A first genotyping assay of French cattle breeds based on a new allele of the extension gene encoding the melanocortin-1 receptor (Mc1r).

The seven transmembrane domain melanocortin-1 receptor (Mc1r) encoded by the coat color extension gene (E) plays a key role in the signaling pathway of melanin synthesis. Upon the binding of agonist (melanocortin hormone, alpha-MSH) or antagonist (Agouti protein) ligands, the melanosomal synthesis of eumelanin and/or phaeomelanin pigments is stimulated or inhibited, respectively. Different alleles of the extension gene were cloned from unrelated animals belonging to French cattle breeds and sequenced. The wild type E allele was mainly present in Normande cattle, the dominant E(D) allele in animals with black color (i.e. Holstein), whereas the recessive e allele was identified in homozygous animals exhibiting a more or less strong red coat color (Blonde d'Aquitaine, Charolaise, Limousine and Salers). A new allele, named E1, was found in either homozygous (E1/E1) or heterozygous (E1/E) individuals in Aubrac and Gasconne breeds. This allele displayed a 4 amino acid duplication (12 nucleotides) located within the third cytoplasmic loop of the receptor, a region known to interact with G proteins. A first genotyping assay of the main French cattle breeds is described based on these four extension alleles.

Identification ability of the first five letters of Christian name and surname.

Identifiers must be easy for access and discrimination. Usual identification by surname and christian name is convenient, for these two identifiers are almost always available. Their discrimination ability was studied in terms of the theory of information and the rate of homonymy. In the French language, the first five letters of the surname provide information equal to 12.11 bits and the rate of homonymy is about 0.659%. If one adds the first three letters of the first name, the gain in the quantity of information is 1.68 bits and the rate of homonymy becomes 0.087%. So the first five letters of a surname and the first three letters of a christian name ensure a relatively satisfactory identification and may constitute a significant way of reinforcing the discrimination power of another identification system.

Modelling and simulation of information systems on computer: methodological advantages.

Modelling and simulation of information systems by the means of miniatures on computer aim at two general objectives: (a) as an aid to design and realization of information systems; and (b) a tool to improve the dialogue between the designer and the users. An operational information system has two components bound by a dynamic relationship, an information system and a behavioural system. Thanks to the behaviour system, modelling and simulation allow the designer to integrate into the projects a large proportion of the system's implicit specification. The advantages of modelling to the information system relate to: (a) The conceptual phase: initial objectives are compared with the results of simulation and sometimes modified. (b) The external specifications: simulation is particularly useful for personalising man-machine relationships in each application. (c) The internal specifications: if the miniatures are built on the concept of process, the global design and the software are tested and also the simulation refines the configuration and directs the choice of hardware. (d) The implementation: stimulation reduces costs, time and allows testing. Progress in modelling techniques will undoubtedly lead to better information systems.

Lying to patients: can it ever be justified?

The acts of withholding information from patients and of deliberately issuing false information have been embodied in nursing and medical practice for centuries. The author considers the relationship of veracity, or truth-telling, to the four established tenets of medical ethics: autonomy, justice, beneficence and non-maleficence. Although there are occasions when withholding information can be supported, particularly when the patient has requested that this be so, she concludes that there can be no moral justification for health professionals lying to patients.

Buying software off the rack. Packages can be the solution to the software shortage.

The rapid pace of the computer revolution may well be slowed by the lack of suitable software. Though computers should be increasing business efficiency and productivity, many companies do not have the software that would enable them to enjoy these benefits. Packaged software can close the gap for both large and small companies. Here are some guidelines for making the decision whether to buy a package and for avoiding trouble once the package is installed. The article also describes the features a company should look for in packaged software and the optimal provisions of a software contract.

Severe cold agglutinin disease and cryoglobulinemia secondary to a monoclonal anti-Pr2 IgM lambda cryoagglutinin.

OBJECTIVE: To present a case of cold agglutinin disease/cryoglobulinemia secondary to a monoclonal anti-Pr2 IgM lambda antibody, and review the literature on the occurrence of this antibody in cold-induced disease and the clinical disease associated with it. METHODS: Cryoantibody characteristics were evaluated by cold precipitation. The antigen specificity of the monoclonal IgM lambda antibody was evaluated using techniques of selective red blood cell absorption. RESULTS: In our patient, we were able to identify an antibody with both cryoglobulinemic and cold agglutinin (cryoagglutinin) properties. This antibody was found to be monoclonal IgM lambda with specificity to the Pr2 antigen on red blood cells. CONCLUSIONS: Monoclonal IgM lambda anti-Pr is a rarely found cold agglutinin antibody. In this report we describe the clinical course of a patient who had this antibody, which not only agglutinated red cells in the cold but also had cryoglobulin properties. The clinical illness of this man was characterized by severe acrocyanosis and digital necrosis with eventual organ necrosis and death. We also review the literature on cold induced disease due to monoclonal anti-Pr IgM lambda antibody. Our patient was found to be unique among the reports reviewed. Our case is the first to report both cold agglutinin and cryoglobulinemic properties with the evaluation of the thermal amplitudes of these activities of the antibody. Also, unlike the lymphoproliferative malignancy observed in the cold agglutinin-associated disease in the other reports, our patient's disease was associated with a monoclonal B-cell expansion on the spectrum between benign monoclonal gammopathy and a low grade lymphoproliferative disorder.

Production and regulation of matrix metalloproteinases and their inhibitors by human peritoneal mesothelial cells.

OBJECTIVE: Human peritoneal mesothelial cells (HPMC) are likely to be involved in maintenance of the peritoneal membrane. We determined whether these cells were able to synthesize the matrix degrading enzymes, matrix metalloproteinases (MMPs), likely to be responsible for the breakdown of this membrane, and whether this secretion could be modulated by cytokines involved in the inflammatory response. DESIGN: MMP activity in conditioned medium of growth-arrested HPMC was measured by zymography. Cultures were incubated in the presence and absence of the cytokines transforming growth factor-beta (TGFbeta) and interleukin (IL)-1beta in order to determine the effects of these cytokines on this process. The mRNA for these MMPs, together with that of their specific inhibitors, tissue inhibitors of metalloproteinases (TIMPs), was also examined by reverse transcriptase polymerase chain reaction RESULTS: HPMC were shown to constitutively secrete the metalloproteinases MMP-2 and MMP-3 in vitro. In response to the proinflammatory cytokine IL-1beta , the protein and mRNA for MMP-9 was induced, while secretion of MMP-2 was unaltered. Similarly, the mRNA for MMP-3 was also increased relative to actin following the addition of IL-1beta. TGFbeta was shown to slightly induce the secretion of MMP-2 together with the mRNA for TIMP I, TIMP II, and, to a greater extent, TIMP III. Used peritoneal dialysate was also shown to induce MMP-9 secretion, and this effect was blocked by the co-incubation of IL-1 receptor antagonist. The secretion of enzyme activity was shown to be from the apical surface of the cells. CONCLUSION: HPMC have the ability to control the accumulation of extracellular matrix by secreting the matrix degrading molecules MMP-2, MMP-3, and MMP-9. In addition, the secretion of these enzymes, together with that of their inhibitors (TIMPs) is regulated by the cytokines IL-1beta and TGFbeta. This process is likely to be important in both the normal maintenance of the integrity of the peritoneal membrane and in the changes that occur following prolonged peritoneal dialysis.
