Novel pattern of editing regions in mitochondrial transcripts of the cryptobiid Trypanoplasma borreli.

In mitochondria of Kinetoplastida belonging to the suborder Trypanosomatina, the nucleotide sequence of transcripts is post-transcriptionally edited via insertion and deletion of uridylate residues. In order to shed more light on the evolutionary history of this process we have searched for editing in mitochondrial RNAs of Trypanoplasma borreli, an organism belonging to the suborder Bodonina. We have cloned and sequenced a 5.3 kb fragment derived from a 37 kb mitochondrial DNA molecule which does not appear to be a part of a network structure and have found genes encoding cytochrome c oxidase (cox) subunit 1, cox 2 and apocytochrome (cyt) b, and genes encoding the small and large subunit mitoribosomal RNAs. The order in which these genes occur is completely different from that of trypanosomatid maxicircle genes. The 5' and 3' termini of both the cytb and cox1 gene are cryptic, the protein coding sequences being created by extensive insertion/deletion of Us in the corresponding mRNA sections. Phylogenetic analyses of the protein and ribosomal RNA sequences demonstrated that the separation between T.borreli and Trypanosomatina was an early event, implying that U-insertion/deletion processes are ancient. Different patterns of editing have persisted in different lineages, however, since editing of cox1 RNA and of relatively small 3'-terminal RNA sections is not found in trypanosomatids. In contrast, cox2 RNA which is edited in trypanosomatids by the insertion of four Us, is unedited in T.borreli.

Comparison of technetium-99m-Q3 and thallium-201 for detection of coronary artery disease in humans.

UNLABELLED: Technetium-99m-Q3 is a myocardial imaging agent that produces prompt myocardial visualization in humans. METHODS: In 19 patients with angiographic coronary artery disease, 2 patients with no angiographic coronary artery stenosis greater than 50% of the luminal diameter and 6 healthy volunteers, exercise and resting myocardial imaging were performed with 99mTc-Q3 and also with 201Tl. Technetium-99m-Q3 imaging began 15 min after injection at rest and with exercise, in a complete imaging sequence that required less than 100 min. RESULTS: Overall accuracy for coronary disease detection was 78% (21 true-positive or true-negative studies among 27 study participants) by tomographic thallium imaging versus 89% for 99mTc-Q3 tomographic imaging (p = ns). Accuracy for detection of individual coronary stenoses was 75% (61 true-positive or true-negative coronary segment classifications among 81 total coronary segments) for 201Tl imaging and 83% for 99mTc-Q3 imaging (p = ns). CONCLUSIONS: Technetium-99m-Q3 when used in a rest-exercise sequence that can be completed in 100 min appears to provide comparable diagnostic accuracy to 201Tl for overall coronary disease detection and detection of individual coronary artery stenoses.

Neopterin in the diagnosis of disorders associated with immune system activation.

While the knowledge concerning the role of the immune system in many internal disorders has grown rapidly in recent years, there are few methods to assess immune system activation in clinical practice. Measurement of urine neopterin, product of a metabolic pathway controlled by interferon-gamma, has been found useful in many clinical conditions. The present study concerns neopterin excretion in 157 patients with different internal disorders. As expected, we found an increase in urine neopterin in patients with malignant tumors, autoimmune disorders like systemic lupus erythematosus or inflammatory bowel disease, and infections. Elevated neopterin levels were also observed in acute pancreatitis and in acute myocardial infarction. In addition, significant correlations between urine neopterin and zinc and neopterin and copper excretion were found suggesting a physiological role of neopterin as urine antioxidant.

A coccidian (Apicomplexa: Eimeriidae) with extracytoplasmally located stages in the kidney tubules of golden carp (Carassius auratus gibelio L.) (Cyprinidae).

Numerous coccidian stages were found in the kidney tubules of the golden carp (Carassius auratus gibelio). The merogonial and gamogonial stages were localized extracytoplasmally in the microvillous region of the epithelial cells. The host-parasite interface consisted of i) a large area where the parasite was separated from the host cytoplasm by the parasitophorous vacuole membrane only, and ii) a zone of multiple fusions of the host cell membrane investing the parasite to the neighbouring microvilli. The taxonomic status of the extracytoplasmic stages is not clear, however, their possible appurtenance to Eimeria scardinii, which was frequently found in the kidneys of golden carps in the same population, is discussed.

The differentiation between freshwater and marine fish trypanosomes by lectin agglutinability.

The agglutinability of three fish trypanosomes, Trypanosoma boissoni and T. triglae senegalensis from marine fish, and T. carassii from freshwater fish was compared. The tests were performed with trypomastigotes and epimastigotes in the exponential and stationary growth phases. The simple agglutination test, performed in microwell plates, used ten purified lectins, and revealed a clear difference between marine and freshwater strains when compared for their agglutinability with the HPA lectin. Whereas the cells of T. boissoni and T. triglae senegalensis became strongly agglutinated by this lectin, those of T. carassii remained unagglutinated in the same conditions. However, the cells of T. triglae senegalensis reacted positively with ConA lectin in the stationary phase only. The differences in structure and composition of surface polysaccharides stable after prolonged cultivation in vitro are inferred on the basis of the results obtained.

Goussia carpelli (Apicomplexa, Eimeriorina) from cyprinid fish: field observations and infection experiments.

Intestinal tissue samples of 9 cyprinid and gobiid fish species from several hatcheries and ponds in South Bohemia were found to harbour Goussia carpelli-like coccidian oocysts. Furthermore, laboratory transmission of Goussia carpelli, obtained from common carp, was attempted to 9 cyprinid fish species, both laboratory reared and obtained from pond environments. Infection experiments, carried out by fecal contamination (direct transmission) and by using tubificid oligochaetes as paratenic hosts resulted in weak infections in 4 out of 15 Carassius auratus used, but it was not possible to infect any of the other fishes used.

Catecholamines in adult iron deficiency patients.

Patients with iron deficiency whether uncomplicated or associated with other types of anemias, had plasma catecholamine levels which were significantly increased above normal controls. Patients with a variety of other anemias had no significant increase in catecholamine levels. Plasma catecholamine levels in uncomplicated iron deficient patients approached normal values as early as 3 h following oral FeSO4.
