Herbaceous vegetation productivity, persistence, and metals uptake on a biosolids-amended mine soil.

The selection of plant species is critical for the successful establishment and long-term maintenance of vegetation on reclaimed surface mined soils. A study was conducted to assess the capability of 16 forage grass and legume species in monocultures and mixes to establish and thrive on a reclaimed Appalachian surface mine amended with biosolids. The 0.15-ha coarse-textured, rocky, non-acid forming mined site was prepared for planting by grading to a 2% slope and amending sandstone overburden materials with a mixture of composted and dewatered, anaerobically digested biosolids at a rate of 368 Mg ha(-1) (dry weight). Tall fescue (Festuca arundinacea Schreb.), orchardgrass (Dactylis glomerata L.), switchgrass (Panicum virgatum L.), caucasian bluestem (Bothriochloa caucasia L.), reed canarygrass (Phalaris arundinacea L.), ladino clover (Trifolium repens L.), birdsfoot trefoil (Lotus corniculatus L.), crownvetch (Coronilla varia L.), alfalfa (Medicago sativa L.), common sericea lespedeza and AULotan sericea lespedeza (Lespedeza cuneata L.), tall fescue-ladino clover, tall fescue-alfalfa, orchardgrass-birdsfoot trefoil, switchgrass-AULotan, and an herbaceous species mix intended for planting on reforested sites consisting of foxtail millet [Setaria italica (L.) Beauv.], perennial ryegrass (Lolium perenne L.), redtop (Agrostis alba L.), kobe lespedeza (Kummerowia striata L.), appalow lespedeza (Lespedeza cuneata L.), and birdsfoot trefoil were established between spring 1990 and 1991. Vegetative biomass and/or persistence were assessed in 1996, 1997, 1998, 2000, 2001, and 2002. The high rate of biosolids applied provided favorable soil chemical properties but could not overcome physical property limitations due to shallow undeveloped soil perched atop a compacted soil layer at 25 cm depth. The plant species whose persistence and biomass production were the greatest after a decade or more of establishment (i.e., switchgrass, sericea lespedeza, reed canarygrass, tall fescue, and crownvetch) shared the physiological and reproductive characteristics of low fertility requirements, drought and moisture tolerance, and propagation by rhizome and/or stolons. Of these five species, two (tall fescue and sericea lespedeza) are or have been seeded commonly on Appalachian coal surface mines, and often dominate abandoned pasture sites. Despite the high rates of heavy metal-bearing biosolids applied to the soil, plant uptake of Cd, Cu, Ni, and Zn were well within critical concentrations more than a decade after establishment of the vegetation.

The prevalence of different human papillomavirus types and p53 mutations in laryngeal carcinomas: is there a reciprocal relationship?

This study investigates whether a reciprocal association occurs between HPV infection and somatic mutation of the tumour suppressor gene p53 in laryngeal carcinomas. Using immunohistochemical techniques, 87 tumours were examined for expression of the mutant form of p53 phosphoprotein using the monoclonal antibody PAB 1801. The prevalence of different HPV types in these tumours was determined by using the polymerase chain reaction and restriction enzyme analysis. Over-expression of p53 was noted in 50/87 (57.5%) of the tumours investigated but not in any of the non-neoplastic laryngeal mucosa (controls). There was no statistical correlation between p53 immunoreactivity and the clinicopathological parameters of laryngeal carcinomas. HPV DNA was detected in 8/36 (22.2%) of the tumours: HPV-6 in three, HPV-11 in one, HPV-16 in two, and unknown HPV type in two. In p53-mutant tumours, HPV was present in 4/20 tumours but none of these were high risk HPV types. In p53-normal tumours, on the other hand, HPV was present in 4/16 tumours but two of these were definitely high risk HPV-16. These results imply the reciprocal association between HPV and somatic mutations of p53 found in the case of cervical carcinoma.

General primer-mediated polymerase chain reaction for simultaneous detection and typing of human papillomavirus DNA in laryngeal squamous cell carcinomas.

The aim of this study was to investigate the prevalence of different human papillomavirus (HPV) types in laryngeal squamous cell carcinomas using general primer-mediated polymerase chain reaction (PCR). Tumour sections from 42 patients with laryngeal carcinomas were investigated. For HPV DNA amplification, consensus primers were used which were directed to the L1 coding region of the HPV genome. Analysis of the PCR products was done using 2% agarose gel electrophoresis followed by restriction enzyme analysis to identify different HPV types. Amplification of the human TGF-beta 1 DNA was successfully performed in 36/42 (85.7%) of samples confirming the presence of sufficient DNA for viral amplification. HPV DNA was detected in 8/36 (22.2%) of the tumours examined (three HPV-6, two HPV-16, one HPV-11, two unknown HPV types). HPV DNA was not detected in any of the non-neoplastic laryngeal mucosa which was used as control (n = 15). Fifty per cent of women had HPV-positive tumours compared with 8% of men (chi2 = 5.8, P < 0.05). Our data indicate that while the overall prevalence of HPV in laryngeal carcinomas is fairly high (22.2%), the frequency of high-risk types (HPV-16 & HPV-18) is low (5.5%). HPV probably acts as a promoter in the multistep process of carcinogenesis in squamous mucosal cells of the larynx.

