Subcellular localization of IgG from the sera of ALS patients in the nervous system.

Immunoglobulin G (IgG) samples isolated from the sera of amyotrophic lateral sclerosis (ALS) and control patients were injected intraperitoneally into mice. After 24 h the mice were processed for immune electron microscopic immunohistochemistry to localize IgG in their nervous system. The injected ALS IgG was observed in the axon terminals of the lower motor neurons (MNs), localized to the microtubules and enriched in the rough endoplasmic reticulum (RER). In post-mortem spinal cord samples from ALS patients, IgG was similarly detected in the vicinity of the microtubules and in the RER of the MNs. IgG was neither found in the corresponding structures of MNs of mice injected with the control human IgG nor in post-mortem human control spinal cord samples. The data suggest that multiple antibodies directing to different structures of the MNs may play a role in their degeneration in ALS.

Widespread increased expression of the DNA repair enzyme PARP in brain in ALS.

Expression of the DNA repair enzyme poly(ADP-ribose) polymerase (PARP) is a known response to oxidative damage of DNA. In ALS brain, PARP expression by western analyses was increased in the motor cortex, parietal cortex, and cerebellum. PARP immunostaining in the motor cortex was increased in ALS neurons and subcortical glia and macrophages. Importantly, there was widespread increased PARP expression in neurons in the parietal cortex and cerebellum, regions that are typically clinically unaffected in ALS, suggesting widespread oxidative stress.

Coincidence of hereditary angioedema (HAE) with Crohn's disease.

A patient with two diseases, based presumably on different immunopathological mechanisms, hereditary angioedema (HAE) and Crohn's disease, was followed for 8 years. For more than three years of this observation period, detailed laboratory data were also available and could be analyzed. Both diseases had severe courses requiring chronic treatment with danazol and sulfasalazine, respectively. During exacerbation of Crohn's disease, the levels of C4 was found to be significantly lower than during the periods free of symptoms of both diseases. This drop was probably due to an impaired C1-inhibitor activity. HAE attacks and acute exacerbation of Crohn's disease never occurred simultaneously. This finding may be a mere chance but may also indicate that the different immunopathological processes underlying HAE and Crohn's disease influence each other.

[Simultaneous occurrence of hereditary angioneurotic edema and Crohn disease].

Hereditary C1 esterase inhibitor deficiency is often associated with immunpathologic disorders. The authors present a case of the rare coincidence of hereditary angioedema (HAE) and Crohn's disease. The history of the patient is analysed along with the familial occurrence of the disease. Characteristic abdominal manifestations of C1 esterase inhibitor deficiency are compared to the clinical signs of Crohn's disease. Differential diagnostic pitfalls are described along with efficatious therapeutic options.

[A new method, based on in situ DNA hybridization, for the detection of papillomavirus in condyloma patients and their partners].

The detection and typing analysis of human papillomavirus (HPV) were carried out by means of BIOHIT in situ HPV DNA hybridization screening and typing kits in condyloma tissues of patients with genital condyloma acuminatum, in cytological smears of their asymptomatic female consorts, and in female patients with vulval condyloma acuminatum. "Low"-risk HPV 6 and HPV 11 were demonstrated most frequently in the condyloma tissues of the male patients. It is noteworthy that all but one female consorts carried similar HPV type(s) in their cervical smears to those of their sexual partners in condyloma tissues. "Medium" and "high"-risk HPV 18, 31 and 33 types (frequently associated with high-grade dysplastic lesions and carcinomas) were demonstrated more frequently in female condylomas than in male. These three HPV types were identified in both tissues in a female patient with condyloma acuminatum and carcinoma epidermoids cornescens.

Papillomavirus DNA typing analysis in condyloma acuminatum patients and their consorts.

The present study demonstrated human papillomavirus (HPV) infection by means of an in situ HPV DNA hybridization screening test (BIOHIT) in 8 male patients with genital condyloma acuminatum, their asymptomatic female consorts and 6 female patients with vulval condyloma acuminatum. The investigations revealed that all but one of the female consorts were infected by HPV but did not show any clinical sign of papillomavirus infection. HPV DNA typing analyses with the most common HPVs present in the genital tract (HPV 6, 11, 16, 18, 31 and 33) revealed the same HPV types in the consorts as found in their partners. (HPV 6 and/or 11 were detected in 4 pairs, HPV 18 and 31 in one pair each, and HPV 18/31 and 16/31 double infections in one pair each.) All cytological specimens of female patients with condyloma acuminatum were HPV-infected, the most frequent type being HPV 31. In the one patient with vulval condyloma acuminatum and carcinoma epidermoides cornescens, the "high-risk" HPV 18 was identified in both condyloma and carcinoma tissues.

