[Immune response in Pneumocystis carinii pneumonia in rats].

OBJECTIVE: To study the inflammatory immune response to Pneumocystis canii pneumonia(PCP) in rats induced by glucocorticoid(GC). METHODS: The model of PCP was set up by injecting GC subcutanously to SD rats. Lymphocyte proportion in peripheral blood, CD4+/CD8+ T cell ratio of PBL and lymphocyte proportion in the BALF were measured. The levels of sIL-2R and TNF-alpha in the BALF were detected. RESULTS: 1. After the rats were immunospressed, the lymphocyte proportion in the peripheral blood and CD4+/CD8+ ratio of PBL, and the lymphocyte proportion in the BALF were decreased, and the levels of sIL-2R, TNF-alpha in BALF were reduced. 2. The lowest levels of TNF-alpha in BALF and CD4+/CD8+ T cells of PBL were observed in PCP group; 3. The lymphocyte proportion in the BALF was significantly higher in PCP group than in PC negative group. CONCLUSION: The reduction in the level of TNF-alpha and CD4+/CD8+ T cell ratio in rats treated with GC might result in PCP infection under immunosuppressive condition.

[Uncomplicated bronchial tuberculosis].

Thirty four cases of uncomplicated bronchial tuberculosis proved by fiberobronchoscopy (FB) and operation in recent ten years were reported. The total diagnostic yield of FB was 85.6%. The preliminary results revealed some new characteristics of uncomplicated bronchial tuberculosis during antituberculosis chemotherapy era: uncomplicated bronchial tuberculosis accounted for great proportion among bronchial tuberculosis; the involvement of middle and lower bronchi was more common; positive rate of sputum smear for AFB was low. Among the fibrobron choscopic manifestations, the percentage of proliferative lesions in bronchi was high.

The effect of endotoxin lipopolysaccharide from different bacterial species on the generation of intracellular inositol trisphosphate and superoxide in a human phagocytic cell line.

The acute effects of endotoxins and lipid A on two intracellular responses, inositol phosphate generation and superoxide production were analysed in the DMSO differentiated premyelocytic leukaemic HL-60 cell line. Short-term incubation (1-10 min) with Escherichia coli-type LPS, Salmonella-type LPS and Lipid A caused significant increases in cellular InsP1 and InsP3, compared with control cells (P less than 0.5-P less than 0.001). The Escherichia coli-type LPS released approximately twice the quantity of InsP3 compared with Salmonella-type LPS (P less than 0.001). Lipid A-dependent stimulation of InsP3 production was also detected. The rate of superoxide production increased 1-10 min after addition of both Escherichia coli- and Salmonella-type LPS and Lipid A. Endotoxins and Lipid A caused a dose-dependent increase in intracellular oxidative activity. The superoxide response showed less species dependence and a higher response to particulate lipid A compared with the inositol phosphate response.

Endotoxin releases platelet-activating factor from human monocytes in vitro.

The role of platelet-activating factor (PAF) in human endotoxaemia was investigated by incubating human monocytes from 27 subjects with S. minnesota endotoxin in vitro. Monocytes synthesized 1.01 +/- 0.4 x 10(-10) M to 5.3 +/- 1.51 x 10(-9) M PAF after 60 min of incubation with endotoxin at concentrations of 1 x 10(-8) M and 4.5 x 10(-6) M respectively. The endotoxin-stimulated release of PAF was not significantly increased in monocytes cultured on a human endothelial cell monolayer. This rapid release of PAF in response to endotoxin is consistent with a role for monocyte-derived PAF as a toxic mediator of the acute systemic changes observed in patients with endotoxin-related septic shock.
