A short synthesis of antimalarial peroxides.

A concise and efficient synthesis of two simplified diastereomeric analogues of a natural peroxide is presented. Both compounds could be isolated in high purity and fully identified. They exhibited moderate antimalarial activity.

Coriadienin, the first annonaceous acetogenin with two double bonds isolated from Annona coriaceae.

A new Annonaceous acetogenin, coriadienin (1), has been isolated from the roots of Annona coriacea Mart. (Annonaceae). This structure is the first reported acetogenin containing two double bonds. Compound 1 showed potent cytotoxicity against VERO and KB cell lines. Compound 1 appears to play an important role in the biosynthetic pathway of mono- and bis-THF acetogenins, and it is proposed as a biogenetic precursor of coriacin. A known cytotoxic acetogenin, gigantetronenin, was also isolated from this plant.

Vidarabine-loaded nanoparticles: a physicochemical study.

The chemical reaction of vidarabine (VIDA) with isohexyl cyanoacrylate nanoparticles in a pH-dependent fashion occurs only in the presence of dioctylsulfosuccinate (DOSS). The formation of an ion pair with DOSS allows a better contact of VIDA with the monomer during the polymerization process taking place in micelles. On the basis of molecular weight profiles of the polymer, determined by gel permeation chromatography (GPC), it is proposed that VIDA induces the polymerization of cyanoacrylic monomers through a zwitterionic pathway. This mechanism allows the covalent linkage of the drug with the polymer, which is consistent with NMR experiments. The present study illustrates the need for physicochemical studies in the design of new colloidal drug delivery formulations.
