Diethazine, amitriptyline and imipramine showed bacteriostatic and bactericidal effect on different bacteria. Chlorpromazine sulphoxide and fluorescein were ineffective even at 1000 microgram/ml. The antibacterial compounds deleted at 40-70% frequency the F'lac plasmid of Escherichia coli K12 LE-140.
It is recommended to introduce screening for fertility and potency determination to disclose genital disturbances the correction of which must be done in childhood or early age. Cryptorchism, undescended testicles or their hypoplasia, hypogonadism, hypogenitalism, hypospadias and finally varicocele are easily detected by screening at school, on military enlistment or by the sports physician.
Five phenothiazine derivatives (chlorpromazine, levomepromazine, diethazine, promethazine and chlorpromazine) sulphoxyde were tested for antimycobacterial activity. The growth of Mycobacterium tuberculosis, M. bovis and M. butyricum was inhibited by chlorpromazine practically at identical concentrations. The minimum inhibitory concentrations for M. tuberculosis were: chlorpromazine and levomepromazine, 10 microgram/ml; diethazine and promethazine 20 microgram/ml, whilst chlorpromazine sulphoxyde was ineffective even at a concentration of 100 microgram/ml. Chlorpromazine and promethazine exerted a measurable bactericidal activity on M. tuberculosis at 50 microgram/ml; total destruction of the organism and loss of acid fastness in part of the cells were shown at 300 microgram/ml. Preliminary studies, in mouse experiments phenothiazine derivatives were ineffective.
The author deals with the anomalies and diseases of the dpididymis, the spermatic ducts and the adnexal glands which may have an influence on the spermatogram and the fertility, respectively. It is reported on the agenesia of the epididymis, the spermatic duct and the glands of the vesicles. Then in turn the concerning relations of the spermatocele and the inflammation in the genital region, above all in their chronic form are discussed.
The sedimentation characteristics and protein composition of the polyadenylic acid-containing ribonucleoprotein (RNP) particles separated from the polysomes of Ehrlich ascites carcinoma cells by means of ribonuclease treatment were studied. The results were compared with the corresponding characteristics of the poly(A)-containing particles of the cell nucleus. The sedimentation coefficient of the poly(A)-containing RNP from poly-ribosomes was found to be 11 S, while that of the nuclear particles was 14 S. In Cs2SO4 equilibrium density gradient values of p=1.25 and 1.34 gcm-3 were obtained for the nuclear particles and the poly(A)-RNP separated from polyribosomes, respectively. The molecular weights of the proteins making up the polysomal poly(A)-particles were found to be over 40 000 daltons, i.e. 54 000, 48 000, 68 000 and 89 000 daltons for the four polypeptides present in considerable amounts. As far as electrophoretic mobility is concerned these proteins were indistinguishable from the proteins of the nuclear poly(A)-containing RNP. However, the ratio of the components in the two types of particle proved to be quite different.
Chlorpromazine, levopromazine and promethazine exerted a bacteriostatic effect on Gram-positive bacteria at 20-60 mug/ml, on Gram-negative bacteria at 130-180 mug/ml concentration. Of the three compounds, chlorpromazine had the most marked bactericidal effect on cultures of Bacillus anthracis growing in minimal medium. In addition, chlorpromazine had a significant bactericidal effect on the resting cells of Escherichia coli suspended in saline. Pseudomonas aeruginosa was resistant to phenothiazines. Experiments have failed to derive resistant mutants from the highly sensitive B. anthracis. An effective R-factor elimination was observed at chlorpromazine concentrations of 50 mug/ml, practically not affecting the growth of multiple resistant E. coli.

Nuclear 14 S RNP particles containing poly (A) from Ehrlich ascites carcinoma cells and rat liver were purified by re-sedimentation in sucrose gradients, by Cs2SO4 density gradient centrifugation and by affinity chromatography on a poly (dT)-Sepharose column. Proteins of these RNP particles were electrophoresed in urea and SDS-polyacrylamide gels. RNP particles of ascites carcinoma cells contain two main bands having molecular weights of 51 000 and 69 000 daltons, respectively, and two or three minor components.

The nuclear ribonucleoprotein particles containing polyadenylic acid were prepared from rat liver nuclear extract by treatment with RNase I. The sedimentation coefficient of the particles was approximately 14 S. These particles represent a component of the pre-mRNA containing ribonucleoprotein complexes of the nucleus. The submicroscopic morphology of the particles was examined in preparations pretreated by platinum-palladium shadowing and by various negative staining methods as well as in ultrathin sections. The 14 S particles are globular structures, 120-140 A in diameter. The particles are characterized by a strong tendency to aggregation. The results are discussed with respect to mRNA transport.
The identification of men's fertility troubles is based upon a modern spermatogram. This procedure comprehends a laboratory action which contents the strict following of the examination requirements, and the preparing of a so-called simple (or, if necessary, an "extended") spermatogram. Then comes the genital, somatic examination, which discovers the possibly existing shortages of this field. In special cases it is also desired to clear up additionally the endocrine, hormonal state of the patient. So the results of the analyses will not only give information about the grade of fertility, but they also give a prognosis, and show the possible ways of therapy, in the same time. If reasonable, the testisbiopsy should be made. This could give some valuable datas to discover the shortages in the parenehyma, and to obtain a right prognosis. In case if both parties in the marriage can be considered as fertile, but yet, no sign of the pregnancy appears, the matter will belong to the immunologist, even regarding the sperma-autoimminulogy. Nowadays the genetist is to be initiated, too, if early, of habitual abortions happen, which can not be explained within the field of gynecology, or if any anomalies of evaluation appear at the offspring. Since these examinations neither cause uncomfortabilities, nor risici to the men, it is reasonable to the men to prepare the andrological clear-ups first. Their results mostly can provide the gynecologists searching the origin of sterility with important depending points.
Nuclear 14 S ribonucleoprotein particles containing poly(adenylic acid) were isolated from Ehrlich ascites carcinoma cells and purified by a two step method. The nuclear extract obtained at an ionic strength of 0.1 was treated by pancreatic and T1 ribonucleases and sedimented in a 15-30% sucrose gradient. The 14 S peak containing 50-60% poly(adenylic acid) was chromatographed on a poly(dT)-Sepharose column. The particles thus obtained were of high purity: they had an adenosine content of 90% and a protein moiety characteristic of the 14 S nuclear particles i.e. polypeptides with molecular weight of 51 000, 61 000 and 69 000 daltons.
After mutations affecting the de novo adenine nucleotide synthesis in Bacillus anthracis, the salvage pathway failed to ensure growth at the prototroph level. In sonicated suspensions of adenyl succinate lyase deficient B. anthracis mutant, in the presence of exogenous 5-phosphoribosyl-1-pyrophosphate (PRPP), part of the labelled adenine was incorporated into adenine nucleotides. Addition of PRPP to cultures of adenyl succinate lyase and adenyl succinate synthetase mutants failed to promote growth. Both kinds of auxotroph showed in vitro a reduced growth rate as compared to the prototrophic strain.
