Analysis of polyphenols in wines: correlation between total polyphenolic content and antioxidant potential from photometric measurements. Prediction of cultivars and vintage from capillary zone electrophoresis fingerprints using artificial neural network.

The polyphenols (some of them are also called phytoalexins, flavonols, flavanons, flavanonols, flavons, flavanols, and anthocyanines) are usually marked as potent antioxidants or radical scavengers which assist the body cells against oxidation. Polyphenols in wine are also considered to explain so called French paradox (long life aging and low number of coronary diseases despite of high alcohol and fat consumption). The total polyphenolic content (TPC) and total antioxidant potential (TAP) were determined by photometry and found strongly correlated. This finding suggests that the determination of TAP can be replaced by a more simple procedure of TPC determination. Capillary zone electrophoresis (CZE) with preconcentration by solid phase extraction (SPE) was applied for some polyphenols determination and for obtaining electropherograms of the SPE extracts (fingerprints). From mathematical evaluation of the fingerprints, prediction of cultivars and vintage using artificial neural networks (ANN) was done with more than 90% correct prediction. The study was performed on a set of 47 samples of young wines (vintage 1999-2002) from south Moravia (Czech Republic) and New South Wales (Australia).

Validation of a capillary zone electrophoresis method for determination of rimantadine hydrochloride in Rimantadin100 tablets and the method application to dissolution test monitoring.

A capillary zone electrophoretic method with indirect UV-detection for determination of rimantadine, an antiviral drug against influenza A, in tablets was validated. Instrumental precision, the method precision, accuracy, calibration curve linearity, selectivity, robustness, and time stability of the sample and the standard were tested. The method was also applied to monitor dissolution tests of the tablets. The possibility of addition of an internal standard for improvement of the method precision was discussed.

Separation of polyphenols in Canary Islands wine by capillary zone electrophoresis without preconcentration.

A method for separation and determination of polyphenols in wine by capillary zone electrophoresis (CZE) without any preconcentration step is described. The sensitivity and limits of detection for gentisic and p-coumaric acid are better than those previously published. The effect of a possible C18 solid-phase extraction prior to the CZE analysis was examined. The developed optimized method (without any extraction step) was applied to the analysis of wines from Tenerife, Canary Islands.

[Long-term survival after palliative embolization of kidneys affected by tumors].

Based on their own experience and data in the literature, the authors recommend to use also in future palliative embolization of the kidney as part of comprehensive treatment of patients with inoperable tumours of the kidney. They describe their experience with a group of 20 patients. In one female patient after palliative embolization an unusually favourable effect was observed with macroscopic disappearance of the tumour and affection of the pleura, associated with prolonged survival.

[Testing bronchogenic carcinoma cells for sensitivity to cytostatics in vitro and in cell cultures].

The authors submit the first experience assembled in Czechoslovakia with testing of the sensitivity of bronchogenic carcinoma cells to cytostatics by the HTCA method (Human Tumor Clonogenic Assay). Based on work by A. W. Hamburger, S. L. Salmon and others the authors elaborated their own modification of the method of cell cultivation in a double layer of soft agar, which can be used in small laboratories attached to clinical departments. The sensitivity tests were made in eight bronchogenic carcinomas--four adenocarcinomas and four epidermoid carcinomas. For the tests five cytostatics were used: 5-fluorouracil, methotrexate, cis-platinum, vincristine and vinblastine. Because of contamination the results in three tested adenocarcinomas could not be evaluated. The evaluated specimen of adenocarcinoma was sensitive to 5-fluorouracil and methotrexate, but was resistant to the remaining tested cytostatics. In the group of epidermoid carcinomas one specimen was completely resistant and one completely sensitive to all tested cytostatics. In one specimen the test with 5-fluorouracil could not be evaluated, and the specimen was resistant to the remaining cytostatics. The last specimen in this group was resistant to cis-platinum and sensitive to the remaining cytostatics.

Large granular lymphocytes in blood of healthy donors and patients with tumors.

The percentage of large granular lymphocytes (LGL) in the peripheral blood of patients with less advanced tumors (T1 and T2) averaged 2.15 +/- 1.3 as compared with 3.8 +/- 1.5 in patients with more progressive tumors (T3 and T4) and with metastases. The values of LGL remained constant as long as 6 weeks both in the diseased and healthy subjects and were not age- nor sex-related. This observation confirms the usefulness and reliability of the test as one of the markers of the immunologic state since LGL are considered to represent NK cells responsible for resistance to malignant growth.

Nucleolar morphology in estimation of urinary bladder cancer in man: study on surface contact preparations.

The special feature of nucleolar morphology which reflects the intensity of ribosomal RNA precursor molecule formation and thus also the intensity of proteosynthesis was studied in contact surface preparations from tissue samples of 28 patients with epithelial tumors of the bladder and in tissue samples of bladder mucosa from 16 patients with prostata hypertrophy (7 of them suffered also from cystolithiasis). In malignant lesions of the bladder mucosa, especially in tumors of medium and high grade malignancy, very low values of metabolically resting cells with ring shaped nucleoli and micronucleoli were found.
