tyrosine in the combining site of anti-PSM antibodies. Specifically purified antibodies showed no restriction of heterogeneity, in comparison with rabbit IgG containing anti-PSM antibodies.

Obtaining, evaluation and clinical application of antileukemic antilymphocyte globulin (ALLG) in the treatment of chronic lymphatic leukemia.

Antileukemic antilymphocyte globulin (ALLG) was obtained by immunizing horses with peripheral blood lymphocytes from patients suffering from chronic lymphatic leukemia. After absorbing the serum with erythrocytes, globulins were precipitated from the serum with ammonium sulfate. ALLG preparations were sterile, nontoxic and nonpyrogenic and contained no antibodies against basement membranes or antierythrocytic or antiplatelet antibodies. Nine patients with chronic lymphatic leukemia were treated with the globulin. Methods for selecting patients for treatment with ALLG and for controlled therapy were elaborated. Clinical and hematologic observation in the patients were reported. The high therapeutic efficacy of ALLG with high cytotoxic titer was emphasized. Beneficial hematologic and therapeutic effects were obtained in six patients. Attention was drawn to the efficiency of the combined therapy with ALLG, cytostatics and adrenal steroids.

The presence of secretory component-free human colostral IgA in early colostrum.

Human secretory IgA was prepared from colostrum. Different elution diagrams from CM-cellulose were obtained depending on the time of collection of colostrum. In case of early colostrum, collected within 6 hours post partum, two IgA fractions were obtained after chromatography on CM-cellulose. Both fractions have different molecular weights and different amino acid compositions. The results obtained suggest that the first fraction is a dimer of IgA monomers containing J chain with a molecular weight of 330,000 whereas the second fraction having the molecular weight 395,000 daltons is composed of IgA dimer, J chain and SC. When colostrum collected 48 or more hours was used as a source of IgA, only the second IgA fraction was obtained. The problem of SC-free IgA immunoglobulins is discussed.

Synthetic antigens. II. Immunogenic, antigenic and adjuvant properties of a copolymer of styrene and maleic anhydride.

Immunogenic, antigenic and adjuvant properties of a polyanionic copolymer of styrene and maleic anhydride (PSM) are described. The results obtained showed that PSM was immunogenic in BALB/c mice when administered without adjuvant, the optimal dose being 0.01 mug/mouse. Antibodies could be detected only by precipitation in gel but not by agglutination. High molecular weight polymer (290 000 daltons) was a stronger immunogen and antigen than copolymers of lower molecular weights (85 000-210 000 daltons). PSM also showed adjuvant activity and enhanced the humoral response of mice against sheep red blood cells. Adjuvanticity was found only when 1 mug of PSM/mouse was applied. The results obtained suggest that PSM interacts with B rather than T cells.