Metabolic cardiac imaging in severe coronary disease: assessment of viability with iodine-123-iodophenylpentadecanoic acid and multicrystal gamma camera, and correlation with biopsy.

Fifteen patients with coronary disease and resting left ventricular ejection fractions of less than or equal to 0.35 underwent resting metabolic cardiac imaging utilizing 1 mCi [123I]iodophenylpentadecanoic acid (IPPA) intravenously and a multicrystal gamma camera. Parametric images of regional rates of IPPA clearance and accumulation were generated. Forty-two vascular territories (22 infarcted) were evaluated by metabolic imaging as well as transmural myocardial biopsy. Despite resting akinesis or dyskinesis in 20/22 (91%) infarcted territories, 16/22 (73%) of these territories were metabolically viable. Transmural myocardial biopsies in all patients (43 sites, 42 vascular territories) during coronary bypass surgery confirmed IPPA results in 39/43 patients (91%). When compared to biopsy, scan sensitivity for viability was 33/36 (92%) with a specificity of 6/7 (86%). Eighty percent of bypassed, infarcted but IPPA viable segments demonstrated improved regional systolic wall motion postoperatively as assessed by exercise radionuclide angiography. We conclude resting IPPA imaging identifies viable myocardium, thereby providing a safe, cost-effective technique for myocardial viability assessment.

Gallium scintigraphy in the detection of amiodarone lung toxicity.

Fourteen gallium scans were obtained in 11 patients suspected of having amiodarone lung toxicity on the basis of clinical findings, pulmonary function tests, and chest radiographs. All 11 patients had abnormal scans. Gallium accumulates in various inflammatory and neoplastic lesions, but despite this nonspecificity, the findings suggest gallium scintigraphy is a useful procedure to detect amiodarone lung toxicity when used in the appropriate clinical setting.

Microarray-based compendium of hepatic gene expression profiles for prototypical ADME gene-inducing compounds in rats and mice in vivo.

To examine species-specific aspects of the induction of absorption, distribution, metabolism and excretion (ADME)-related genes, we used 25 000 gene oligonucleotide microarrays to construct a rodent gene-response compendium that compared hepatic gene expression profiles and developed consensus aryl hydrocarbon receptor (AhR), constitutive androstane receptor (CAR) and pregnane X-receptor (PXR) ligand signatures relevant to drug clearance. Twenty-six inducer compounds were chosen from the literature. Rats and mice received one of six dose levels (log2 dose escalation, 32-fold dose range) of each compound daily for 3 days. Animals were necropsied 6-9 h after the last dose, and tissues were collected for RNA analysis. Hepatic gene expression profiles were obtained using Rosetta Resolver expression analysis system, and ADME-related genes were extracted. Cross-talk among nuclear receptors or hepatoxicity at high dose levels resulted in large signatures (usually >1000 genes at p < 0.01) for most compounds. After ADME gene transcript enrichment, agglomerative clustering separated AhR ligands from CAR/PXR ligands, but it was difficult to distinguish CAR from PXR ligands. Consensus signatures were derived from groups of AhR, CAR and PXR ligands; and cross-talk among responding genes was determined. Many compounds had distinct log dose-response profiles, and relative potencies for ligands were established. Robust responses by CYP1A1, CYP2B10 (CAR responsive in mice) and CYP2B15 (CAR responsive in rats) and CYP3A1 (PXR responsive in rats) were used to benchmark the relative potency of different ligands and to determine the relative selectivity for AhR, CAR or PXR. By using a compendium of gene expression profiles, we defined species-specific induction patterns across the ADME transcriptome.

Opti-Free chemical disinfectant: a safety study with various soft contact lenses.

A 6-month, Phase I clinical study evaluated Opti-Free, a new polyquaternium-1 chemical disinfectant intended for use with all types of soft contact lenses. Fifty-one patients were enrolled and divided into two groups. Group A used the product under exaggerated conditions, with daily exposure of Durasoft 3 (55%) and Hydrocurve II (55%) medium water content, ionic lenses to 5 mL of Opti-Free for 16 hours. Beginning on day 15, group A patients began a normal daily use schedule. Patients in group B wore CSI (38%), Permalens (71%), and Softcon (55%) contact lenses and used the regimen under normal daily use conditions throughout the study. The result indicate that Opti-Free is safe and effective under both normal and exaggerated use conditions. The implications of these data for patient safety are discussed.