[Detection of antimitochondrial antibodies by immunodiffusion, ELISA and immunofluorescence methods].

Ouchterlony immunodiffusion and ELISA were used for the detection of antimitochondrial antibodies. The results were compared with those of immunofluorescence measurements. The occurrence of antimitochondrial antibodies was investigated in 20 patients with primary biliary cirrhosis, in 30 with autoimmune disease and in 28 with porphyria cutanea tarda. Sonicated rat liver mitochondria fractions were used as antigens. Among the patients with primary biliary cirrhosis, Ouchterlony immunodiffusion revealed antimitochondrial antibodies in 18 cases, whereas antibodies were not found in the other groups of patients, except for 1 case of systemic lupus erythematodes. ELISA demonstrated that in all 20 patients with primary biliary cirrhosis where immunofluorescence revealed antimitochondrial antibodies, there was an increase in the quantity of antibodies against mitochondrial inner membrane fractions, and in 7 cases even against the outer membrane fractions. These 7 were the most serious cases clinically. These methods allow the differentiation of antimitochondrial antibody subtypes and promote the establishment of the diagnosis.

Identification and cDNA cloning of a new nodule-specific gene, Nms-25 (nodulin-25) of Medicago sativa.

A new nodule-specific gene, Nms-25 (nodulin-25), was identified in cDNA clones isolated from a nodule-specific cDNA library of Medicago sativa. The first transcript of this gene appeared 9 days after inoculation of the roots with Rhizobium meliloti. The time of expression and the quantity of the transcripts of the Nms-25 gene was similar to that of leghemoglobin genes suggesting a similar regulation. A protein of 246 amino acids could be deduced from a full-length cDNA clone. The first 24 amino acids at the N-terminal end of this protein formed a signal sequence which might direct membrane transport into the peribacteroid space. Using different predictive methods the signal sequence cleaved protein was tentatively predicted to be a water-soluble enzyme, but not hydrolase.

Effects of precocene analogs on the nematode Caenorhabditis remanei (var. Bangaloreiensis). I. Structure/activity relations.

Precocenes (PI and PII) and 114 of their analogs (PAs) were synthetized and tested on C. remanei embryos for their precocene-like (P-like) activities resulting in unusual development at sublethal doses. The P-like activity was quantitated by plotting the probit of the percentage of the developmentally affected survivors against the (log) dose to obtain the EC plot and the half effective concentration (EC50). All five PAs (PI, PII, 7-ethoxy-PII, 7-(prop-2-ynyloxy)-PI, and 6-methoxy-7-(prop-2-ynyloxy)-PII) which exert both antiallatal activity in insects and P-like activity in nematodes are 7-alkoxy-substituted 2,2-dimethylchromenes. Both activities can be enhanced by an additional 6-MeO-substitution or by an asymmetric 6,7-dialkoxy-substitution, on condition that R-7 is longer than R-6. There are many more similarities than dissimilarities in the structural requirements needed for antiallatal and P-like activities. All but three nonantiallatal PAs effective in nematodes are 7-prop-2-ynyloxy-subsituted; two are symmetrically 6,7-disubstituted, and one is heterosubstituted (thio-PI). All PAs with antiallatal but without P-like activity are 7-monosubstituted with a relatively long alkoxy group. Certain substitutions favor antiallatal activity and others P-like activity. The severe nematocidal effect of 6,7-methylenedioxy-2,2-dimethylchromene (inert in insects) is not accompanied by P-like activity. The present findings lend some indirect support to the supposition that JH-producing cells and/or JH-dependent function(s) might exist in the nematodes.

Effect of calcium on the structure-function relationship of (Na+ + K+)-ATPase in cardiac sarcolemma.

Calcium-induced changes in (Na+ + K+)-ATPase activity and structural changes of membrane bound proteins in rat heart sarcolemma were investigated. Increasing concentrations of Ca2+ (0.1-8.0 mmol.l-1) gradually inhibited the (Na+ + K+)-ATPase activity and decreased the alpha-helix content of sarcolemmal proteins. Mathematical and graphical analysis of observed data yielded a quantitative relationship between Ca2+-induced changes in (Na+ + K+)-ATPase activity and the secondary structure of membrane proteins in cardiac sarcolemma.

Quantitative relationship between the protein secondary structure in cardiac sarcolemma and the activity of the membrane-bound Ca2+-ATPase.

In the absence of ATP, increasing concentrations of calcium within a range between 0.1--8.0 mmol . 1(-1) gradually lowered the alpha-helix content of proteins in rat heart sarcolemma requiring no energy supply. In the presence of ATP, similar concentrations of calcium stepwise activated the sarcolemmal low-affinity Ca2+-ATPase. A mathematical analysis of the data obtained revealed a quantitative relationship between calcium-induced stimulation of the Ca2+-ATPase activity and a diminution of the alpha-helix contents of membrane proteins in cardiac sarcolemma. The cooperation between changes in protein conformation and energy consumption in relation to the supposed role of low-affinity Ca2+-ATPase in gating the calcium channel are discussed.

Secondary structure of heart sarcolemmal proteins during interaction with metallic cofactors of (Na+ + K+)-ATPase.

he secondary structure of membrane proteins was studied in rat heart sarcolemma by circular dichroism under conditions of interaction with metallic cofactors of (Na+ + K+)-ATPase at their optimal concentrations and under metal free conditions. Approximately 80 per cent of polypeptide chains in the membrane were organized in alpha-helical structure. Upon stabilizing the E1. Na conformation state of (Na+ + K+)-ATPase by Mg2+ and Na+ ions, only a slight increase in the protein alpha-helix content (to 83 per cent) was observed. On the other hand, simultaneous addition of Mg2+ and K+ ions resulting in the establishment of the E2 . K conformational state of the enzyme, was followed by a significant decrease in the membrane protein helicity (to 72 per cent). The presence of all three metallic cofactors of (Na+ + K+)-ATPase did not induce any further conformational change in sarcolemmal proteins as compared to the state induced by the interaction with Mg2+ and Na+ ions. In contrast to results obtained with Mg2+ ions, the interaction of Na+ with the sarcolemmal membranes led to a considerable decrease and that of K+ to a significant increase in alpha-helicity of the membrane polypeptides. These findings have confirmed the regulatory role of magnesium in transition of the conformational state from E1 to E2 in the reaction sequence of (Na+ + K+)-ATPase. Specific modulation by Na+ and K+ of the helicity of sarcolemmal proteins in the presence of Mg2+ and in the absence of ATP might be considered as a preprint of conformational changes which will occur in the presence of ATP.

Protochlorophyll forms with different molecular arrangements.

Spectral properties of protochlorophyll ()PChl) forms were investigated in solid-film model systems by absorption fluorescence and circular dichroism (CD) spectroscopy. The solid films were prepared from diethyl ether solution of PChl on a cover glass surface by evaporation of the solvent. After preparation the films usually showed an absoprtion maximum at 635 nm or in some cases at 640 nm. The PChl form with 635 nm absorption maximum had no CD signal, whilst the films with absorption maximum at 640 nm gave an intense negative CD band at about 640 nm and a positive one at 668 nm. The treatment of the films with ammonia or acetone vapour resulted in a red shift of the absorption maximum from 635 nm or 640 nm to 650 nm. The study of the CD spectra of the films with different PChl forms showed that, depending on the treatment, forms of PChl with similar absorption and fluorescence spectra, but with opposite CD signals, can exist. It is suggested that the differences of the CD spectra are mainly due to different arrangements of the aggregates.

Heart block: histological and clinical features.

The clinical types of A-V-block and the localization of the underlying lesion to the conduction pathway have been examined in 25 patients. On the evidence of the post mortem studies the lesions were of the following types: 1) Necrosis or fibrosis of ischaemic origin: 13 cases; 2) lesions of inflammatory character: 5 cases; 3) nonspecific lesions; 6 cases; 4) malignant infiltration: one case. Correlations between the site of lesion and the clinical type and pattern of block could be demonstrated in a large proportion of the cases. The characteristic features of the individual types of block and their localization are discussed and a review of the literature is given.

[Anoxic myocardial lesions in man, developed during extracorporeal circulation, studied by electron microscopy].

Anoxemic changes of the myocardium were examined at 20 patients operated on in extracorporal circulation. At a mild degree hypothermia (30 degrees C) morphological changes appear after a 20 minutes period of anoxaemia. Damage to the myocard of irreversible character could be revealed after an anoxaemic period of 43--55 minutes. An intermitting perfusion of the coronary arteries delayes the formation of the lesions, although in cases of ligation of the aorta for 90 minutes severe mitochondrial changes and deformation of cells were seen. The best method for protecting the myocard is an isotherm perfusion of the coronary arteries. Clinical and laboratory findings show that necrosis of the myocard has a focal character and may play a part in formation of postoperative lesions.
